Between May 2002 and February 2003 a longitudinal survey was carried out in Mbale and Sironko Districts of Eastern Uganda to determine the influence of agro-ecological zones (AEZ) and grazing systems on tick infestation patterns and incidence of East Coast Fever (ECF) in bovine calves. The study area was stratified into AEZ (lowland, midland and upland) and grazing systems {zero grazing (ZG), restricted-outdoor grazing (ROG) and communal grazing (CG)}, whose strata had previously been shown to influence the prevalence of ECF, babesiosis and anaplasmosis. One hundred and eighty-five smallholder dairy farms with a total of 198 calves of both sexes, between the ages of 1 day and 6 weeks, were purposively selected from the AEZ–grazing system strata. Nine dynamic cohorts (11–51 calves in each) of these calves were examined and sampled monthly. Ticks infesting the calves were counted from one side of the animal body and categorized into the different species, sex and feeding status. Sera were collected at recruitment and monthly thereafter and antibodies against Theileria parva, T. mutans, Babesia bigemina, B. bovis and Anaplasma marginale were measured using ELISA. Tick challenge (total and specific) varied with AEZ and grazing system. The risk of infection with T. parva was higher in the lowland zone compared to the upland zone (hazard ratio (HR) = 2.59; 95% CI: 1.00–6.34). The risk of infection with T. parva was higher in the CG system than the ZG system (HR = 10.00; 95% CI: 3.61–27.92). The incidence risk for sero-conversion, over the 10 months study period, was 62, 16 and 9% in the lowland, midland and upland zones, respectively. Ninety-eight percent of the calves in lowland-CG stratum sero-converted by the age of 6 months, while 56 and 8% did so in the lowland-ROG and the lowland-ZG stratum, respectively. The results of this study show the need to consider farm circumstances and the variation in ECF risk, both spatially and temporally when designing control strategies for ECF.  相似文献

Theileria parva is the causative agent of a lethal tick-borne disease of cattle occurring in eastern, central and southern Africa. Variations in the sensitivity of the serological and molecular tests with seasonal vector occurrence and discrepancies between low PCR prevalence and high T. parva vector density are a setback to estimate true prevalences. Therefore, the objectives of the present studies were to evaluate (1) the sensitivity of three serological tests (IFAT, ELISA and SELISA) and one molecular test (PCR) in the diagnosis of chronic T. parva infections in four different agro-ecological zones of Rwanda and (2) the effect of tick challenge and animal's age on the sensitivity of PCR. Blood samples from 635 bovines were collected in four agro-ecological zones of Rwanda. All sera were screened using the IFAT, ELISA, SELISA and PCR. The binary results of the four diagnostic tests were introduced separately for each agro-ecological zone in a Bayesian model to estimate the prevalence of T. parva infections and the sensitivity of the four diagnostic tests. All test specificities were set to 100%. The estimated T. parva prevalence was much higher (83–85%) than estimations based on single diagnostic tests. The estimated sensitivity of serological tests was relatively constant and ranged from 57 to 75% in the various areas. The sensitivity of PCR showed more pronounced variations, ranging from 66% in the low T. parva transmission (high land) zones compared to 24% in the highly vector infested (low land) zones. Calves and adult cattle (n = 194) were also sampled in regularly and irregularly dipped herds in the low land region. The apparent T. parva prevalence detected by PCR was significantly higher in calves than in adult cattle and in herds regularly treated with acaricides, while no significant differences were found with IFAT. The conditional probability that a sample was positive at PCR while it was positive at IFAT was significantly lower in adults. The implication of these findings in the use of diagnostic assays for epidemiological studies is discussed.  相似文献

Thirty-one calves born into five Maasai zebu cattle herds over a period of 1 month in the Trans-Mara Division of Kenya, endemic for theileriosis, were recruited for an intensive study of theileriosis. No calves up to 6 months of age died but all developed Theileria infections as judged by slide examination and serology. Parasitosis by T. mutans schizonts in lymph node smears was usually higher than that of T. parva. The T. mutans schizonts usually occurred at an earlier age but persisted at a patent level for a shorter time than those of T. parva. Serological findings using the indirect fluorescent antibody test confirmed the parasitological findings. It was evident that colostral transfer of Theileria antibodies was frequent. Theileria piroplasm parasitaemia had developed in all calves by 111 days of age. The earlier parasitosis by T. mutans reflected the higher infection rates in Amblyomma spp. than in Rhipicephalus appendiculatus. The mean number of R. appendiculatus on the ears of calves during the observations was 9.1 adults and 1.5 nymphs. Clinical episodes of T. mutans and T. parva infection were associated with febrile responses, enlarged lymph nodes, anaemia and other symptoms and about 80% of calves had poor weight gains or weight losses during either clinical infection. It would appear that theileriosis is one of the most important factors in the stunting of calf development in the area.  相似文献

Bovine theileriosis was a kind of hemo-protozoan diseases that seriously hindered the sustainable development of cattle industry.In the present study, the infection of Theileria was detected using microscopic examination and species-specifc PCR of T.annulata, T.sergenti and T.sinensis from 18 blood samples collected from a breeding cattle farm in China.Then ITS and MPSP genes were amplified and cloned using universal primers of ITS and allele-specific primers of 4 major piroplasm surface protein (MPSP) from the positive samples.The sequences were used to make alignment, polymorphism and phylogeny analysis.The results showed that 3 samples were positive for Theileria and the 3 positive samples were all the T.sergenti infection.The amplification of allelic MPSP gene of T.sergenti from the 3 positive samples showed that two of them were single infection by Ikeda-type and another one was co-infection with both Chitose-type and Ikeda-type, while Buffeli-type and Thai-type were not detected.Moreover, on the basis of phylogenetic tree constructed with MPSP gene sequences, types 1 and 2 of MPSP were confirmed to be present in the cattle farm.The results revealed that there were T.sergenti infection in the breeding cattle farm, and these parasites at least with 2 allelic MPSP gene types were present, which indicated that the immune prevention and control of the disease became more complicated.Our research laid foundation of the further study on T.sergenti infection and disease prevention and control.  相似文献

牛泰勒虫病是严重危害养牛业可持续发展的血液原虫病,本试验采用血涂片镜检和特异性PCR检测技术,对中国某种牛场的18份血样进行了泰勒虫检测,然后分别用ITS基因通用引物和4种MPSP等位基因特异性引物自阳性样品中扩增出对应的基因,克隆测序后,进行序列比对和进化分析,确定泰勒虫基因型。结果显示,自18份牛血样中检出3份阳性样品,且全为瑟氏泰勒虫感染;3个阳性样品的瑟氏泰勒虫MPSP等位基因扩增结果显示,1个阳性样品为I (Ikeda) 和C (Chitose)型的混合感染,另2个样品为I型单一感染,而均无B (Buffeli)和Thai型;用扩增的MPSP基因测序,构建进化树,确认其感染的瑟氏泰勒虫存在MPSP 1型和2型。这些结果表明,该种牛场存在瑟氏泰勒虫感染,且同时存在2种MPSP等位基因型;MPSP等位基因的复杂性可能使该病的免疫防控更加困难。本试验结果为深入研究瑟氏泰勒虫感染情况及免疫防控提供了数据支持,并为养防一体做好监控。  相似文献

The responses were monitored of young crossbred calves vaccinated against tropical theileriosis during the winter against a field tick challenge in the disease season. Thirty-eight calves below 2 months of age, born after the end of the disease season, were selected at an organized farm. Twenty-five animals were vaccinated with Theileria annulata (Hisar) cell culture vaccine (developed at CCS HAU Hisar laboratory) after the end of the disease season and 13 calves were kept as non-vaccinated controls. These calves were observed for their susceptibility to theileriosis in the new disease season. There was an increase in antibody titre in 18 of the 25 vaccinated animals one month after vaccination. The antibody titre then declined gradually, but remained higher than those of the non-vaccinated animals at month 0. No fever or other clinical signs of tropical theileriosis were observed in any of the vaccinated animals. Nine out of 25 (36%) vaccinated calves showed occasional piroplasms (<;0.5%) in blood smears. All the vaccinated animals withstood the field tick challenge. On the other hand, 9 of the 13 (69%) unvaccinated calves exhibited occasional piroplasms, and included three clinical cases of tropical theileriosis. These observations suggest that young crossbred calves vaccinated with the T. annulata (Hisar) cell culture vaccine at the end of the disease season were relatively resistant during the next disease season.  相似文献

An enzyme-linked immunosorbent assay (ELISA) using sonicated purified Theileria annulata piroplasms was compared with an indirect immunofluorescent antibody test (IFAT) during a vaccination trial in cattle to test different doses and passage numbers of an attenuated T. annulata-infected lymphoblastoid cell-line, and also with Giemsa-stained blood smears during an epidemiological field study of tropical theileriosis in Morocco. The sensitivities of both the ELISA (0.56) and the IFAT using T. annulata piroplasm antigen (0.56) were lower than the IFAT using schizont antigen (0.94) for detecting serum antibodies from 18 cattle immunised 38 days previously with cell-line. The ELISA was, however, the most sensitive test after 180 days (0.50 compared with 0.06 for the piroplasm IFAT and 0.39 for the schizont IFAT), and each test detected antibodies in all sera after challenge with live T. annulata sporozoites. There were minor differences in the ability of blood smear examinations and the ELISA to detect infected and uninfected cattle in the field study at the start and end of the disease season. Initially, the sensitivity and specificity of blood smears were both 0.96 and for the ELISA were 0.83 and 0.86, whereas at the end of the season sensitivity and specificity of blood smears were 0.96 and 0.86 and for the ELISA were 0.95 and 0.94. The specificity of the ELISA was affected by the presence of calves with colostral antibodies, and if these were disregarded the specificities before and after the season were 0.94 and 1.00.  相似文献

This work extends basic knowledge of tropical theileriosis in taurine and crossbred cattle. Infection of Bos taurus and Bos taurus cross Bos indicus (Sahiwal) calves with graded doses of sporozoites of Theileria annulata (Hissar), an Indian stock of the parasite, showed the following to be dose dependent in both cattle types: the time to appearance and population size of macroschizonts, microschizonts and piroplasms, time and severity of pyrexia, anaemia manifested by erythrocyte counts and haematocrit. All infections were accompanied by a prompt and severe panleucopenia. This effect was dose related in both the taurine and the Sahiwal crossbred calves. Lymphocyte counts returned to preinfection levels in the blood of animals which recovered, but death from theileriosis was characteristically accompanied by a persistent and severe lymphocytopenia. Flow cytometry using monoclonal antibodies to bovine mononuclear cells was used to identify the lymphocyte subsets involved in lymphocytopenia. The outcome of infection was dose dependent in the crossbred calves but not in taurine calves. Although the results obtained did not differ qualitatively between the two cattle types, they provided some preliminary evidence for resistance to tropical theileriosis in Sahiwal crossbred calves.  相似文献

Blood from sick cattle in Bahrain transmitted piroplasms of Theileria annulata to a splenectomized calf. Larvae of Hyalomma anatolicum anatolicum were infected on the calf and, after moulting, induced clinical theileriosis, associated with numerous schizonts, in the same calf. The animal was cured by specific treatment. Antigenic differences thus shown between piroplasms on the one hand, and sporozoites and schizonts on the other hand, were confirmed in the indirect fluorescent antibody test, as a significant titre to T. annulata piroplasm antigen developed after the inoculation of blood, but to schizont antigen only after the infective ticks had induced the appearance of schizonts.  相似文献

The Theileria parva carrier-state in cattle on commercial farms on Zimbabwe was investigated using parasitological and serological methods. The proportion of cattle showing Theileria piroplasms on two farms, which had recent histories of disease outbreaks, were 64% (n = 106, total of heifers and weaned calves examined) and 71.5% (n = 60) while the proportion of T. parva antibodies for the same animals were 59% and 98.5%, respectively. On four farms where no cases of the disease occurred for over 10 years, the average proportion of animals showing piroplasms and antibodies were 55.4% (range 32-82, n = 223) and 73% (range 47-91, n = 223), respectively. However, on another three farms which had no history of theileriosis outbreaks these proportions were very low, being 11.4% (0-24, n = 157) for piroplasms and 12.2% (5-23, n = 157) for antibodies. The mean infection rate in unfed Rhipicephalus appendiculatus adults collected from farms with a high prevalence of cattle which were carriers of Theileria piroplasms during the tick activity season was 29% (range 12-60%) with 9.3 (range 2-18.7) mean infected acini per infected tick. The infectivity of different tick batches to susceptible cattle produced a wide spectrum of theileriosis reactions. Laboratory controlled experiments were carried out to study the persistence of T. parva (Boleni) piroplasms in cattle immunized with this strain as well as its infectivity for ticks and its subsequent transmissibility to cattle. Examination of the salivary glands of 15 batches of ticks collected from six immunized cattle on three different occasions over 18 months showed that none were infected with Theileria parasites. However, the infectivity of other ticks in the same batches to susceptible animals was demonstrated 6, 10 and 18 months after cattle had been immunized with Boleni stabilate.  相似文献

The prevalence of Theileria spp. infection was studied in sheep in the South Khorasan province in Iran from 2003 to 2004. A total of 840 sheep from 34 flocks were clinically examined and investigated for the presence of Theileria spp. in the appropriate blood smears and any tick species on the body of the animals. In this study, 11.9% of sheep were infected with Theileria spp., with a parasitemia of 0.02–0.1%. Differences in the infection rates were statistically significant among different areas of the South Khorasan province. The highest prevalence was found in the Ferdows area (31.4%) and the lowest rate in the Nehbandan area (0.7%). The prevalence of Theileria spp. infection in males and females and between different age groups of sheep were not statistically significant. Seasonally, the prevalence of Theileria spp. infection in sheep reached its highest level in June (26.3%), whereas it decreased in July and August. It was found that 50.5% of the animals harbored Rhipicephalus sanguineus, 48.5% harboured Hyalomma anatolicum and 0.89% harboured Hyalomma dromedari.  相似文献

The 18S rRNA genes of Theileria species detected in sika deer, Cervus nippon centralis in Yamaguchi and Cervus nippon yesoensis in Hokkaido, were analyzed. The percent identities of the nucleotide sequences of Theileria from Cervus nippon centralis and Cervus nippon yesoensis were more than 99%. The percent identities of the Theileria sp. from sika deer and Theileria sergenti, Theileria buffeli and Theileria cervi were 97, 96 and 95%, respectively. Phylogenetic analysis of the gene sequences also revealed that Theileria sp. detected from sika deer comprise a clade that is clearly distinct from the clade comprised of Theileria from cattle.  相似文献

We investigated the prevalence of Cryptosporidium infection in relation to age and clinical status in cattle in the central region of Viet Nam. A total of 266 fecal samples from diarrheic and non-diarrheic cattle were examined by the modified Ziehl-Neelsen staining method. Prevalence of Cryptosporidium parvum type infections, those of the Cryptosporidium andersoni type, and mixed infection of both types was 33.5% (89/266), 5.6% (15/266), and 3.4% (9/266), respectively. The infection rate of 44.3% (35/79) of C. parvum in calves less than 6 months old was significantly higher than that of 28.9% (54/187) in cattle greater than 6 months old (P < 0.01). Although no C. andersoni oocysts were detected in calves less than 3 months old, no significant difference was observed between the age groups in the prevalence of C. andersoni infection and mixed infection. The percentage of diarrheic and non-diarrheic cattle identified to be shedding C. parvum oocysts was 46.5% (74/159) and 14.0% (15/107), respectively (P < 0.0001). The risk of diarrhea was 1.7 times greater in C. parvum-infected calves than in their non-infected counterparts. DNA sequences of 18S rRNA genes of C. parvum type and C. andersoni type indicated that they were C. parvum bovine genotype and C. andersoni, respectively. This is the first genetic identification of C. parvum bovine genotype and C. andersoni from cattle in Viet Nam.  相似文献

为了解吕氏泰勒虫在中国不同地区山羊和绵羊中的流行情况,本研究应用基于吕氏泰勒虫18S rRNA基因位点的PCR检测方法,对采自中国河南、甘肃、陕西、山西、贵州、云南、新疆7个地区的281份羊血液样品进行检测,并对阳性样品测序以进行序列分析。结果显示,羊吕氏泰勒虫总感染率为35.23%(99/281)。不同采样点羊吕氏泰勒虫感染率差异极显著(P<0.01),其中河南省羊吕氏泰勒虫感染率最高(98%,49/50),新疆最低(0)。绵羊和山羊吕氏泰勒虫感染率分别为51.67%(31/60)和30.77%(68/221),差异极显著(P<0.01);放牧羊吕氏泰勒虫感染率(41.55%,86/207)极显著高于舍饲羊(17.57%,13/74)(P<0.01);羊吕氏泰勒虫感染率在春、夏、秋及冬四季分别为56.00%(28/50)、42.75%(59/138)、9.43%(5/53)和17.50%(7/40),差异极显著(P<0.01);≥ 12月龄和<12月龄羊吕氏泰勒虫感染率分别为33.50%(67/200)和39.51%(32/81),差异不显著(P>0.05)。此外,遗传进化分析表明,本研究获得的羊吕氏泰勒虫分离株与中国流行的吕氏泰勒虫分离株同源性高达99.60%以上,且位于同一分支上。本研究为进一步了解中国不同地区羊吕氏泰勒虫的流行现状及分布提供了重要参考依据。  相似文献

Intestinal establishment and reproduction of adult Trichinella spiralis, Trichinella nativa, Trichinella britovi and Trichinella pseudospiralis were examined as single species or mixed species infections in foxes. This is the first study of intestinal dynamics of Trichinella spp. in a carnivore model and the results suggest that the intestinal phase is relatively short as only very few worms were recovered 10 days post-inoculation (dpi). In mixed species infection with equal doses of T. nativa and T. spiralis, molecular typing demonstrated that 64% of the intestinal worms and 78% of the muscle larvae were T. nativa. Conversely, T. spiralis dominated in the mixed species infections with T. pseudospiralis, constituting 66% of the intestinal worms and 94% of the muscle larvae. Although, the individual recoveries of intestinal worms were only up to 5.6% on day 1, and up to 1.5% on day 4 post-infection, the muscle larvae establishment was comparable to other fox studies. Infectivity, measured as muscle larvae burden did not differ among the four species of Trichinella, which is in contrast to other models with mice, rats, pigs or herbivores. Although statistically significant differences in intestinal worm burdens were found for some days, no distinct species were recovered in consistently higher numbers than the others.  相似文献

A prospective study was carried out on 845 heifer calves born during 1991 on 30 Holstein dairy farms in southeast Minnesota. The objectives of the study were to describe the epidemiology of morbidity and mortality in dairy calves from birth to 16 weeks of age (with an emphasis on respiratory disease), to examine individual calf and herd management practices as risk factors for calf morbidity and mortality, and to validate producer diagnosis of mortality. Incidence rates for all morbidity, enteritis, and pneumonia were 0.20, 0.15, and 0.10 cases per 100 calf-days at risk for the period of the study. Risk of enteritis was highest in the first 3 weeks of life, with pneumonia risk highest at 10 weeks of age. Case fatality rates averaged 11.8%, 17.9%, and 9.4% for all diagnoses, enteritis, and pneumonia, respectively. Average daily rates of gain from birth to 16 weeks of age differed between farms that had inadequate calf housing (0.8 kg day−1) versus those with adequate calf housing (1.0 kg day−1). Approximately half of the calves in the cohort (418) had blood samples taken monthly from birth until 16 weeks of age. Of the calves sampled, only 19 calves showed a four-fold rise in serum titers to respiratory viruses. Sixteen calves seroconverted to BVDV, two calves to IBRV, and one calf to PI3 virus. Of 98 calves less than 10 days of age tested for adequacy of passive transfer, 35 (35.7%) had serum immunoglobulin levels of less than 800 mg dl−1. There were no significant differences in mortality or morbidity between calves that had adequate passive transfer and those that did not. The incidence of mortality was 0.08 deaths per 100 calf-days at risk; 64 calves died during the 16 months of the study. The risk of death was highest at 2 weeks of age. Enteritis was the most common cause of death (28 deaths, 44% of all deaths) followed by pneumonia (19 deaths, 30% of all deaths). Comparing producer diagnosis of mortality with necropsy results yielded sensitivities of 58.3% and 56% and specificities of 93% and 100% for producer diagnoses of enteritis and pneumonia, respectively. The kappa statistic comparing producer diagnosis with necropsy result was 0.47. The most common pathogens isolated from calves that died of enteritis were rotavirus (five calves), and Escherichia coli (four calves). Pathogens isolated from pneumonic lungs included Pasteurella multocida (three calves), Haemophilus somnus (three calves), and Pasteurella haemolytica (one calf).  相似文献

The association between mean daily weight gain, Theileria parva infections, clinical East Coast fever and other possible determinants of weight gain were examined in a longitudinal observational study that was conducted in cohorts of female calves from five agro-ecological zone (AEZ)-grazing strata. The strata were upper-midlands (UM) 1 zero-grazing, UM 1 open-grazing, UM 2 zero-grazing, UM 4 zero-grazing and UM 4 open-grazing. In total, 225 calves on 188 smallholder dairy farms were visited within the first 2 weeks of life and thereafter at biweekly intervals up to the age of 6 months between March 1995 and August 1996. During each visit, the calves were weighed and other calf-management practices in the farm during the visit such as housing, feeding and tick control also were recorded. Other events such as morbidity and mortality between or during the visits were also recorded.

The overall mean daily weight gains were 0.24–0.29 kg (S.D.=0.17–0.22 kg) and were lower than the recommended targets for smallholder farms of 0.40–0.50 kg. The major tendency in variability of daily weight gains was due to visit-to-visit variation (especially in calves >3 months old).

Differences in mean daily gains were associated with AEZ-grazing strata and calf-level factors that included breed of calf, calf sickness, incidence of ECF, feeding of milk, concentrate feeds and minerals and interaction between calf age and AEZ-grazing strata (P<0.05). ECF and other calf sicknesses exerted a temporal effect on calf-growth at the height of illness and immediately after; calves later recovered the lost growth except where other factors such as poor calf nutrition prevailed. Improvement in calf-growth in Murang’a District is achievable and extension services should continue to target individual-calf-level management practices.  相似文献

A study was undertaken to determine the prevalences of Cryptosporidium and Giardia in dairy calves less than 6 months of age in Norway. Faecal samples were collected from a total of 1386 calves, between 3 and 183 days of age, in 136 dairy farms from three different areas of Norway. Faecal samples were examined for Cryptosporidium oocysts and Giardia cysts after concentration and immunofluorescent staining.

Giardia was found in 93% (127 out of 136) of the farms and in 49% (679 out of 1386) of the calves. Cryptosporidium was found in 53% (72 out of 136) of the farms and in 12% (167 out of 1386) of the calves. The level of Giardia and/or Cryptosporidium was low in the majority of the infected calves.

Infection peaked in the age group 2–3 months for both Cryptosporidium and Giardia. The prevalences of both parasites were higher in samples taken during winter than in samples taken during summer, and statistically significant differences were found when prevalences in different age groups of calves were compared between the three areas. A significantly lower prevalence of Cryptosporidium was found in calves housed in shared pens that were thoroughly washed more than three times a year than in calves from pens washed less often. For Giardia there was a trend for decreasing intensity of infection with increasing age in the sampled calves. For Cryptosporidium there was a trend for increasing herd prevalence with increasing number of calves in the herd, but this trend was not statistically significant. Other parameters which were investigated such as housing, feeding or management routines were not associated with prevalence or intensity of infection with either parasite.  相似文献

Fecal specimens were collected from 30 calves from birth to 24 months of age at a dairy farm in Maryland to determine the prevalence and age distribution of Cryptosporidium species/genotypes. After centrifugation to remove debris and concentrate oocysts, specimens were examined by immunofluorescence microscopy and polymerase chain reaction (PCR). Fragments of the SSU-rDNA gene amplified by PCR were purified and PCR products were sequenced. All 30 calves shed Cryptosporidium oocysts at some time during the 24 months of the study. Of 990 specimens, 190 were Cryptosporidium-positive (19.2%). The highest prevalence of infection was at 2 weeks of age when 29 of the 30 calves were excreting oocysts. Prevalence was higher in pre-weaned calves (1-8 weeks of age) (45.8%) than in post-weaned calves (3-12 months of age) (18.5%) and heifers (12-24 months of age) (2.2%). Sequence data for 190 PCR-positive specimens identified: C. parvum, C. bovis, the Cryptosporidium deer-like genotype and C. andersoni, with cumulative prevalences of 100, 80, 60, and 3.3%, respectively. C. parvum constituted 97% of infections in pre-weaned calves but only 4% and 0% of infections in post-weaned calves and heifers, respectively.
