Glutathione S-transferases (GSTs) are important detoxification enzymes in insects. This study examined the inhibitory effects of five insecticides (beta-cypermethrin, fenpropathrin, phoxim, abamectin, acetamiprid) and two plant-derived allelochemicals (quercetin, tannic acid) on GST activity in Micromelalopha troglodyta (Graeser) and Clostera anachoreta (Fabricius). GST activities were suppressed by all the insecticides and allelochemicals tested; the two allelochemicals were the most potent inhibitors, reducing GST activity in vitro in a dosage-dependent manner. Kinetic analyses of GST inhibition by quercetin and tannic acid in M. troglodyta and C. anachoreta suggested biochemical differences in their GSTs. 相似文献

In lindane-treated house flies, a cis-dehydrogenated metabolite, , was identified by gas-liquid chromatography and mass spectrometry. The in vitro metabolism study showed that in the presence of NADPH the microsomal fraction of house flies converted lindane to three hexane-soluble metabolites. This conversion was inhibited by piperonyl butoxide, SKF-525A, and carbon monoxide. These metabolites were identified as , (PCCHE) by gas-liquid chromatography. They, as well as lindane, were excellent substrates for the reaction with the postmicrosomal fraction in the presence of glutathione. While the reaction with lindane-d6 showed a significant deuterium isotope effect (6.82), that of did not (1.18). Enzymatic conjugation with glutathione probably occurs at the stage of PCCHE. 相似文献

This paper describes a method for the determination of fenoxaprop ethyl and fenoxaprop residues in four soil types using two extraction procedures: extraction with 0.1 M hydrochloric acid/methanol, partitioning with dichloromethane and extraction with ethyl acetate. The extracts were purified on florisil or alumina cartridge. The analyses were performed by reverse phase HPLC with UV detection at 280 nm. The best results in terms of recovery, clean-up efficiency and independence of soil characteristics were obtained with the combination ethyl acetate extraction alumina clean-up. Under these conditions, the recoveries were higher than 70 % and the detection limit was 0.02 mg kg?1 soil. 相似文献

The relationship between glutathione S-transferase activity toward 3,4-dichloronitrobenzene and O-alkyl or O-aryl conjugation of diazinon was investigated in eight strains of house flies. No significant difference was found in the amount of O-aryl conjugation. In contrast, house flies which had higher glutathione S-transferase activity toward 3,4-dichloronitrobenzene also had higher O-alkyl conjugating activity toward diazinon. The glutathione S-transferase(s) in phenobarbital-pretreated flies degraded diazinon faster than those in the nontreated ones. The present results showed that the formation of the O-alkyl conjugate was enhanced by phenobarbital pretreatment, while the formation of the O-aryl conjugate was not affected by induction. Based on these findings, it would appear that one of the multiple forms of glutathione S-transferase is specifically induced and responsible for the increase in O-alkyl conjugation. 相似文献

The separate enantiomers of diclofop-methyl and fenoxaprop-ethyl were incubated with two soils under aerobic conditions. Rapid hydrolysis to the corresponding acids diclofop and fenoxaprop was observed. At various time intervals after soil treatment the parent compound and its free acid were extracted and the extracts were investigated by high-performance liquid chromatography. The determination of the K:S-ratio using a chiral stationary phase revealed that no racemisation of the esters took place. However, inversion of the hydrolysis products was observed. In both soils the acids with the S-configuration showed a significantly higher inversion tendency than the acids with the R-configuration. In one soil the S-acids of both herbicides were inverted (50% inversion in 4–7 days) to yield, ultimately, a residue containing 70% R- and 30% S-enantiomer. 相似文献

Herbicide‐resistant populations of Alopecurus myosuroides (black grass) have become widespread throughout the UK since the early 1980s. Previous observations in this laboratory have demonstrated that natural climatic fluctuations caused increases in endogenous glutathione S‐transferase (GST) enzyme activity in A. myosuroides plants as they mature, which is thought to be linked to herbicide resistance in this species. The present study has investigated the effects of plant growth at 10°C and 25°C, and reports GST specific activity and glutathione (GSH) pool size in resistant and susceptible A. myosuroides biotypes. Findings demonstrate differences in GST activity between resistant and susceptible populations, which are transient at lower growth temperatures. The GSH pool size was elevated at lower growth temperature in both biotypes. We speculate that these endogenous responses are part of a natural mechanism of acclimation to environmental change in this species and suggest that resistant plants are more able to adapt to environmental stress, as indicated in this instance by temperature change. These observations imply that the control of resistant A. myosuroides by graminicides may be more effective when applied at lower temperatures and at earlier growth stages. 相似文献

In vitro assay procedures for measuring the activity of cysteine biosynthesis from serine (CBS), which is a coupled reaction catalyzed by serine acetyltransferase and cysteine synthase, were developed using crude extracts from sorghum shoots. Cysteine biosynthesis from serine activity was dependent on acetyl‐CoA concentrations (up to 1.5 mmol L?1), serine (at least up to 20 mmol L?1) and sulfide (up to 0.25 mmol L?1), respectively, and was proportional to the protein concentration in the reaction mixture below 0.4 mg mL?1. The reaction rate was 6.6 nmol min?1 per mg of protein during the first 5 min, but increased to 45.6 nmol min?1 per mg of protein between 30 and 45 min after reaction initiation. Sorghum had the highest CBS total activity (222.4 nmol min?1 per g of fresh weight), and large crabgrass had the lowest CBS total activity (4.7 nmol min?1 per g of fresh weight) when CBS activity in shoots was extracted from sorghum, corn, johnsongrass, barnyardgrass, goosegrass, green foxtail and large crabgrass. Similar results were obtained for CBS specific activity (nmol min?1 per mg of protein). There was no correlation between total CBS activity and susceptibility to metolachlor; however, when corn was excluded, a correlation of R2 = 0.690 was found. Flurazole seed treatment (1.25 g per kg of seed) conferred metolachlor resistance by sorghum, and enhanced total CBS activity and non‐protein thiol content by 27 and 61%, respectively. The increase in thiol content presumably contributed to metolachlor tolerance in sorghum. From these results, the difference in CBS activity partially contributes to the selectivity to metolachlor among certain grass species, and to the safening action of flurazole by increasing thiol content. 相似文献

The pattern and extent of 14C-glyphosate [N-(phosphonomethyl)glycine] translocation from the treated leaf and metabolism of 14C-glyphosate were studied in field bindweed (Convolvulus arvensis L.), hedge bindweed (Convolvulus sepium L.). Canada thistle [Cirsium arvense (L.) Scop.] tall morning glory [lpomoea purpurea (L.) Roth.] and wild buckwheat (Polygonum convolvulus L.). 14C was translocated throughout the plants within 3 days with accumulation in the meristematic tips of the roots and shoots evident. Cross and longitudinal sections of stems and roots showed that the 14C was localized in the phloem. Field bindweed translocated 3–5% of the applied 14C from the treated leaf, hedge bindweed 21.6%, Canada thistle 7.8%, tall morningglory 6.5%, and wild buckwheat 5%. Field bindweed, Canada thistle, and tall morningglory metabolized the parent glyphosate to aminomethylphosphonic acid to a limited extent. This metabolite made up less than 15% of the total 14C. Of the total 14C applied to excised leaves, 50% had disappeared within 25 days. 相似文献

采用自动记录装置测试了黑龙江地区6种主要农业害鼠的昼夜活动节律和活动强度。结果表明,达乌尔黄鼠为昼行性鼠种,中午时段出现明显的活动间歇期以避开强烈的日晒;莫氏田鼠为昼夜活动型鼠种,日夜活动强度差异不显著,与其生活于草滩底部营食草根的生活方式相适应。大仓鼠、小家鼠、褐家鼠和黑线姬鼠都为夜行性鼠种,其中以小家鼠和黑线姬鼠最为典型,而大仓鼠和褐家鼠在日间都具有相当强度的活动,褐家鼠则是供试鼠种中唯一在各个时段都有活动的夜行鼠种。6种供试鼠种的主要活动特征与前人研究的结果基本相符,但其活动节律与其他研究结果存在活动高峰时间、活动强度等差异,表现出这些鼠种与黑龙江地区特定气候条件的适应。 相似文献

The influence of water stress on the absorption and translocation of 14C-labelled fenoxapropethyl and imazamthabenz-methyl in Avena fatua L. (wild oat) was studied. The phytoioxicity to A. fatua of both herbicides with a droplet application was also examined under water stress conditions. The absorption of both fenoxaproethyl and imazamethabenz-methyl was reduced by waler stress when the plants were harvested within 24 h after herbicide application. Up to 48 h after the application, the translocation out of the treated lamina of both herbicides, based on percentage of applied 14C. was reduced under water stress conditions. When havested 96 h after herbicide application, however, water stress no longer significantly affeaed the absorption and translocation of either herbicide. When the herbicides were applied as individual droplets, water stress reduced the phytotoxicity of fenoxaprop-ethyl but not that of imazamethabenz-methyl. It is concluded that the changes in herbieide absorption and translocation may not be the major physiological processes associated with differential whole-plant response oi A faiua to fenoxaprop-ethyl and imazamefhabenz-methyl under water stress. 相似文献

Fusarium rot of melon, caused by species of the genus Fusarium, has become an important postharvest disease for many Brazilian producers. Due to the delayed onset of symptoms, this disease is often only detected when fruits arrive at the importing country, thus generating economic loss for the exportation of the fruit. This study was developed with the aim of investigating which Fusarium species cause fruit rot in melon and to evaluate any differences in aggressiveness and development of symptoms. Species were identified through phylogenetic analysis of two loci and morphological markers. The 28 isolates obtained from diseased melon fruits of different commercial cultivars were identified as Fusarium falciforme (FSSC), F. sulawesiense, F. pernambucanum (FIESC), and F. kalimantanense (FOSC). Three isolates belong to a new phylogenetic lineage within the F. fujikuroi species complex (FFSC). All isolates were tested for pathogenicity, and first symptoms of rot in Canary melon were observed 2 days after inoculation. Isolates of F. falciforme and F. sulawesiense were shown to be more aggressive. Our results extend information on Fusarium species that cause fruit rot in melon and support the development of management strategies, as there is currently no efficient control for this disease. To our knowledge, this is the first report of the occurrence of species of the FSSC, FOSC, and FFSC from muskmelon fruits in Brazil. 相似文献

The toxicity of carbaryl to three species of fleshflies Sarcophaga bullata Parker, S. crassipalpis Macquart, and S. argyrostoma (Robineau-Desvoidy) varied considerably with age and sex. In contrast, the susceptibility of a blowfly, Phormia regina (Meigen) to carbaryl decreased with age and that of two muscid flies, Musca autumnalis DeGeer and Stomoxys calcitrans (L.), remained relatively constant. The synergistic activity of piperonyl butoxide varied inversely with the innate toxicity of carbaryl to each species suggesting that the observed age- and sex-dependent variations in carbaryl toxicity result mainly from differences in detoxifying capability. This was supported by in vitro measurements of oxidative microsomal metabolism. It was further established that differences in the rates of penetration and excretion of carbaryl and in the susceptibility of the head cholinesterase to carbaryl inhibition were of little importance in determining the susceptibility of the flies to this insecticide. 相似文献
