Holstein steer calves (130 kg) were used to monitor venoarterial concentration differences of plasma peptide and serum protein amino acids across the hindlimbs. Calves were fed purified diets containing soy protein or urea as the sole source of dietary N. Twenty-four hourly feedings per day were imposed to promote "steady-state" metabolism. Negative venoarterial differences of glutamic acid (P less than .10), valine (P less than .10), lysine (P less than .05) and histidine (P less than .01) were observed in soy-fed calves and isoleucine (P less than .10) in urea-fed calves from the peptide fraction. Negative venoarterial differences of amino acids from the peptide fraction were generally similar for the two dietary treatments, except for glutamic acid (P less than .01). A large negative venoarterial difference of glutamic acid was observed when soy protein was fed and, conversely, a large positive venoarterial difference was observed when urea was fed. Two groups of serum proteins were evaluated: fraction I (primarily globulins) and fraction II (primarily albumin). Hindlimb amino acid venoarterial concentration differences for soy-fed calves were inconsistent and nonsignificant for both fractions I and II. Conversely, hindlimb negative venoarterial concentration differences in urea-fed calves for both fractions I and II were large and statistically significant for many amino acids. Fraction II venoarterial differences were much greater than differences observed in fraction I. Soy and urea treatment differences were also much more pronounced in fraction II, being statistically significant for 8 of 17 amino acids. Eight male Holstein calves (body weight 68 +/- 5 kg; age 75 +/- 6 d), each with a permanent re-entrant pancreatic cannula and T-type ileal and duodenal cannulas, were used in a crossover design with four animals per group to determine amino acid kinetics and digestibilities in the digestive tract of calves fed soybean meal (SBM) and canola meal (CM) protein. The SBM and CM diets were fed twice daily at a level of 900 g at each feeding time (air-dry basis). With the exception of methionine, crude protein and amino acid flows at the proximal duodenum, expressed as a percentage of intake, were not influenced by dietary protein source. Apparent ileal and total tract digestibilities of CP and amino acids were reduced (P less than .05) by feeding CM compared to SBM, but apparent ileal digestibility of methionine was not affected by dietary protein source. Except for methionine, net disappearance of all amino acids in the small intestine, relative to the amount fed, was higher for the SBM diet than for the CM diet. Net disappearance or synthesis of amino acids in the large intestine were not affected by dietary protein source. Similarly, dietary protein source did not affect (P greater than .05) the secretion of pancreatic juice or concentrations of protein, chymotrypsin and trypsin in pancreatic juice. Soybean meal protein has higher ileal and total gastrointestinal tract digestibility than CM protein for young, growing calves.  相似文献

Eight male Holstein calves 7 to 10 d of age were fed a milk replacer containing a skim milk powder subjected to low-temperature drying either with or without addition of an oxalate-NaOH buffer known to prevent curd formation. The calves were used in a completely randomized design to study the effect of milk clotting on digestibility and blood parameters. Plasma glucose and plasma insulin were similar (P greater than .05) for the clotting and the nonclotting milk replacers. For both treatments, concentrations of glucose and insulin reached a peak 2 h postfeeding (P less than .01). Plasma triglycerides were higher (P less than .01) postfeeding for the nonclotting than for the clotting milk replacer. Plasma essential amino acids and plasma urea were higher, whereas plasma calcium was lower, for the nonclotting milk (P less than .01). Digestibility of dry matter, protein and fat was similar (P greater than .05) between clotting and nonclotting milk. The dry matter content of feces was not affected by clotting (P greater than .05). The data are interpreted to indicate that clotting of the milk replacer modifies blood concentrations of triglycerides, essential amino acids and urea without changing the digestibility of the diet.  相似文献

The aim of the present study was to investigate the relationships between serum amino acid profiles in normal and calves with Mycoplasma bronchopneumonia. Serum free amino acid concentrations in serum obtained from 34 calves with or without Mycoplasma bronchopneumonia were determined by high-performance liquid chromatography. The calves with Mycoplasma were characterized by significantly lower total amino acid and total essential amino acid concentrations and molar ratios of branched-chain amino acid (BCAA) to aromatic amino acid (BCAA/AAA) and BCAA to tyrosine (BTR), and by a significantly higher molar ratio of serine phosphorylation (SPR). The proposed diagnostic cutoffs for BCAA/AAA, BTR and SPR in serum based on ROC analysis for detection of catabolic states associated with Mycoplasma bronchopneumonia were set at <1.75, <2.86 and >0.85, respectively. Our results suggest that determining the profiles of amino acids, especially BTR and SPR, could provide useful diagnostic information in terms of predicting protein catabolism in Mycoplasma bronchopneumonia.  相似文献

A trial was conducted to examine the potential of using plasma amino acid responses to graded levels of escape protein to determine limiting amino acids in cattle. Growing calves (n = 120; mean BW = 220 +/- 21 kg) were fed a basal diet of corncob:sorghum silage (61:39) and were individually supplemented with distillers' dried grains (DDG), heat-damaged DDG (H-DDG), feather meal (FTH), or urea. The urea supplement was mixed with DDG and H-DDG to allow 0, 20, 35, 50, 65, or 80% of the supplemental CP to come from distillers' protein and maintain an 11.5% CP diet. Urea supplement was mixed with FTH to allow 0, 22, 39, 56, 73, or 90% of the supplemental CP to come from FTH. Dietary CP ranged from 11.5% at the 0% level to 17.3% at the 90% level. Plasma concentration of most essential plasma amino acids responded (P less than .10) linearly and(or) quadratically to increased escape protein. The broken-line response of plasma methionine at low DDG intake suggested that methionine was limiting at low levels of escape protein. An initial decrease followed by a plateau fit by a broken line indicated that histidine became limiting in FTH diets, and lysine eventually became limiting for DDG, H-DDG, and FTH diets before maximum BW gain was reached. Results indicate that plasma amino acid responses may identify amino acids that become limiting with increasing escape protein.  相似文献

Trials were conducted with four adult sheep of the Merino breed, live weight 45 to 50 kg, fed two diets with different nitrogen levels (7.6 g nitrogen per day as protein or 24 g nitrogen per day with an addition of 17.4 g urea nitrogen to 7.6 g nitrogen as protein). The following findings were obtained: the concentration of free amino acids in blood plasma in the 9th, 12th, 15th, 18th and 22nd weeks from the beginning of feeding with both diets was lower, in most of the analyzed amino acids, three hours after feeding than before feeding. The results show that at an increased intake of urea nitrogen the concentration increases of the free amino acids lysine, serine, glutamic acid, glycine, valine and isoleucine before feeding. Three hours after feeding these differences are less pronounced. An increase is only recorded in the concentration of arginine, glutamic acid and glycine, whereas the concentration of alanine decreases.  相似文献

本研究探讨了在日粮中添加尿素替代部分豆粕对山羊瘤胃发酵、上皮细胞增殖、凋亡和吸收转运能力的影响。将18 只波杂山羊随机分为3 组(n=6),分别饲喂3 种日粮:LC组(纯粗料)、MC组(30%精料)以及Urea组(1%DM缓释尿素替代部分豆粕+30%精料)。饲喂Urea组和MC组的山羊瘤胃中短链脂肪酸(SCFA)浓度、pH值均显著高于LC组,而该两组之间无显著差异;但Urea组在MC组的基础上进一步显著提高了瘤胃NH3和血浆尿素氮(BUN)浓度。因此日粮添加尿素对于瘤胃上皮中受瘤胃SCFA浓度、pH值调节的上皮生长、细胞周期、增殖凋亡相关基因和SCFA转运载体mRNA表达的影响与MC组相似,但对受瘤胃NH3调节的尿素转运、细胞内pH(pHi)调节相关蛋白mRNA表达则有显著的抑制效果,即显著高于LC组,但低于MC组。  相似文献

Venoarterial differences in the amino acid composition of serum proteins were measured across the hindlimbs of growing Holstein steer calves (147 kg) surgically fitted with abdominal aorta and caudal vena cava cannulae. Steers were maintained in near steady-state metabolism by feeding at hourly intervals in a continuously lighted environment. The steady-state blood samples were obtained 10 d post-surgery; then the steers were fasted and resampled. Arterial and venous serum proteins were chromatographically separated into four fractions and the amino acid composition of each was determined. Under steady-state conditions, there appeared to be a tendency for serum protein amino acids to appear on the venous side of the tissue in either greater or lesser concentrations than on the arterial side, depending upon the protein fraction. However, after the steers had fasted for 72 h, virtually all amino acids from all protein fractions were present in greater concentrations in venous serum than in arterial serum. Furthermore, the quantitative differences were several-fold larger than during steady-state sampling. It appears that serum proteins may contribute to the transport of amino acids to and from peripheral tissues. Data further suggest that all serum proteins do not appear to function in the same manner, and physiological state influences this process.  相似文献

Supplementation of the diets of rats with branched chain amino acids (BCAA: leucine, isoleucine, valine) did not alter their susceptibility to chronic poisoning by tansy ragwort (Senecio jacobaea), which contains hepatotoxic pyrrolizidine alkaloids (PA). Phenobarbital in the diet, which alters liver microsomal enzyme activity, also did not alter susceptibility to PA poisoning. A combination of butylated hydroxyanisole (BHA), cysteine and BCAA did increase (P less than .05) survival time of rats fed tansy ragwort. Dietary BHA and cysteine increased the survival time of rats injected with the PA monocrotaline, with evidence that addition of vitamin B12 and folic acid improved the effectiveness of this treatment. In a chronic feeding trial with tansy ragwort, a combination of BHA and cysteine increased (P less than .05) the survival times of rats, showing protective activity against PA poisoning. A mixture of B-complex vitamins, or vitamin B12-folic acid, was not effective in improving the response.  相似文献

The addition of graded amounts of L-lysine to a basal diet of cereals and groundnut meal given to growing pigs caused the expected improvements in rate of growth, N retention and carcass quality. Best responses were obtained when the diet contained 9 g lysine/kg; above that level there were no significant additional responses.Blood samples were taken from the anterior vena cava before and at intervals after a meal of each diet, and the concentration of amino acids and urea in the plasma determined. The concentrations of most amino acids in blood plasma increased after a meal, reaching maxima about 2 h after feeding.At all times of sampling the concentrations in plasma of essential amino acids other than lysine were largely unaffected by the lysine content of the diet, but the concentration of lysine increased linearly over a wide range of lysine intake.The concentration of urea in blood plasma fell as the lysine content of the diet increased. Lowest concentrations were found with diets containing 9 g or more lysine/kg.It was concluded that measurement of the concentrations of urea in blood plasma of growing pigs fed twice daily may be of more value in assessing the amino acid requirements and efficiency of protein utilization than determination of changes in concentration of blood plasma amino acids.  相似文献

Two metabolism trials were conducted with 12 yearling crossbred wethers per trial (34 and 38 kg for trials 1 and 2, respectively). The wethers, equipped with ruminal, abomasal and ileal cannulae, were randomly allotted for each trial to the following treatments: 1) hay alone or hay supplemented with 2) .9% urea, 3) 1% urea and 6.5% molasses or 4) 1% urea and 5.2% corn. Two digestive flow markers were used: Cr2O3 powder and Co-ethylenediaminetetraacetic acid (Co-EDTA). Urea and Co-EDTA were infused continuously into the rumen via cannula. Daily dry matter (DM) intake averaged 517 g. Urea supplementation improved N retention (P less than .01). Apparent digestibility of DM, acid detergent fiber (ADF) and energy was not affected by treatment. Urea and carbohydrate supplementation increased ruminal propionic acid molar proportions (P less than .05). Apparent ruminal DM digestion accounted for 41% of the total DM degraded, whereas 77.4% of the digestible ADF was degraded in the rumen. Urea supplementation increased ADF digestion in the large intestine (P less than .01). Urea and carbohydrate supplementation resulted in a stepwise increase in N flowing with the liquid phase at the abomasum. Mean retention times of the solid and liquid phases of digestive contents were similar across treatments. Overall, benefits of supplementation of poor-quality fescue hay diets by small amounts of urea and readily available carbohydrates remain questionable for sheep fed at a fixed level of intake below maintenance.  相似文献

In order to examine the relationship between white scour and fatty acids, we added fatty acids to the milk replacer. Twenty healthy Holstein calves were divided into 4 groups, five calves per group; a control group with no fortified fatty acid, and 3 groups fed either with oleic, stearic, or palmitic acid, respectively. The calves were fed milk replacer (5% of the calf's body weight) twice a day but the fatty acids (2 wt % of milk replacer) were added only once. The fecal and blood samples were obtained at 0, 12, 24, 36, and 48 h after feeding of the acids. All five calves in the palmitic acid group, and 3 out of 5 calves each in the stealic and the oleic acid groups had whitish feces after feeding fatty acid. The stearic acid group had a significantly elevated stearic acid concentration in the feces during 24–36 h compared to the pre-feeding level. The fecal concentration of palmitic acid increased significantly at 24–36 h in the palmitic acid group. We concluded that the long-chain saturated fatty acids are one of the causes of white scour in calves.  相似文献

Five sheep (average BW 62 kg) were fed 65% roughage: 35% concentrate diets (CP = 15%) in a 5 x 5 Latin square design to study the effects of combinations of defaunation and N supplements (soybean meal [SBM], corn gluten meal [CGM], blood meal [BM], urea, and casein) differing in ruminal degradation on ruminal microbial numbers and activity. Diets were fed twice daily (DM intake 1,759 g/d). Defaunation was accomplished with doses of 30 ml of alkanate 3SL3.sheep-1.d-1 for 3 d with 2 d of fasting. Treatment 1 (control) involved feeding faunated sheep a diet in which the supplemental N was 67% SBM N and 33% urea N. Treatment 2 involved feeding defaunated sheep the same diet as the control. Treatments 3, 4, and 5 involved feeding defaunated sheep diets in which the supplemental N source was either 67% CGM-BM N (CGM and BM combined on a 1:1 N ratio): 33% urea N, or 33% CGM-BM N:67% urea N or 33% CGM-BM N:33% urea N:33% casein N, respectively. Compared with the faunated control, defaunation (Treatments 2, 3, 4, and 5) increased (P less than .05) total direct counts of ruminal bacteria (2.7 vs 1.3 x 10(11)/ml), fungal zoospores (2.8 vs 1.4 x 10(5)/ml), and ruminal microbial protease activity (1.4 vs 1.0 mg azocasein/[ml ruminal fluid.h]). Defaunation did not have a consistent effect on ruminal microbial deaminase activity. Compared with the control, defaunation resulted in lower (P less than .05) total perchloric acid-soluble amino N in ruminal fluid at 4 and 10 h after the morning feeding. Defaunation did not decrease (P greater than .05) total free amino acid concentrations in ruminal fluid, but it altered the profile of free amino acids. Although defaunation increased (P less than .05) ruminal bacterial numbers, no increases in total microbial CP or OM concentrations in ruminal contents were observed.  相似文献

Previous research has demonstrated that increasing the CP concentration from 16 to 26% in milk replacers fed to male preruminant dairy calves at 1.5% of BW (DM basis) daily resulted in increased ADG, G:F, and deposition of lean tissue. However, the effects of dietary CP would be expected to vary depending on ME intake. Here, male Holstein calves < 1 wk old were used to determine the effects of feeding rate and CP concentration of isocaloric, whey protein-based milk replacers on growth and body composition. After a 2-wk standardization period, calves were assigned randomly to an initial baseline group or to treatments in a 2 x 4 factorial arrangement of feeding rate (1.25 or 1.75% of BW daily, DM basis) and milk replacer CP concentration (14, 18, 22, or 26% of DM). No starter was offered, but calves had free access to water. After a 5-wk feeding period, calves were slaughtered and body composition was determined. Increasing the feeding rate increased (P < 0.05) ADG, G:F, empty-body gains of chemical components and energy, the percentage of fat in empty BW gain and in the final empty body, and concentrations of IGF-I and insulin in plasma. Increasing the feeding rate decreased (P < 0.01) percentages of water and protein in the empty body and decreased urea N in plasma. Increasing dietary CP concentration linearly increased (P < 0.05) ADG, body length, heart girth, and gains of water and protein but linearly decreased (P < 0.05) fat gain. As dietary CP increased, fat content in empty body gain decreased, and water and protein increased. Increasing CP concentration increased (quadratic, P < 0.02) G:F, with greatest efficiencies for calves fed 22% CP. Gross energetic efficiency (retained energy:intake energy) was greater (P < 0.05) for calves fed at 1.75% of BW daily. Efficiency of dietary protein use for protein gain was greater for calves fed at 1.75% of BW daily but was not affected by dietary CP. The ratio of protein gain to apparently digestible protein intake above maintenance decreased as dietary CP increased. Interactions (P < 0.05) of feeding rate and CP concentration for gains of water and protein indicated that when dietary CP was 26% the ME supply limited protein use by calves fed at 1.25% of BW daily. Body composition of preruminant calves can be markedly altered by manipulating the protein to energy ratio in milk replacers. These dietary effects on body composition and growth are not predicted by current NRC standards.  相似文献

A total 262 Angus steer calves averaging 207 kg from none (trial 1) and five farms (trial 2) were identified on the farm of origin and one-half of the calves on each farm was implanted with 36 mg of zeranol before entering the feeder calf assembly and marketing system. Calves were assembled, fasted for 24 h and fed hay for 72 h, then shipped 1,368 km. Upon arrival, the following receiving diets were fed for 4 wk to one-third of the calves from each farm and implant group: 1) control diet, 2) high-potassium diet, 3) escape-protein diet using corn gluten meal, which can escape ruminal degradation as a protein source. Implanted calves gained more weight (P less than .10) than nonimplanted calves while on the farm, but changes in weight during assembly, transit and the 28-d receiving period were not different (P more than .10). Plasma glucose and total protein concentrations were higher (P less than .05), while urea nitrogen concentration was lower (P less than .05) for implanted calves following transit. Increasing the potassium concentration of the receiving diet increased (P less than .05) post-transit weight gains, but had no affect on dry matter consumption. Corn gluten meal replaced soybean meal in the escape-protein diet and resulted in similar weight gains. Plasma urea nitrogen concentrations were higher (P less than .05) at 2 wk post-transit in the escape-protein diet group as compared with the control group. Receiving diet had no effect on the incidence or severity of bovine respiratory disease, but affected (P less than .05) performance during the subsequent 69-d stocker phase of ad libitum access to medium quality hay and 2.27 kg of supplement per day.  相似文献

Plasma glucose concentrations in neonates are influenced by colostrum feeding and by glucocorticoids. We have tested whether a high-glucocorticoid status after birth, as well as colostrum feeding, influences glucose metabolism in association with changes of hepatic expression and activities of gluconeogenic enzymes phosphoenolpyruvate carboxykinase (PEPCK; EC 4.1.1.32) and pyruvate carboxylase (PC; EC 6.4.1.1) in neonatal calves. Calves (n = 14 per group) were fed either colostrum or a milk-based formula with nutrient and energy contents similar to colostrum. Half the calves in each feeding group were treated with dexamethasone (DEXA; 30 microg/[kg BW x d]). Pre- and postprandial blood samples were taken on d 1, 2, 4, and 5 and liver samples were collected on d 5 of life. Dexamethasone treatment increased (P < or = 0.05) plasma concentrations of glucose, insulin, and glucagon more in colostrum-fed than in formula-fed calves but increased (P < or = 0.05) urea concentrations and decreased (P < or = 0.05) concentrations of NEFA, ACTH, and cortisol independent of colostrum vs. formula feeding. Colostrum feeding increased (P < 0.05) plasma glucose, but decreased (P < 0.05) plasma urea concentrations. Glucagon-to-insulin ratios in DEXA-treated and colostrum-fed calves were decreased (P < 0.05). Dexamethasone treatment decreased hepatic mRNA levels and activities of PC (P < 0.001 and P < 0.10) and activities of PEPCK (P < 0.001) but increased (P < 0.001) the glycogen content. Colostrum feeding increased (P < 0.05) mitochondrial PEPCK mRNA levels and PEPCK activities in calves not treated with DEXA but decreased (P < 0.1) amounts of PC mRNA. In conclusion, increased plasma glucose concentrations after DEXA treatment were not associated with a stimulation of hepatic gluconeogenic enzyme activities; however, colostrum feeding probably raised plasma glucose concentrations because of increased hepatic gluconeogenic activities.  相似文献

The effect of different ratios of urea to amino acid N at a fixed concentration of soluble sugars as energy source and varying levels of soluble sugars at optimum urea cell suspension was obtained from the rumen fluid of buffalo (Bubalus bubalis) calves fed on a growth ration. Under glucose fermentation, the bacterial protein content of the incubation mixture (I. M.) was increased to 3.91, 6.31 and 5.08 times the control value (urea alone) when 25, 50 and 75% of urea-N was replaced with amino acid N, respectively. With cellobiose, the corresponding increase was 4.06, 5.29 and 5.63 times. At 50% urea-N replacement with amino acid N (a ratio for maximum protein synthesis), the bacterial content was maximum when 1 g glucose or cellobiose per 100 ml of I. M. was added. Per cent incorporation of radioactivity from amino acids into bacterial protein was maximum at 25% amino acid N level with both the soluble sugar sources. The total amino acids incorporated into bacterial protein were, however, more at 50% than at 25% amino acid N level.  相似文献

Brahman calves (n = 28) were used to evaluate the effect of environmental temperature during the 1st or 2nd d after birth. Calves were removed from their dams within 30 min of birth (newborn; D0) before suckling or at 20 h of age and fasted for 4 h before treatment (day-old; D1). Calves were placed in either a warm (W; 25 degrees C) or a cold (C; 5 degrees C) environment for 2 h and either maintained in or transferred to, respectively, W for 22 h. Blood samples were collected via jugular catheters at 15-min intervals beginning at initial placement in W or C through 3 h and at 4, 5, 6, 8, 10, 12, 14, 18, and 24 h. Rectal temperature (Tr) was recorded with each sample. Following the 60-min and 12-h samples, each calf was administered 1 liter of colostrum from its dam. Serum or plasma was analyzed for glucose, lactate, plasma urea nitrogen, triglycerides, nonesterified fatty acids, insulin, cortisol, triiodothyronine (T3), and thyroxine (T4). Rectal temperature of D0C calves was lower (P less than .05) than that of other calves from 75 min through 3 h. Insulin, lactate, T3, and plasma urea nitrogen concentrations were not different among all calves. Higher (P less than .01) cortisol and T4 concentrations were observed in D0 than in D1 calves. Cortisol (P less than .008) and nonesterified fatty acid (P less than .05) levels were greater in C than in W calves. This study investigated effects of birth weight and postnatal nutrition on regulation of energy metabolism in the neonatal lamb. Low (mean +/- SD 2.289 +/- 0.341 kg, n = 28) and high (4.840 +/- 0.446 kg, n = 20) birth weight male Suffolk x (Finnsheep x Dorset) lambs were individually reared on a liquid diet to grow rapidly (ad libitum fed, ADG = 337 g, n = 20) or slowly (ADG = 150 g, n = 20) from birth to live weights (LW) up to approximately 20 kg. At birth, small newborns had higher plasma concentrations of urea nitrogen (mean +/- SEM 8.31 +/- 0.25 vs 6.39 +/- 0.32 mM, P = 0.002) and somatotropin (ST, 49.1 +/- 17.0 vs 10.8 +/- 4.3 ng/mL, P = .045) and lower IGF-I (36.1 +/- 6.8 vs 157.7 +/- 21.8 ng/mL, P < 0.001) than large newborns. Plasma glucose (1.42 +/- 0.23 vs 2.63 +/- 0.95 mM, P = 0.147) and insulin (0.09 +/- 0.02 vs 0.13 +/- 0.06 ng/mL, P = 0.264) concentrations did not differ. Urea nitrogen concentration in plasma peaked and then declined rapidly in all lambs during the first week postpartum, and plasma ST declined on a body-weight-related basis from birth. During rearing to 20 kg LW, plasma insulin was higher in low- vs high-birth-weight lambs. Lambs fed ad libitum had greater plasma concentrations of glucose, urea nitrogen, insulin, and IGF-I compared to those fed a restricted diet (ADG = 150 g). The results suggest that during the early postpartum period, newborn lambs exhibit the fetal characteristic of high rates of amino acid oxidation. The results also support the notion that, at birth, low-birth-weight lambs are less mature than high-birth-weight lambs in aspects of metabolic and endocrine development, which may enhance their capacity to utilize amino acids for energy production and to support gluconeogenesis during the immediate postpartum period. Being small at birth also resulted in elevated plasma insulin concentrations when adequate nutriment to support moderate or rapid growth was provided postpartum, although it remains to be elucidated whether this more chronic effect persists in the longer term.  相似文献
