A novel method named cell membrane affinity chromatography was used to screen antimicrobial peptides from Jatropha curcas . A cationic antimicrobial peptide (KVFLGLK, JCpep7) was successfully isolated and identified. Antimicrobial assays indicated that JCpep7 was active against the tested microorganisms ( Salmonella typhimurium ATCC 50013, Shigella dysenteriae ATCC 51302, Pseudomonas aeruginosa ATCC 27553, Staphylococcus aureus ATCC 25923, Bacillus subtilis ATCC 23631, and Streptococcus pneumoniae ATCC 49619) with minimal inhibitory concentration (MIC) values ranging from 24 to 64 μg/mL. The antimicrobial mechanisms based on Fourier transform infrared (FTIR) spectroscopy and transmission electron microscopy (TEM) techniques showed that JCpep7 killed microbes principally via breaking of their cell walls and membranes, followed by cell lysis. The results indicated that cell membrane affinity chromatography could be a promising approach for high-throughput screening of antimicrobial peptides from J. curcas .  相似文献

A rapid bacterial identification method by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) using ribosomal proteins coded in S10 and spc operons as biomarkers, named the S10-GERMS (the S10-spc-alpha operon gene encoded ribosomal protein mass spectrum) method, was applied for the genus Bacillus a Gram-positive bacterium. The S10-GERMS method could successfully distinguish the difference between B. subtilis subsp. subtilis NBRC 13719(T) and B. subtilis subsp. spizizenii NBRC 101239(T) because of the mass difference of 2 ribosomal subunit proteins, despite the difference of only 2 bases in the 16S rRNA gene between them. The 8 selected reliable and reproducible ribosomal subunit proteins without disturbance of S/N level on MALDI-TOF MS analysis, S10, S14, S19, L18, L22, L24, L29, and L30, coded in S10 and spc operons were significantly useful biomarkers for rapid bacterial classification at species and strain levels by the S10-GERMS method of genus Bacillus strains without purification of ribosomal proteins.  相似文献

本文对33株枯草芽孢杆菌群菌株进行β-甘露聚糖酶活性筛选,其中的32株具有β-甘露聚糖酶活性,只有1株无β-甘露聚糖酶活性。通过基因克隆测序的方法获得33株枯草芽孢杆菌群菌株β-甘露聚糖酶基因编码区全序列,对酶基因进行同源性分析并构建系统发育树;在β-甘露聚糖酶基因系统发育树中,33株枯草芽孢杆菌群菌株聚为3个分支,分别是枯草芽孢杆菌分支、地衣芽孢杆菌分支和解淀粉芽孢杆菌分支;枯草芽孢杆菌、地衣芽孢杆菌和解淀粉芽孢杆菌β-甘露聚糖酶基因种内同源性大于91%,而种间同源性为60%~69%。  相似文献

The antibacterial activity of a series of alkyl gallates (3,4,5-trihydroxybenzoates) against Gram-positive bacteria was tested using a broth dilution method. All of the Gram-positive bacteria tested were susceptible to alkyl gallates, and this activity was found to correlate with the alkyl chain length. The antibacterial activity of alkyl gallates against Bacillus subtilis was a parabolic function of their lipophilicity and maximized with alkyl chain length between C(8) and C(11). Notably, alkyl gallates were found to be bactericidal against B. subtilis ATCC 9372, but this activity was significantly affected by the endospore formation in the culture. The antibacterial activity of alkyl gallates likely comes at least in part from their ability to inhibit the membrane respiratory chain but is not due to the prooxidant action.  相似文献

从西瓜根围分离得到的一株有效抑制西瓜枯萎病菌的枯草芽孢杆菌B 11菌株(B acillus subtilisstra in B 11)。为获得高产拮抗物质的诱变菌株,采用紫外线诱变方法对B 11菌株进行诱变,结果获得抑菌活性比野生菌B 11菌株强的6株诱变菌株,其中G 17菌株的抑菌活性及粗拮抗物质含量均高于B 11菌株,且遗传性较稳定;再以G 17菌株为供试菌株,用亚硝基胍(NTG)对其进行化学诱变,结果获得抑菌活性较强的4株诱变菌株,其中BV 22菌株对西瓜枯萎病菌的抑菌活性最高,其抑菌活性比G 17和B 11菌株分别提高了36.88%和80.00%;其粗拮抗物质的含量分别提高了30.15%和62.53%。研究结果说明,用物理诱变B 11菌株后获得的诱变菌株再用化学方法进行诱变,可选育出高产拮抗物质的诱变菌株。  相似文献

枯草芽孢杆菌对盐碱土水分运动和水稳性团聚体的影响   总被引:5,自引:0,他引:5  
为探明微生物在盐碱地改良方面的应用,通过在盐碱土中加入枯草芽孢杆菌,研究不同浓度微咸水(0,1,2,3,4g/L)条件下,盐碱土中的水分运动和土壤水稳性团聚体的变化规律。结果表明:(1)枯草芽孢杆菌可以显著减少土壤的入渗能力,同时可减缓水分的迁移能力,添加枯草芽孢杆菌的土壤与相对应的对照组相比,累积入渗量和湿润锋推进距离均显著减少,其中在微咸水浓度为2g/L时,土壤累积入渗量减少幅度最大,为40.97%,其他处理减少程度依次为3,0,1,4g/L,湿润锋的推进距离也有类似的结果。(2)利用Philip和Kostiakov入渗模型拟合试验数据,发现Philip公式中的吸渗率S均小于相相应对照组,Kostiakov公式中的经验系数K均小于相应对照组,而经验指数β均大于相应对照组。(3)枯草芽孢杆菌可以增加土壤的保水能力,在微咸水浓度为2g/L时,对土壤水吸力的影响较大。与对照组相比,van Genuchten公式中土壤滞留含水率θr、饱和含水率θs及与进气值相关系数α均有所增大,形状系数n逐渐减小。(4)含有枯草芽孢杆菌的土壤,在微咸水入渗后其土壤团粒结构均有较大的改善。  相似文献

枯草芽孢杆菌改良盐碱土过程中水盐运移特征   总被引:3,自引:2,他引:1  
该文通过室内一维土柱试验,设置5种不同枯草芽孢杆菌质量分数(0、1、3、5、7 g/kg)处理,研究不同含量枯草芽孢杆菌对盐碱土水盐运移的影响。结果表明:施加枯草芽孢杆菌后,土壤累积入渗量、入渗速率及湿润锋运移距离均显著降低,在入渗时间为3 600 min时,枯草芽孢杆菌质量分数为1、3、5、7 g/kg的累积入渗量相比0 g/kg分别减少了18.49%、21.85%、12.18%、3.78%;当湿润锋运移至27 cm时,枯草芽孢杆菌含量为1、3、5、7 g/kg所用时间相比0 g/kg分别增大了96.21%、108%、37.84%、16.76%,在枯草芽孢杆菌施加量为3 g/kg时,单位入渗历时内累积入渗量、入渗速率及湿润锋的运移距离最小。枯草芽孢杆菌同样影响Philip方程和Green-Ampt入渗公式参数显著,土壤水饱和导水率KS、吸渗率S、稳定入渗率A随着枯草芽孢杆菌含量的增加先减少后增大,湿润锋处的吸力hf先增大后减少,当施加量为3 g/kg时,S、KS、A均取得最小值,hf取得最大值。枯草芽孢杆菌可提高土壤的保水性能,在土层深度为27 cm处,施加量为1、3、5和7 g/kg相比0 g/kg的剖面含水量分别增加了17.65%、31.76%、11.76%、7.06%。施加枯草芽孢杆菌的土壤在入渗结束后,土壤的含盐量分别降低了22.37%、31.29%、17.78%、10.67%。施加枯草芽孢杆菌后,1、3、5、7 g/kg的水稳性团聚体含量相比0 g/kg分别增加了13.02%、17.59%、9.68%和5.24%。综上,在盐碱土中施加3 g/kg的枯草芽孢杆菌,对盐碱土壤的治理具有积极作用。  相似文献

Fourier transform infrared spectroscopy (FT-IR, 4000-600 cm(-)(1)) was used to discriminate between intact and sonication-injured Listeria monocytogenes ATCC 19114 and to distinguish this strain from other selected Listeria strains (L. innocua ATCC 51742, L. innocua ATCC 33090, and L. monocytogenes ATCC 7644). FT-IR vibrational overtone and combination bands from mid-IR active components of intact and injured bacterial cells produced distinctive "fingerprints" at wavenumbers between 1500 and 800 cm(-)(1). Spectral data were analyzed by principal component analysis. Clear segregations of different intact and injured strains of Listeria were observed, suggesting that FT-IR can detect biochemical differences between intact and injured bacterial cells. This technique may provide a tool for the rapid assessment of cell viability and thereby the control of foodborne pathogens.  相似文献

从羽毛废弃物中筛选出一株高效降解羽毛、产角蛋白酶的革兰氏阳性芽孢杆菌YYW-1,根据形态学特征、生理生化特征、生长特征和16S rDNA序列特征(GenBank接受号:DQ017585)将其鉴定为枯草芽孢杆菌(Bacillus sublitis),命名为Bacillus sublitis YYW-1。初步分析了YYW-1产角蛋白酶性质,该酶最适反应pH值为7.2,最适反应温度为60℃,该酶活性能够被EDTA和PMSF彻底抑制,说明该酶既是金属蛋白酶又是丝氨酸蛋白酶。  相似文献

The present study was designated to evaluate the antimicrobial and antioxidant activities of the essential oil, obtained by using a Clevenger distillation apparatus, water soluble (polar) and water insoluble (nonpolar) subfractions of the methanol extracts from aerial parts of Satureja hortensis L. plants, and methanol extract from calli established from the seeds using Gamborg's B5 basal media supplemented with indole-3-butyric acid (1.0 ppm), 6-benzylaminopurine (N(6)-benzyladenine) (1.0 ppm), and sucrose (2.5%). The antimicrobial test results showed that the essential oil of S. hortensis had great potential antimicrobial activities against all 23 bacteria and 15 fungi and yeast species tested. In contrast, the methanol extract from callus cultures and water soluble subfraction of the methanol extract did not show antimicrobial activities, but the nonpolar subfraction had antibacterial activity against only five out of 23 bacterial species, which were Bacillus subtilis, Enterococcus fecalis, Pseudomonas aeruginosa, Salmonella enteritidis, and Streptococcus pyogenes. Antioxidant studies suggested that the polar subfractions of the methanol extract of intact plant and methanol extract of callus cultures were able to reduce the stable free radical 2,2-diphenyl-1-picrylhydrazyl to the yellow-colored diphenylpicrylhydrazine. In this assay, the strongest effect was observed for the tissue culture extract, with an IC(50) value of 23.76 +/- 0.80 microgram/mL, which could be compared with the synthetic antioxidant agent butylated hydroxytoluene. On the other hand, linoleic acid oxidation was 95% inhibited in the presence of the essential oil while the inhibition was 90% with the chloroform subfraction of the intact plant. The chemical composition of a hydrodistilled essential oil of S. hortensis was analyzed by gas chromatography (GC)/flame ionization detection (FID) and a GC-mass spectrometry system. A total 22 constituents representing 99.9% of the essential oil were identified by GC-FID analaysis. Thymol (29.0%), carvacrol (26.5%), gamma-terpinene (22.6%), and p-cymene (9.3%) were the main components.  相似文献

为了获得四环素类抗生素的高效降解菌,从长期受四环素类抗生素污染的土壤中分离、重复驯化筛选得到6株对四环素类抗生素具有降解能力的菌株;采用高效液相色谱法研究了6株降解菌的降解效果,并通过形态、生理生化特征及16S rDNA序列分析对其进行鉴定。结果表明,菌株TJ-2#对土霉素、四环素和金霉素3种四环素类抗生素的降解效果最好,降解率分别为58.3%、63.9%和65.5%,TJ-6#其次;6株降解菌均能以四环素类抗生素作为唯一碳源生长。经形态、生理生化特征及16S rDNA鉴定,菌株TJ-2#为木糖氧化无色杆菌(Achromobacter sp.),TJ-6#为枯草芽胞杆菌(Bacillus sp.)。本研究结果对四环素类抗生素的生物降解具有一定的现实意义。  相似文献

【目的】 从岷山红三叶根际分离的109株促生菌,鉴定、筛选了具有生防能力的菌种资源并研究其生防特性,为生产应用提供服务。 【方法】 采用平板对峙法筛选优良生防菌株,对其产铁载体能力进行定性、定量测定和16S rRNA基因序列分析鉴定。 【结果】 平板对峙试验显示,109株促生菌中有8株促生菌对3种病原真菌 (立枯丝核菌、黄瓜枯萎菌、西瓜尖镰孢菌) 具有拮抗效果,分别是菌株MHS3、MHS7、MHS9、MHS27、MHS31、MHS38、MHS39和MHS48。其中,菌株MHS27、MHS31发酵液对立枯丝核菌抑菌率分别为40.1%、47.1%;菌株MHS7和MHS48发酵液对黄瓜枯萎菌抑菌率分别为26.1%、22.8%;菌株MHS27和MHS31发酵液对西瓜尖镰孢菌抑制率分别为34.0%、30.5%;产嗜铁素是生防菌 (MHS7、MHS27、MHS31、MHS48) 拮抗3种病原真菌的主要途径之一。经16S rRNA基因序列分析初步鉴定,菌株MHS7为特基拉芽孢杆菌 (Bacillus tequilensis),菌株MHS27为溶蛋白芽孢杆菌 (Bacillus proteolyticus),菌株MHS31为Pseudomonas paralactis,菌株MHS48为克雷伯氏菌 (Klebsiella oxytoca)。 【结论】 从岷山红三叶根际分离筛选出4株具有优良生防作用的PGPR菌株,为今后生物菌肥应用推广提供了菌种资源支持。   相似文献

为了合理开发利用蕨类植物资源,本研究利用圆纸片法分析了云南省4种蕨类植物单芽狗脊蕨(Woodwardia unigemmata)、蜈蚣蕨(Pteris vittata)、鸡足山耳蕨(Polystichum jizhushanense)和灰绿耳蕨(Polystic-hum eximium)的提取液对金黄色葡萄球菌(Staphylococus aureus)、大肠杆菌(Escherichia coli)和枯草芽孢杆菌(Bacillus subtilis)的抑菌活性。抑菌试验结果表明,当4种蕨类植物提取液浓度分别为10%、8%、6%和4%时,对照组无抑菌圈出现,单芽狗脊蕨提取液对大肠杆菌和金黄色葡萄球菌无抑菌活性,灰绿耳蕨提取液对金黄色葡萄球菌无抑菌活性,其它的蕨类植物提取液对供试微生物都表现出不同程度的抑菌活性;并且这4种蕨类植物提取液至少对一种供试细菌具有抑菌活性,抑菌活性范围为8~14mm,平均抑菌圈直径为11.2mm,4种提取液对枯草芽孢杆菌抗菌活性效果最明显。由此,我们推测本研究中的4种蕨类植物均有抗菌物质存在,并将有望成为筛选抗菌新型药物的种质资源。  相似文献

Natural dihydrocoumarin, which is of great interest in the flavor industry, was biotechnologically produced from pure coumarin or tonka bean meal with Pseudomonas orientalis, Bacillus cereus, and various Saccharomyces cerevisiae strains. Coumarin was shown to be converted to melilotic acid, which yielded dihydrocoumarin upon distillation during purification. About 1.0 g/L product was obtained from 25 g/L tonka beans with S. cerevisiae within 147 h. This dihydrocoumarin thus fulfills all of the criteria of a natural raw material and can be used as a natural flavoring in accordance with U.S. and European Union regulations.  相似文献

白酒酿造行业产生的酒糟含有大量纤维素,不仅原料利用率低,而且丢弃的酒糟会对环境产生污染。为了获得酒糟纤维素降解能力强的微生物并进行应用,从酱香型酒醅、清香型酒醅、浓香型大曲、竹林里的土壤腐殖质中分离筛选酒糟纤维素降解菌,并对菌种分类、理化特征和降解酒糟的特性进行研究。pH值、温度和酒精胁迫性试验进一步确定了最佳酒糟纤维素降解菌为B2菌株,该菌株在pH值3.0、温度44 ℃、酒精含量为体积分数4%环境中生长良好。基于形态学、生理生化和分子生物学分析,鉴定B2菌株为枯草芽孢杆菌(Bacillus subtilis)。在单因素试验结果的基础上,采用Box-Benhnken响应面法进行优化,确定了B2菌株降解酒糟的最佳工艺条件为酒糟添加量71 g/L、温度37 ℃、pH值6.4、接种量8%,此时酒糟降解率为15.23%。该研究丰富了酒糟纤维素降解菌的微生物资源库,同时为酒糟的资源化利用提供技术参考。  相似文献

豆浆富含植物蛋白,但由于植物蛋白相较动物蛋白不易被人体吸收。利用产蛋白酶菌株发酵豆浆,可将植物蛋白降解成多肽和氨基酸,提高其消化吸收率。传统农家酱是以大豆为主要原料的传统发酵食品,含有大量能够水解大豆蛋白的优良菌株。该研究从传统农家酱中分离到8株产蛋白酶的芽孢杆菌并用于豆浆处理,处理后豆浆的可溶性肽含量显著提高,其中菌株BJ-20处理效果最显著;氨基态氮含量和水解度也有显著变化,其中BJ-6处理后的豆浆最佳,氨基态氮含量提高了10.87%,水解度为23.49%。同时,处理后豆浆的组织状态得到了明显改善。本研究丰富了豆类发酵菌种库,提高了大豆蛋白利用率,丰富了大豆产品种类,为大豆保健产品开发提供参考及理论支持。  相似文献

从山东和江苏采集的菜园土样中分离筛选得到4株芽孢杆菌B15、B110、B113和B122,它们均对植物病原真菌具有较强的拮抗能力且具有广谱性。4株芽孢杆菌可以致使黄瓜枯萎病原菌菌丝生长形态异常,出现菌丝前端膨大或变细、菌丝扭曲、分支加剧、生成大量圆珠形囊状体和原生质凝聚等异常现象。通过对4个菌株培养物形态特征的观察和一系列的生理生化实验得到其初步鉴定结果,其中B15和B113为短短芽孢杆菌(Brevibacillus brevis),B110为多粘类芽孢杆菌(Paenibacillus polymyxa),B122为枯草芽孢杆菌(Bacillus subtilis)。  相似文献

The plasmid pHG contains a cyclodextrin glycosyltransferase (CGTase) gene (cgt) derived from Bacillus macerans. Two transformants, Bacillus subtilis (pHG) and Escherichia coli (pHG), were found to produce CGTases with the same primary structure as the enzyme from B. macerans. However, the beta-cyclodextrin coupling activity of the CGTase from E. coli (pHG) was 14-fold higher than that of the enzymes from the other strains. By contrast, no differences in alpha-cyclodextrin coupling activities were observed among these CGTases. CGTase from E. coli (pHG) was found to be less thermostable than the other CGTases. When the CGTase produced by B. subtilis was treated with increasing urea concentrations (10-1000 mM) to promote increasing degrees of protein unfolding, a bell-shaped beta-cyclodextrin coupling activity profile was obtained. Subtle differences in the conformation of the CGTase produced by E. coli are therefore proposed to be responsible for the markedly increased beta-cyclodextrin coupling activity of this enzyme.  相似文献

为探究商陆不同极性、部位提取物的抑菌性能,本实验分别从商陆根和茎中提取石油醚相、乙酸乙酯相、正丁醇相和水相4种不同极性的提取物,再采用滤纸片法测量提取物对大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphyloccocus aureus)、巨大芽孢杆菌(Bacillus megaterium)和副溶血弧菌(Vibrio parahaemolyt-icus)4种细菌的抑制性能。结果表明,部分商陆的提取物有一定的抑菌活性,如根正丁醇相对巨大芽孢杆菌和副溶血弧菌,茎水相对副溶血弧菌的抑菌圈在10mm以上,但总体上商陆的提取物的抑菌性能远不及头孢氨苄好。实验还发现商陆抑菌活性最强的物质大都存在于根中,且抑菌活性最强的物质大都存在于水和正丁醇这种极性较高的溶剂的提取物中,不同提取物对不同细菌的抑菌活性情况不一致。因此本研究结果可以为商陆提取物抑菌性能的进一步探索提供参考。  相似文献
