A seroepidemiological study of border disease was conducted in sheep and goats in various areas of Quebec. Sera of 10% of animals of selected flocks were collected and specific antibodies against bovine viral diarrhea- mucosal disease were tested by seroneutralization. Results show that 10.9% and 16% of sheep and goats respectively gave a positive reaction. The lower serological prevalence was found in sheep flocks of the Sherbrooke area (5.4%) while the highest percentage of positive sera was observed in the Quebec area (24.7%). The prevalence in goats varied according to areas (2.6 to 45.5%). No relation was observed in seropositive animals between age, sex, breed and the presence of abortions in the flocks. Our serological results indicate that border disease is probably present in Quebec sheep and goat flocks but that the clinical diagnosis of this disease is not well established.  相似文献

In this study serological investigations were performed to determine the prevalence of pestiviral infections in sheep in one Federal State of Austria, namely Carinthia. 1527 blood samples from sheep in 147 flocks were collected and tested by Enzyme-linked immunosorbent assay and virus-neutralisation tests for antibodies to ruminant pestiviruses. The estimated flock prevalence was 47.6%, the individual prevalence 16.3%. Significant geographical variations in the flock as well in the individual prevalence were found. The highest prevalence in sheep and in sheep flocks was established in the region Spittal/Drau with 25.9% and 69.7%.The individual and the flock prevalence was significantly higher on farms where cattle or sheep from other farms were present than on farms with no cattle (p < 0.017). All Enzyme-linked immunosorbent assay positive sera were tested for Bovine viral diarrhea virus-1 (strain NADL), Bovine viral diarrhea virus-2 (strain 125) and for Border disease virus (strain MOREDUN) by virus neutralisation tests. Seventy out of 249 positive samples revealed the highest titres (> or = two-fold) to Bovine viral diarrhea virus-1 and 25 to Border disease virus. The remaining positive samples did not show clear results because of cross reactions.  相似文献

883 sheep from 68 flocks were tested for the prevalence of antibodies against Maedi/Visna-(M/V)virus. As serological method the Agar Gel Immunodiffusion Test (AGID) was performed, 9.5% of the blood samples were positive against M/V, 1.7% showed questionable results. The highest incidence of serum-antibodies showed the Texel (47.7%) and Milk sheep (26.1%). No antibody titers were found in Mountain and Suffolk sheep. Merino sheep had antibody titers against M/V in 2.6% and Mountain sheep crossbreeds in 1.7%.  相似文献

Two hundred and fifty-five biological samples (106 aborted foetal tissue samples and 149 blood samples from aborted sheep and goats) were collected from 188 animals during the lambing season from September 2009 to April 2010 from the Mafraq region of Jordan. The sampled animals belonged to 93 goat and sheep flocks that had cases of abortion. A total of 169 (66.3%) biological samples were collected from sheep and 86 (33.7%) from goats. Seventy-six (29.8%) biological samples (45 blood and 31 tissue samples) were positive for Toxoplasma gondii by PCR assay. The positive samples were obtained from 43 sheep and 23 goats. The overall toxoplasma-specific prevalence rate was 35.1% (66/188). Forty flocks (43%) had at least one T. gondii PCR-positive animal. The risk factors related to flock health status and farm management that are hypothesized to be associated with T. gondii PCR positivity were also assessed using multiple logistic regressions. The presence of cats (OR = 4.74), a large flock size (OR = 2.76) and the method of disposing the aborted foetuses (OR = 3.77) were all statistically significant (P < 0.05) risk factors that were positively associated with toxoplasma positivity in goat and sheep flocks.  相似文献

Sera from 155 ewes representing 21 different sheep flocks from the central parts of Sweden were examined for the presence of antibodies to Toxoplasma gondii by the indirect fluorescent-antibody test. Seropositive animals were found in 14 flocks, which corresponded to 55 % of the tested sera. Animals from the same flock were with few exceptions either all positive or all negative. Some ewes with a history of abortion a few months before blood sampling showed high titers indicating causative correlation with toxoplasmosis. The diagnostic performance of an ELISA for the detection of antibodies to the small ruminant lentiviruses (SRLVs) maedi-visna virus and caprine arthritis-encephalitis virus in milk and corresponding blood samples was evaluated in 50 sheep. The agreement between ELISA results in blood and milk was 90 per cent, and the κ value was 0.79. In addition, a serological survey in the central zone of Spain was performed using milk samples from 413 animals (250 sheep and 163 goats) from 12 flocks/herds. All flocks/herds had some animals that were positive for SRLV. Among the animals, 60.0 per cent of the sheep and 8.0 per cent of the goats tested were seropositive. Each sample was also tested using a PCR technique, which increased the percentage of positive animals detected. Using a combination of ELISA and PCR gave a total of 72.2 per cent of sheep and 28.8 per cent of goats positive for SRLV.  相似文献

ABSTRACT

Aims: To determine whether sheep that co-grazed with cattle that were suspected to be positive for bovine viral diarrhoea (BVD) virus had serological evidence of exposure to the virus.

Methods: Eighteen commercial farms that routinely co-grazed cattle and sheep in the same paddocks were recruited through purposive sampling. The recruiting veterinarians identified nine farms with cattle herds that were known or highly suspected to be positive for BVD and nine farms that were considered to be free of BVD. Blood samples were taken from 15 ewes aged 1 year on each farm and samples were submitted to a commercial diagnostic laboratory to test for antibodies against pestiviruses using an ELISA. All samples that were positive were then tested using a virus neutralisation test (VNT)for antibodies against BVD virus.

Results: Of the 270 blood samples, 17 were positive for pestivirus antibodies by ELISA and these originated from two farms that were known or suspected to have BVD virus-positive cattle. None of the samples from the nine flocks co-grazed with cattle herds that were known or suspected to be BVD virus-negative were positive for pestivirus antibodies. Within the two positive farms, 2/15 samples from the first farm and 15/15 samples from the second farm were antibody-positive. When the 17 positive blood samples were submitted for VNT, all 15 samples from the second farm tested positive for BVD virus antibodies with the highest titre being 1:512.

Conclusions and clinical relevance: In this small sample of New Zealand sheep and beef farms with suspected BVD infection in cattle, there was evidence of pestivirus exposure in co-grazed sheep. Although we were unable to confirm the origin of the exposure in these sheep, these findings highlight that farmers who are trying to eradicate BVD from their cattle should be mindful that the infection may also be circulating in sheep, and both populations should be considered a possible risk to each other for generating transient and persistent infections. Further work is needed to estimate the true prevalence of New Zealand sheep flocks that are affected by BVD and the associated economic impacts.  相似文献

A serological survey of Toxoplasma gondii infection in adult breeding sheep in Great Britain was conducted using surplus sera taken during a seroprevalence study of Brucella melitensis in 2009. Of the 3539 sera collected from 227 flocks, 2619 (74 per cent) were found to be positive for T gondii specific antibody when tested using latex agglutination. Multilevel logistic modelling suggested that the likelihood of infection increased with age and this effect appeared to be amplified in animals vaccinated against T gondii. The model also indicated that the odds of sheep being seropositive were increased on premises where cattle were also kept. These results suggest a high level of Toxoplasma infection in breeding sheep in Great Britain and provide further evidence to suggest that postnatal infection is more common than congenital infection in sheep.  相似文献

The contents of toxoplasma antibodies were estimated by a micro-modification of the dye test for up to 3 years after the initial stage of infection both in ewes which had aborted and in ewes with normal pregnancies. During the first 2 years the titres in ewes which had aborted were significantly higher than in ewes with normal pregnancies. The investigation indicated that dye test titres ≥ 1/512 usually occur during the first stage of infection, and are mainly found in ewes with clinical toxoplasmosis.The dye test titres in lambs due to antibodies transferred with the colostrum were up to 4 twofold dilutions higher than in their dams during the first 2 days after birth. Later the titres declined, and at the age of about 2 months only 3 of 29 lambs had higher titres than their dams. After the age of about 3 months maternally derived antibodies were not detected.The contents of toxoplasma antibodies in sheep with listeric encephalitis were nearly the same as found by a serological survey of the local sheep population. The examination indicated that the dye test titres in sheep are little influenced by conditions that may affect the defence mechanism.Sheep with haemoglobin type B had significantly higher dye test titres than sheep with the haemoglobin types A and AB when examined less than about 6 months after they had acquired the infection. Serum samples from 704 animals from 54 Swedish sheep flocks were analysed by ELISA twice during 1 breeding season for antibodies to Toxoplasma gondii and border disease virus (BDV). An ELISA, originally developed for the detection of antibodies to bovine viral diarrhoea virus (BVDV) in cattle, was assessed on sheep sera and the results were compared with those obtained in a virus neutralization test. The correlation between the 2 assays proved good. Before breeding, 132 (19%) sheep in 42 flocks had antibodies to T. gondii and 7 (1%) sheep in 5 flocks were seropositive to BDV. During the observation period 4 sheep seroconverted to T. gondii and 13 to BDV, giving an incidence rate of 0.7% and 1.9% respectively. No clinical signs due to the infections were observed. In 5 flocks the frequency of barrenness, abortion or stillbirths exceeded 5%, 5% and 8%, respectively, but there was no evidence that this was attributable to the agents studied. The proportion of BDV-positive flocks was significantly higher among flocks that had been in contact with cattle than among those that had not.  相似文献

A serological survey was conducted in an attempt to detect antibodies against bovine respiratory viruses in sheep and goats from seven geographical areas of Quebec. Sera from 10% of the animals in 182 sheep flocks and 40 goat flocks were collected and specific antibodies against parainfluenza-3, reovirus type 3, respiratory syncytial and infectious bovine rhinotracheitis viruses were detected by hemagglutination-inhibition tests for the former viruses and complement fixation and seroneutralization assays for the latter viruses. Results showed prevalence rates of serological reaction to parainfluenza-3, reovirus type 3 and respiratory syncytial viruses of 28, 72 and 35% in sheep and 26, 64 and 36% in goats, respectively. No antibodies in infectious bovine rhinotracheitis virus were detected in sheep or goats tested. Prevalence rates varied according to the geographical area. No relationships were detected between age, sex, breed, size of flock and prevalence rates of different antibodies except that parainfluenza-3 antibodies were more common in large goat flocks and in sheep flocks with total confinement housing. A relationship between presence of clinical signs in the flocks and prevalence rates of antibodies was only demonstrated for parainfluenza-3 infection in goat flocks.  相似文献

The objectives of this study were to describe the serological prevalence of maedi-visna in a sample of Ontario sheep flocks, and to identify management and demographic variables that were associated with seroprevalence for maedi-visna. A sample of 103 sheep flocks in Ontario was randomly selected from those flocks participating in the Red Meat Plan. The owners of these flocks were surveyed regarding management procedures on their farms, and blood samples were taken from a random sample of ewes in each flock. At least one ewe tested serologically positive, based on the agar gel immuno-diffusion test, in 69.9% of the farms. Positive serological reactions occurred in 20.9% of the 3880 sheep tested. Flock demographics and farm management variables were considered in a multiple regression model, and several factors were positively associated with higher maedi-visna seroprevalence rates. These included the average age of the flock, the number of years the owner had been sheep farming, the practice of using foster ewes, the practice of allowing lambs to have contact with other ewes that are lambing, and the average pasture acreage per ewe.  相似文献

The aim of this study was to determine the prevalence of pestivirus antibodies in sheep and goats in India. A total of 2803 serum samples collected between 2004 and 2008 from 1777 sheep in 92 flocks and 1026 goats in 63 flocks belonging to 13 states were tested by competition ELISA for detection of pestivirus antibodies. In sheep, the true prevalence rate was 23.4% (95% confidence interval: 22.9%–27.0%) and in goats it was 16.9% (95% CI: 16.4%–21.3%). The flock level seroprevalence was 66.3% for sheep and 54.0% for goats. Geographical variation in individual and flock prevalence was highly significant. A significant association (p?<?0.05) was found between sheep and goat flocks having cattle contact and the flock level seroprevalence. The seroprevalence was lower in 6 months–1 year age group compared to the 1–2 year and >2 year age groups in both sheep and goats. Cross neutralization studies on 61 seropositive sheep and 34 seropositive goat samples representing all positive flocks, exhibited > four fold higher titre to bovine viral diarrhoea virus type 1 (BVDV-1) in 41 sheep and 23 goat samples and to BVDV-2 in one sheep and goat each. This study for the first time showed serological evidence of wide spread BVDV infections in Indian sheep and goats, with BVDV-1 predominating and BVDV-2 occasionally besides highlighting the potential risk of infection to other species, which needs to be considered whenever BVD control measures are initiated.  相似文献

In the course of three years, 620 cats of all age categories were investigated for the occurrence of antibodies to Toxoplasma gondii. The serological examinations included the Sabin-Feldman reaction (SFR), complement-fixation reaction (CFR) and microprecipitation in agar gel (MPA), according to methods prescribed for use in Czechoslovakia. When the Sabin-Feldman reaction was used, 250 (40.3%) of the examined cats were found to be positive at the titre of 4-128. By the complement-fixation reaction, antibodies were demonstrated in 144 (23.2%) of the cats at the titre of 5-80. Microprecipitation in agar gel was positive in 106 animals, i.e. 17.1%. Antibodies to Toxoplasma gondii were demonstrated by one serological reaction in 150 (24.2%) cats; coincidence of two serological reactions occurred in 104 (16.8%) of the animals, and coincidence of all the three serological reactions was demonstrated in 48 (7.7%) cats. Specific antibodies were detected by minimally one serological reaction (which means one to three positive findings) in 302 (48.7%) of the examined cats. The diagnostic value of all serological reactions is discussed. It is recommended to repeat the examination after two to three weeks.  相似文献

Serum samples from 1,133 dairy cows (187 herds), 3,712 ewes (103 flocks) and 1,317 adult pigs (877 herds), were tested for neutralizing antibodies against the NADL strain of bovine virus diarrhoea virus. The prevalence rate of seropositive animals was 18.5% in cattle, 4.5% in sheep and 2.2% in pigs, such seroreactors being found in 28% of the cattle herds and 18% of the sheep flocks. In all three species the rate showed considerable herd and geographical variation. In cattle the seroreactor rate was similar in herds with normal reproduction and in 62 herds with problems of repeat breeding. Of 31 pig sera containing antibodies against the NADL strain, 27 were also positive in a neutralization test for antibodies against swine fever virus (Baker strain). However, all sera showed a higher titre of antibodies against the bovine strain than against the swine fever virus. It was concluded that the immune response of the pigs had been induced by ruminant pestivirus, and not by swine fever virus.  相似文献

为研究小鼠弓形虫病病状及卵巢功能的变化,将雌性小鼠随机地分为实验组与对照组,每组各30例.实验组雌性小鼠感染弓形虫,观察其临床症状及体温变化.体温升高并出现稽留热.在感染弓形虫后第45、90d的发情间期采血,对照组也在相同的时间采血.采用放射性免疫方法,测定血清中的雌二醇含量,并于两次采血后将两组小鼠剖检.结果4-5d时实验组与对照组小鼠血清雌二醇含量分别为12.29 ±4.39pg/mL与10.56±8.34pg/mL,90d时实验组与对照组小鼠血清雌二醇含量分别为7.76±3.67pg/mL与10.23±4.85pg/mL;实验组90d血清雌二醇水平降低,与实验组45d时及对照组45d时比较,差异均有显著性.弓形虫感染的小鼠主要脏器严重受损,肠淋巴结可见大量的巨噬细胞.脑组织可见慢性炎症改变及弓形虫包囊体,卵巢呈炎性变化.结论:弓形虫感染引起轻度临床症状和严重病理变化,导致小鼠卵巢功能下降.  相似文献

To determine the levels of background scrapie-like pathology in the brains of clinically normal adult sheep, the brains of 1106 sheep from 28 known scrapie-infected flocks and nine apparently uninfected flocks were examined during 1998 and 1999. One per cent of the brains had vacuolar pathology and disease-specific accumulations of prion protein consistent with a diagnosis of scrapie. All the positive animals had at least one allele of the prion protein gene encoding valine at codon 136, and originated from flocks in which cases of clinical scrapie had been confirmed within the last four years. The parasympathetic nucleus of the vagal nerve was the most consistently and severely affected nucleus in the medulla oblongata, suggesting that the infection enters the brain via ascending fibres of the vagus nerve.  相似文献

A serological survey for Coxiella burnetii was undertaken on a randomly selected population of 103 Ontario sheep flocks. Twenty-two flocks had at least one positive ewe; seven flocks had two or more reactors. The positive flocks were geographically clustered northwest of Guelph. Crutch-clipping of the ewe's wool prior to lambing, and total confinement housing at lambing in winter and spring seemed to lower the probability of seroreactivity of the flock (p less than 0.05). The study suggests that sheep are not a major reservoir for Coxiella burnetii in Ontario.  相似文献

In this study 2058 blood samples from sheep of 150 flocks from the province of Tyrol were tested by ELISA and serum neutralisation tests for antibodies to ruminant pestiviruses. In the ELISA, positive results were obtained with 34.9% of individual sheep sera and in 89.3% of the sheep flocks. The prevalence in sheep and sheep flocks varied according to areas. Seroprevalence of pestiviruses was significantly (p < 0.05) higher in small ruminants pastured during summertime on the Alps. Comparative neutralisation studies were carried out on all positive blood samples with BVDV-1, BVDV-2 and BDV. 443 seropositive sheep samples exhibited clearly the highest titre against one of the pestivirus strains tested. 413 revealed the highest titres (2 or more fold) to BVDV-1, 6 to BVDV-2 and 24 to BDV. In some areas a very high rate of pestivirus seroprevalence could be found. This fact could be harmful to the BVDV-Elimination and Controlling Program in cattle in Austria.  相似文献

Blood variables (cell values, contents of haemoglobin, minerals and proteins) were studied retrospectively in 13 lambs which had acquired toxoplasmosis during the summer, and in 2 lambs which had been infected during the perinatal period. Examination by immunoelectrophoresis was also performed. The lambs had been in experimental groups in a research project of blood values in sheep on pastures of different qualities. The groups comprised a total of 40 lambs, which had been bled once monthly. Examinations were also carried out in lambs experimentally infected with the RH strain of T. gondii, in lambs experimentally infected with a sheep strain of the parasite, and in a control group. No significant changes in the blood variables were found.Meat samples from the experimentally infected lambs and their controls were examined parasitologically. T. gondii was recovered from all dye test positive (titre ≥ 1/16) lambs inoculated with the sheep strain, but not from any of the lambs inoculated with the RH strain. The results indicated that the RH strain does not produce cysts in the muscular tissue in sheep.
