Background

Heart rate and plasma cortisol concentration are often used in evaluation of physiological reactions to stress and fear, but arterial blood pressure is rarely measured in farm animals. Goats are prey animals and can be expected to react strongly to a predator, especially when they have kids. We hypothesized that exposure to a dog elicits a flight response during pregnancy and a fight response when goats have kids to defend. Arterial blood pressure and heart rate should increase in both these cases, due to a synchronized discharge of the sympathetic nervous system.

Methods

Seven goats were exposed to a dog for 15 minutes at 12 ± 3 days before, and again at 10 ± 1 days after, parturition. Arterial blood pressure, heart rate, and activity were registered by telemetry. Behavioral data were collected during 5 minute sessions, followed by blood samples obtained via intrajugular catheters. Plasma cortisol concentration was analyzed by radioimmunoassay.

Results

At the appearance of the dog, the mean arterial blood pressure of the goats increased from 90 ± 8 to 111 ± 8 mmHg (p < 0.001) during pregnancy and from 96 ± 8 to 108 ± 8 mmHg during lactation (p < 0.001). Heart rate did not change at dog exposure during lactation, but increased from 117 ± 6 to 126 ± 10 beats/min (p < 0.01) during pregnancy. Dog exposure resulted in plasma cortisol concentration increasing from 17 ± 1 to 43 ± 7 nmol/l (p < 0.01) during pregnancy and from 21 ± 1 to 49 ± 6 nmol/l (p < 0.01) during lactation. In response to the dog, goats vocalized at a higher frequency and started to ruminate later during lactation compared to pregnancy.

Conclusions

When goats were exposed to a dog during pregnancy, their heart rate, blood pressure, and plasma cortisol increased, in contrast to lactation when only their blood pressure and plasma cortisol increased. However, when they were lactating, goats vocalized more and started to ruminate later compared to when they were pregnant.  相似文献

Serum and colostrum samples from goats at parturition and serum samples from their kids at 3 days and at 4, 7, 10 and 12 weeks after birth were examined for the presence of antibodies to Corynebacterium pseudotuberculosis hemolysins. The hemolysis inhibition test (HIT) was used. High correlation was found between titre values of antihemolysins in serum and colostrum of goats at parturition (correlation coefficient r = 0.83; P < 0.01). Intermediate correlation was found between antihemolysin titre in colostrum of goats and in the sera of their kids 3 days old (r = 0.56; 0.01 < P < 0.05). Furthermore, titre values for 3 day-old kids showed high correlation with the antihemolysin titres when the kids aged 4 and 7 weeks (r = 0.76 and 0.85, respectively; P < 0.01). Antihemolysin titres decreased linearly in kids from 3 days to 10 weeks old. Calculated half life of antibodies was 12 days. Most of the kids had detectable antibodies up to the age of 5–6 weeks. None of the kids were seropositive at 2½ months of age.Serum samples collected monthly from a group of kids chosen at random, aged 7–10 months, contained antibodies to hemolysins in half of the animals at the first testing. At the age of 10 months 14 out of 15 kids were seropositive. Thus, most of the kids from this herd were exposed to G. pseudotuberculosis antigens during summer and autumn of their first year of life.Prophylactic measures against caseous lymphadenitis is briefly discussed.  相似文献

The objectives were to investigate if partial substitution of a grazing diet with a supplemental high-starch feed during the last 12 d of gestation improved colostrum yield and neonatal activity in goat does grazing semi-arid rangeland. For the first objective, 25 pregnant does were randomly assigned to 1 of 2 treatments: 1) grazing only (control; n = 11), and 2) grazing plus 0.6 kg/d of supplemental corn (as fed), a high starch feed, during the last 12 ± 1.0 d before parturition (SC; n = 14). Colostrum was collected at parturition and again at 1, 3, 6, and 10 h postpartum for yield and composition. In all goats, blood concentrations of glucose and plasma progesterone were determined. Total colostrum yield/10 h was greater (P = 0.002) in the SC does (1,102 ± 144 g) than in control does (405 ± 50 g). From 6 to 10 h postpartum, contents of colostrum protein and solids not fat were less (P ≤ 0.012) whereas lactose contents were greater (P = 0.035) in the SC does than in control does. Concentration of glucose in the blood at parturition was greater (P = 0.037) in the SC does (160 ± 13 mg/dL) than in control does (115 ± 12 mg/dL). A significant decrease (P = 0.001) in plasma progesterone concentrations occurred 1 d before parturition in control does, whereas the decrease (P = 0.008) occurred 3 d before parturition in SC does. For the second objective, 20 does (10 per group) and their single kids were randomly assigned to the same treatments as above for evaluation of dietary treatment on neonatal activity. Kid activity was assessed using videos recorded during the first 90 min after birth. Frequency of low-pitched bleats was greater (P < 0.001) in kids of SC does than in kids of controls. Frequencies and durations of teat seeking from 30 to 90 min after birth and of suckling activity were greater (P ≤ 0.015) in kids of SC does than in kids of controls. Duration of parturition was longer (P = 0.001) in control does (58 ± 10.2 min) than in SC does (21 ± 2.8 min). Finally, percentage of fetal malpresentation was greater (P = 0.036) in control does (33.3%) than in SC does (9.0%). We conclude that partial substitution of the grazing diet with a high-starch feed, corn, during the last 12 d of gestation improved colostrum yield and neonatal activity in goats grazing subtropical semi-arid rangeland.  相似文献

The present experiments on goats were conducted with the aim of verifying (a) whether the identity of the goat kids can affect the characteristics of milk let-down during suckling (Study 1) and (b) whether the presence of the goat kids may improve the milk let-down during hand milking (Study 2). In Study 1, 16 lactating goats with twins and 12 with single kids were used. Dams with single goat kids ejected more milk when suckling their kids at Days 5 and 20 of lactation, than when suckling aliens. Dams with twins ejected more milk when suckling their own kids than aliens only at Day 5 of lactation. Taking into account all the dams, they ejected more milk with more fat, protein, and lactose during suckling their own litter than when suckling aliens. In Study 2, when dams (n = 13) were hand milked in the presence of their own litter or in its absence, they ejected more milk with more fat, protein, and lactose than when milked in the presence of alien kids. We conclude that milk let-down in goats can be influenced by the type of stimuli applied or to which they are exposed to during suckling and milking.  相似文献

Data from 82 kiddings from mixed-breed (native x dairy goats) goats reared on rangeland (n = 47) or confinement (n = 35) were used to determine the influence of various factors on length of labour, standing of newborn kids and time to first suckling in pluriparous goats. Both groups were mated at the same time to bucks of the same genotype in February. During the kidding time in July, the goats were monitored for 24 h in order to register parturition traits and kid birth weight and body measurements. For all goats, blood samples were collected at parturition in order to establish the nutritional state of goats. Grazing goats had lower body weight and body condition score (P < 0.01) than pen-fed goats, as well as lower values for some blood metabolites indicative of low body energy reserves. For both grazing and pen-fed goats, the majority of kiddings (>93%) occurred during daylight hours. No kiddings were registered from 2300 to 0500 hours. No effects of height at withers, thoracic circumference and sex of kids at birth were found to influence length of labour. Labour was shorter (P < 0.01) for pen-fed does than grazing goats (10.6 ± 1.18 min vs. 15.4 ± 1.2 min; mean ± SD). The length of parturition was considerably longer (P < 0.05) for twin-bearing does than for single-bearing does (16.6 ± 1.76 min vs. 11.2 ± 0.81 min, respectively). The interval between parturition and standing of kids was shorter (P < 0.05) for kids from litters weighing >3.3 kg (entire litter) than kids from litters with <3.3 kg (22.7 ± 1.82 min vs. 31.1 ± 2.76 min). Time to first suckling was longer (P < 0.05) for kids delivered from pen-fed does than kids from grazing does (57.8 ± 7.57 min vs. 42.9 ± 3.05 min). These results confirm that, independently of type of production system (intensive or extensive), kiddings of does are concentrated during daylight hours. These results also indicate that conformation of kids does not alter duration of labour, and that nutritional stress of goats during gestation (extensive conditions) does not interfere with the neonate’s level of alertness and arousal.  相似文献

Background

Since transport evokes physiological adjustments that include endocrine responses, the objective of this study was to examine the responses of circulating β-endorphin, adrenocorticotrophic hormone (ACTH) and cortisol levels to transport stress in stallions.

Methods

Forty-two healthy Thoroughbred and crossbred stallions were studied before and after road transport over distances of 100, 200 and 300 km. Blood samples were collected from the jugular vein: first in a single box immediately before loading (pre-samples), then immediately after transport and unloading on arrival at the breeding stations (post-samples).

Results

An increase in circulating β-endorphin levels after transport of 100 km (P < 0.01), compared to basal values was observed. Circulating ACTH levels showed significant increases after transport of 100 km (P < 0.001) and 200 km (P < 0.001). Circulating cortisol levels showed significant increases after road transport over distances of 100, 200 and 300 km (P < 0.001). An effect of transport on β-endorphin, ACTH and cortisol variations was therefore evident for the different distances studied. No significant differences (P > 0.05) between horses of different ages and different breeds were observed for β-endorphin, ACTH and cortisol levels.

Conclusion

The results obtained for short term transportation of stallions showed a very strong reaction of the adrenocortical system. The lack of response of β-endorphin after transport of 200–300 km and of ACTH after transport of 300 km seems to suggest a soothing effect of negative feedback of ACTH and cortisol levels.  相似文献

Background

Objectives were to examine the effects of selenium (Se) supply and maternal nutritional plane during gestation on mammary gland growth, cellular proliferation, and vascularity at parturition and d 20 of lactation. Rambouillet primiparous ewes (n = 84) were allocated to treatments in a 2 x 3 factorial. Factors were dietary Se (adequate Se [ASe, 11.5 μg/kg BW] or high Se [HSe, 77.0 μg/kg BW]) and nutritional plane (60% [RES], 100% [CON], or 140% [EXC]). At parturition, lambs were removed and 42 ewes (7/treatment) were necropsied. Remaining ewes were fed a common diet meeting requirements for lactation and mechanically milked twice daily until necropsy on d 20. At both necropsy periods, mammary glands were dissected and tissues harvested. Samples were analyzed for RNA, DNA, and protein content, cell proliferation, and vascularity. Where interactions were present (P ≤ 0.05), least squares means from the highest-order interaction are presented.

Results

Final body weight of ewes was least (P ≤ 0.002) in RES, intermediate for CON, and greatest for EXC, regardless of stage of the ewe at necropsy (parturition or d 20 of lactation). In ewes necropsied at parturition, mammary glands were heavier (P = 0.02) in EXC compared to RES, with CON intermediate. Concentration of RNA (mg/g) was decreased (P = 0.01) in EXC compared to CON at parturition. There was a tendency (P = 0.07) for a Se by nutrition interaction in percentage of cells proliferating where ASe-EXC ewes had greater (P ≤ 0.02) number of proliferating cells then all other treatments. Mammary vascular area tended (P = 0.08) to be affected by a Se by nutrition interaction where ASe-CON had less (P = 0.007) vascular area than HSe-CON ewes. In ewes necropsied at d 20 of lactation, the number of alveoli per area was decreased (P ≤ 0.05) in RES compared to CON and EXC-fed ewes.

Conclusions

Results of this study indicate that proper maternal nutritional plane during gestation is important for mammary gland development, even out to d 20 of lactation.  相似文献

Background

This study was performed to characterize a gene-addition transgenic mouse containing a BAC (bacterial artificial chromosome) clone spanning the human CYP2C18&19 genes (tg-CYP2C18&19).

Methods

Hemizygous tg-CYP2C18&19, 11 week old mice were compared with wild-type littermates to obtain information regarding clinical status, clinical pathology and anatomical pathology. After one week of clinical observations, blood samples were collected, organs weighed, and tissues collected for histopathology.

Results

In males, the tissue weights were lower in tg-CYP2C18&19 than in wild-type mice for brain (p ≤ 0.05), adrenal glands (p ≤ 0.05) and brown fat deposits (p ≤ 0.001) while the heart weight was higher (p ≤ 0.001). In female tg-CYP2C18&19, the tissue weights were lower for brain (p ≤ 0.001) and spleen (p ≤ 0.001) compared to wild-type females. Male tg-CYP2C18&19 had increased blood glucose levels (p ≤ 0.01) while females had decreased blood triglyceride levels (p ≤ 0.01).

Conclusion

Despite the observed alterations, tg-CYP2C18&19 did not show any macroscopic or microscopic pathology at the examined age. Hence, these hemizygous transgenic mice were considered to be viable and healthy animals.  相似文献

The effect of induction of parturition with a PGF(2)α analog on plasma concentration of prolactin (PRL) and its effects on colostrum concentration of IgG and chitotriosidase (ChT) activity were studied in 16 pregnant Majorera goats. Treated goats, those in which parturition was induced, had greater concentrations of PRL than control goats 24 h before parturition (P < 0.05) and 48 h after parturition (P < 0.05). Control goats had greater concentrations of PRL than treated goats 96 h after parturition (P < 0.05). Plasma concentration of IgG did not differ between groups during the experimental period, but colostrum concentrations of IgG were greater in control goats than in treated goats at parturition (P < 0.05). Plasma ChT activity decreased during the period 72 h before parturition to 24 h after parturition in control and treated goats. Time evolution after partum affected the colostrum ChT activity, being greater at parturition than after parturition in both groups (P < 0.05). In summary, concentration of IgG in colostrum is slightly diminished if parturition is induced. Induction of parturition causes an early increase in PRL, which is most likely responsible for preterm suppression of IgG transport into mammary secretions.  相似文献

Background

Dairy goats are commonly housed at a space allowance of 0.7 – 0.8 m2/goat in commercial Norwegian goat herds, which is very low compared to regulations and recommendations in other European countries. One easy and cheap way to increase space allowance is to allow the animals’ access to outdoor area. The aim of this study was to investigate the effect of access to an outside enclosure and environmental enrichment for dairy goats kept in slatted floor pens with low space allowance on their activity pattern and social behaviour.

Methods

A group of 82 dairy goats on a commercial Norwegian dairy farm were kept inside during the winter period from October to April. In April the goats were given access to an outside enclosure for 8 hours per day. After having access to the enclosure for another for two days, enrichment (branches) was provided, and after 19 days the enrichment were removed. The goats were observed for 5 hours per day for the two last days before they got access to the outside enclosure, the two days in the enclosure, the two first and the two last days with enrichment and for the following two days without enrichment by two trained observers.

Results

When allowed access to the enclosure, the goats spent nearly 50% of the time outside, and later the time spent outside was reduced to less than 40% (P < 0.0001), but there was no clear effect of enrichment. All the goats appeared to have a regular use of the enclosure. Time spent resting decreased 59.2% to only 25.2% when the goats first got access to the enclosure, but then started to increase again (P < 0.0001). Initially time spent exploring and chewing the branches was 20%, but this was reduced to around 12% in the last part of the ENRICH period (P < 0.0001). Number of aggressive interactions tended to increase when the goats were allowed access to the outdoor enclosure whereas play behaviour was only observed in the outside enclosure (P < 0.05).

Conclusions

In conclusion, the goats preferred to use the outside enclosure when being active, and branches were perceived as an attractive enrichment.  相似文献

Two experiments were conducted to determine whether exposure to a photoperiod of artificial long days in autumn increased milk yield in subtropical goats milked once (Exp. I) or twice daily (Exp. II). In Exp. I, starting at d 10 of lactation, 1 group of does was kept under naturally decreasing photoperiod (DD1X; n = 8), whereas the other group was submitted to an artificial photoperiod of long days (LD1X; n = 8; 16 h light:8 h darkness). The kids were weaned 28 d after parturition, and dams were manually milked once daily. Milk yield and milk components (fat, protein, and lactose) were assessed up to 140 d of lactation. From d 0 to 28 of lactation (suckling phase), mean daily milk yield did not differ between DD1X and LD1X goats (2.3 ± 0.2 kg vs. 2.4 ± 0.2 kg; P = 0.717). However, between d 29 and 84 (early milking phase), mean daily milk yield was greater in LD1X does than in DD1X does (2.6 ± 0.1 kg vs. 2.1 ± 0.1 kg; P = 0.001). Finally, between d 85 and 140 (late milking phase), mean daily milk yield was greater in LD1X goats than in DD1X goats (P ≤ 0.05) only during the first 2 wk. In Exp. II, one group of goats was exposed to a photoperiod of naturally decreasing days (DD2X; n = 8) and another group was submitted to an artificial photoperiod of long days (LD2X; n = 7). In both groups, kids were weaned on d 28 of lactation and the dams were manually milked twice daily. During the nursing phase, mean daily milk yield did not differ between the DD2X and LD2X groups (2.5 ± 0.3 kg vs. 2.6 ± 0.2 kg; P = 0.767). In the early milking phase, mean daily milk yield was greater in LD2X than in DD2X goats (3.3 ± 0.2 kg vs. 2.8 ± 0.2 kg; P = 0.022), whereas during the late milking phase, milk yield did not differ between the 2 groups (P = 0.946). In both experiments, milk composition was not significantly influenced by exposure to long-day photoperiod. We conclude that, in subtropical female goats that start lactation in late autumn, exposure to an artificial long-day photoperiod stimulates milk yield, even if goats are milked once daily. In addition, combining exposure to long days with twice-daily milking will increase further milk yield in such goats without affecting milk components.  相似文献

Background

Insulin secretion and tissue sensitivity to insulin is considered to be one of the factors controlling lipid metabolism post partum. The objective of this study was to compare glucose-induced blood insulin and metabolite responses in Estonian Holstein (EH, n = 14) and Estonian Red (ER, n = 14) cows.

Methods

The study was carried out using the glucose tolerance test (GTT) performed at 31 ± 1.9 days post partum during negative energy balance. Blood samples were obtained at -15, -5, 5, 10, 20, 30, 40, 50 and 60 min relative to infusion of 0.15 g/kg BW glucose and analysed for glucose, insulin, triglycerides (TG), non-esterified fatty acids (NEFA), cholesterol and β-hydroxybutyrate (BHB). Applying the MIXED Procedure with the SAS System the basal concentration of cholesterol, and basal concentration and concentrations at post-infusion time points for other metabolites, area under the curve (AUC) for glucose and insulin, clearance rate (CR) for glucose, and maximum increase from basal concentration for glucose and insulin were compared between breeds.

Results

There was a breed effect on blood NEFA (P < 0.05) and a time effect on all metabolites concentration (P < 0.01). The following differences were observed in EH compared to ER: lower blood insulin concentration 5 min after glucose infusion (P < 0.05), higher glucose concentration 20 (P < 0.01) and 30 min (P < 0.05) after infusion, and higher NEFA concentration before (P < 0.01) and 5 min after infusion (P < 0.05). Blood TG concentration in ER remained stable, while in EH there was a decrease from the basal level to the 40th min nadir (P < 0.01), followed by an increase to the 60th min postinfusion (P < 0.01).

Conclusion

Our results imply that glucose-induced changes in insulin concentration and metabolite responses to insulin differ between EH and ER dairy cows.  相似文献

Background

The main objective of this study was to estimate the effect of supplementation with Saccaromyces cerevisiae (SC) (Yea-Sacc® 1026) on milk production, metabolic parameters and the resumption of ovarian activity in early lactation dairy cows.

Methods

The experiment was conducted during 2005/2006 in a commercial tied-house farm with an average of 200 milking Estonian Holstein Friesian cows. The late pregnant multiparous cows (n = 46) were randomly divided into two groups; one group received 10 g yeast culture from two weeks before to 14 weeks after calving. The groups were fed a total mixed ration with silages and concentrates. Milk recording data and blood samples for plasma metabolites were taken. Resumption of luteal activity was determined using milk progesterone (P4) measurements. Uterine bacteriology and ovarian ultrasonography (US) were performed and body condition scores (BCS) and clinical disease occurrences were recorded. For analysis, the statistical software Stata 9.2 and R were used to compute Cox proportional hazard and linear mixed models.

Results

The average milk production per cow did not differ between the groups (32.7 ± 6.4 vs 30.7 ± 5.3 kg/day in the SC and control groups respectively), but the production of milk fat (P < 0.001) and milk protein (P < 0.001) were higher in the SC group. There was no effect of treatment on BCS. The analysis of energy-related metabolites in early lactation showed no significant differences between the groups. In both groups higher levels of β-hydroxybutyrate (BHB) appeared from days 14 to 28 after parturition and the concentration of non-esterfied fatty acid (NEFA) was higher from days 1–7 post partum (PP). According to US and P4 results, all cows in both groups ovulated during the experimental period. The resumption of ovarian activity (first ovulations) and time required for elimination of bacteria from the uterus did not differ between the groups.

Conclusion

Supplementation with SC had an effect on milk protein and fat production, but did not influence the milk yield. No effects on PP metabolic status, bacterial elimination from the uterus nor the resumption of ovarian activity were found.  相似文献

Background

Proximal cytoplasmic droplets (PCDs), a remnant of germ cell cytoplasm, are common non-specific morphological defects in bovine semen. This study evaluated the effect of higher percentages of PCDs on the quality of frozen-thawed bovine semen, embryo production and early embryo development.

Methods

Three ejaculates from each of five (group 1: PCD ≤ 1%, control) and eight adult Bos indicus bulls (group 2: PCD ≥ 24%) were analysed. Semen samples were examined for: post-thaw motility, vigour of movement, concentration, sperm morphology, slow thermoresistance test (STT), membrane integrity, acrosome status, mitochondrial function using fluorescent probes association (FITC-PSA, PI and JC-1) and sperm chromatin integrity using acridine orange assay. Two bulls from group 2, with 28.5% and 48.5% PCD, respectively, and three bulls from the control group, each with 0% PCD, were selected for IVF (in vitro fertilisation).

Results

Semen analyses revealed a significant correlation (P < 0.01) between increased rates of PCD and sperm quality traits. Nevertheless, no differences were observed in sperm motility and vigour either before or after the STT or in the percentage of intact acrosomes (analysed by differential interference contrast microscopy (DIC) after STT), but membrane integrity, acrosome status (evaluated with FITC-PSA staining method after thawing) and mitochondrial function were reduced, when compared with group 1 (P < 0.05). The higher incidence of PCD was positively correlated to chromatin damage, especially after three hours of incubation at 37°C. IVF showed similar results for bull C2 (group 1, control) and bull P2 (group 2, group with higher PCDs).

Conclusion

Higher PCD levels influenced spermatozoa quality traits. IVF and embryo development data showed that cleavage, blastocyst formation and blastocyst hatching may have been influenced by the interaction of morphology traits and individual bull effects.  相似文献

Forty female goats in the third parity were randomly divided into four similar groups. The experiment was started 20 days before mating and lasted until the end of the suckling period for 60 days and weaning their kids. The first group were fed diet without supplementation and kept as control while in the second, third and fourth groups, each doe was fed diets with rumen protected choline (RPC) at the rate of 10, 20 and 40 g/day, respectively. Results showed that number of doe kidding twins and triplets and litter weight of kids born per group increased with increasing the level of RPC in the diet of goats and viability rate of born kids during the suckling period improved due to RPC supplementation in the diets of their mothers. Duration of estrous, days from weaning to estrous, days from kidding to estrous and kidding interval decreased significantly, while conception rate increased due to adding RPC. Milk choline concentrations and total choline secretion though milk were progressively increased significantly with increasing the level of RPC supplementation. Live body weight and daily body gain of their suckling male and female kids at weaning increased significantly with increasing RPC levels in the diets of their mothers.  相似文献

Twenty-two pregnant goats of the Canarian breed were administered 10 mg of the prostaglandin analogue luprostiol intramuscular (IM) on day 145 of pregnancy; seventeen pregnant goats were injected IM with saline solution the same day. Twenty hours after luprostiol application, goats were continuously observed in order to record the following parameters: parturition, dystocia incidence, placental delivery and live births. Parturition was successfully induced in all goats; the mean interval to parturition (first kid delivery) was of 32.9 ± 1.0 (SEM) hours (h) and the range varied from 21 to 41 h; in addition, no significant differences (p > 0.1) were detected between nuliparous and multiparous goats (31.3 ± 1.3 h and 34.7 ± 1.2 h, respectively). In the control group, the interval from saline injection to parturition was 75.9 ± 12.5 h (mean ± SEM), showing a range between 34 and 138 h. All the goats expelled the fetal membranes in the first 6 h after the induced or natural parturition. The incidence of dystocia due to fetal maldisposition was not significantly different between induced and control goats (9.0% and 11.8%, respectively). In addition, the percentage of live kids was practically similar between both groups (90.7% and 88.0%, induced vs control goats). This study confirms the effectiveness of the luprostiol to induce the parturition in goats, showing a narrow range (30–37 h) in most of the induced females (86.3%; 19/22 goats). The role of opioid peptides in the secretion of oxytocin (OT) and prolactin (PRL) induced by sucking was studied in goats. Seven goats were isolated with their kids (four singletons and three twins) in individual corrals 3–4 weeks after parturition. On day 1 of the experiment, the kids were separated from the does for 7 h and were weighed before and 15 min after being reunited with their mothers to assess the amount of milk obtained by sucking. The does were blood‐sampled 10 min before and at the end of the sucking period. On day 2, a similar protocol was followed, but naloxone was given immediately after the first blood sample. On day 3, the protocol was repeated but saline vehicle was injected instead of naloxone. On day 5, the naloxone experiment was repeated as on day 2. Milk ejection was evaluated as the difference in the weight of the kids before and after sucking for 15 min, and the maternal serum levels of OT and PRL were measured by radioimmunoassay. A significant decrease in the weight gain of the kids was obtained when the mothers were treated with naloxone on day 2. Consistently, serum levels of OT and PRL induced by sucking were significantly reduced; indicating that sucking‐induced OT secretion for milk ejection in lactating goats is facilitated by opioid peptides. In a second experiment performed in the same animals 10 days later, the administration of OT, immediately after naloxone administration, prevented the decrease in the weight gain induced by naloxone, suggesting that the effect of the opioid antagonist on milk ejection in goats is a result of a reduced OT secretion. The results of this study confirm the importance of sucking‐induced OT secretion for milk ejection in lactating goats, and indicate that OT and PRL secretion are regulated by opioid peptides in this species.  相似文献

Pregnant goats were induced to parturition on day 145 of pregnancy, with three different protocols: group Cl (n = 19) was injected intramuscularly (IM) with 75 μg of the prostaglandin analogue R‐Cloprostenol; group L (n = 20) was treated IM with 7.5 mg of the prostaglandin analogue Luprostiol; group L50 (n = 18) was injected IM with 3.75 mg of Luprostiol (IM); in addition, Group S (Control, n = 15) was injected IM with 1 ml of saline solution. Thereafter, goats were continuously observed to record the following parameters: parturition, dystocia incidence, placental delivery and kid and maternal survival. Moreover, blood sampling was performed around kidding and plasma progesterone concentrations were analyzed. The interval from injection to parturition (mean ± SEM) was not significantly different among the experimental groups: 35.1 ± 1.5 h, 33.3 ± 0.9 h and 34.1 ± 1.8 h (groups Cl, L and L50, respectively). In the control group, time to parturition was 99.4 ± 12.1 h (range: 34–166 h). All the goats expelled the foetal membranes within the first 2 h after the induction. The incidence of dystocia due to foetal posture was not significantly different between induced and control goats (21.1%, 20.0%, 22.0% and 20%, for groups Cl, L, L50 and S, respectively). The percentage of live kids was practically similar between induced goats (93.9%, 94.9% and 92.1%, for groups Cl, L and L50, respectively); in addition, there was a case of maternal mortality in control group (6.7%; 1/15), whereas there was no mortality in induced goats (0%; 0/57). Plasma concentrations of progesterone showed an intense drop (<2 ng/ml) at 24 h after induction. This study confirms the effectiveness of the luprostiol to induce the parturition in goats, within a narrow range (30–40 h) in most of the induced females (80.0%, 7.5 mg; 77.8%, 3.75 mg).  相似文献

Background

Sedation with α2-agonists in the horse is reported to be accompanied by impairment of arterial oxygenation. The present study was undertaken to investigate pulmonary gas exchange using the Multiple Inert Gas Elimination Technique (MIGET), during sedation with the α2-agonist detomidine alone and in combination with the opioid butorphanol.

Methods

Seven Standardbred trotter horses aged 3–7 years and weighing 380–520 kg, were studied. The protocol consisted of three consecutive measurements; in the unsedated horse, after intravenous administration of detomidine (0.02 mg/kg) and after subsequent butorphanol administration (0.025 mg/kg). Pulmonary function and haemodynamic effects were investigated. The distribution of ventilation-perfusion ratios (VA/Q) was estimated with MIGET.

Results

During detomidine sedation, arterial oxygen tension (PaO2) decreased (12.8 ± 0.7 to 10.8 ± 1.2 kPa) and arterial carbon dioxide tension (PaCO2) increased (5.9 ± 0.3 to 6.1 ± 0.2 kPa) compared to measurements in the unsedated horse. Mismatch between ventilation and perfusion in the lungs was evident, but no increase in intrapulmonary shunt could be detected. Respiratory rate and minute ventilation did not change. Heart rate and cardiac output decreased, while pulmonary and systemic blood pressure and vascular resistance increased. Addition of butorphanol resulted in a significant decrease in ventilation and increase in PaCO2. Alveolar-arterial oxygen content difference P(A-a)O2 remained impaired after butorphanol administration, the VA/Q distribution improved as the decreased ventilation and persistent low blood flow was well matched. Also after subsequent butorphanol no increase in intrapulmonary shunt was evident.

Conclusion

The results of the present study suggest that both pulmonary and cardiovascular factors contribute to the impaired pulmonary gas exchange during detomidine and butorphanol sedation in the horse.  相似文献

Background

Exogenous melatonin could induce cashmere growth. However, induced growth of cashmere fleece by melatonin implants cannot be combined with the typical growth, resulting in earlier shedding followed by another cycle of cashmere growth. To address this issue, we examine the effects on the cashmere yield, fibre characteristics, and the growth and reproductive performance of cashmere goats of planned administration of melatonin.

Methods

Eighteen half-sib, female goats were assigned to two treatments (n = 9) including a control and a treatment where melatonin (2 mg/kg BW) was implanted at the end of April and end of June. Cashmere growth and shedding were observed for approximately 1 year following implantation. Fibre samples were collected monthly to determine cumulative cashmere length. Initiation and cessation dates for cashmere growth as well as the rate of cashmere growth were calculated. Cashmere yield, weight gain of dam, kidding date, litter size, and birth weight were also recorded.

Results

Melatonin implantation increased cashmere yield by 34.5 % (control 553.7 g vs. melatonin 745.0 g; P < 0.01), cashmere length by 21.3 % (control 95.2 mm vs. melatonin 115.4 mm; P < 0.01), and decreased fibre diameter by 4.4 % (control 14.6 μm vs. melatonin 14.0 μm; P < 0.03). In melatonin-treated goats, the average initiation date was earlier than in control goats (May 18, 2013 vs. July 2, 2013; P < 0.01) but there was a similar cessation date (March 22, 2014 vs. March 27, 2014). Consequently, the duration of cashmere growth was longer in melatonin-treated goats than in control goats (307 vs.270 days; P < 0.01). The final BW, average daily gain, kidding date, litter size, and birth weight were not influenced by melatonin implantation.

Conclusions

These data indicate that melatonin implantation (2 mg/kg BW) on two occasions (late April and June) increased cashmere yield by combining the induced growth of cashmere fleece with the typical growth and decreased the fibre diameter without changing dam growth rate or reproductive performance.  相似文献
