Constituents of the leaves of Holarrhena pubescens

The leaves of Holarrhena pubescens yielded a new naringenin glycoside (1), together with naringin, naringenin 7-O-beta-D-glucoside, lupeol, lupeol beta-hydroxyhexadecanoate and ursolic acid. The structure of 1 was determined by spectral analysis.

介绍了银杏叶烷基酚化合物的分离过程，并鉴定出烷基酚化合物单体3类6种，其中银杏酸类化合物2种，白果酚类化合物3种，白果二酚类化合物1种。这些化合物经UV、IR、MS、^1HNMR、^13CNMR等波谱鉴定为：3-（8-十五碳烯基）苯酚；3-[4（Z），7（Z）-十七碳二烯基]苯酚；3-（10-十七碳烯基）苯酚；6-（8-十五碳烯基）水杨酸；6（10-十七碳烯基）十七碳烯基）水杨酸；5-（8-十五碳烯基）间苯二酚。     

乐昌含笑叶挥发油GC-MS及其活性初步测定

采用气-质联用法对乐昌含笑叶挥发油的化学成分研究表明,该油含23种化合物,其含量较高的物质有:十四氢化芘(18.891%);3,6-二甲基-5-丙烯基-6-乙烯基-4,5,6,7-四氢-1-苯并呋喃(16.066%);2,5-二环戊亚基-环戊酮(9.436%);3,3-二甲基-5-叔丁基-1-茚酮(8.892%);乙酸-3a,9b-二甲基-7-羰十氢化环戊并a萘-3-酯(8.024%);1,3-二甲基-2-丁烯基-5-叔丁基苯(5.720%);β-丁香烯(4.719%);5,11(13)-桉叶二烯-8,12-内酯(4.005%);β-橄香烯(3.232%);γ-杜松烯(1.977%);β-法尼烯(1.810%);含木香内酯(1.084%)。文章还对其挥发油的活性进行了初步测定。       

Further degradation product of hyperforin from Hypericum perforatum (St John's Wort)

Repeated examination of the aerial parts of Hypericum perforatum yielded a new degradation product of hyperforin (1) namely deoxyfurohyperforin A (2), together with the previously identified furohyperforin (3), furoadhyperforin (4), furohyperforin A (5a and 5b), pyrano[7,28-b]hyperforin (6) and 3-methyl-4,6-di(3-methyl-2-butenyl)-2-(2-methyl-1-oxopropyl)-3-(4-methyl-3-pentenyl)-cyclohexanone (7). The antimicrobial activity of the compounds 3, 5a and 5b, 6 and 7 was tested against Staphylococcus aureus, Candida albicans, Bacillus subtilis and Escherichia coli.     

Iridoid, phenylethanoid and flavonoid glycosides from Sideritis trojana

From the MeOH extract of Sideritis trojana, a new iridoid glycoside, 10-O-(E)-feruloylmelittoside (1) was obtained in addition to four known iridoid glycosides [melittoside (2), 10-O-(E)-p-coumaroylmelittoside (3), stachysosides E (4) and G (5)]. Moreover, five phenylethanoid glycosides [verbascoside (6), isoacteoside (7), lamalboside (8), leonoside A (9), isolavandulifolioside (10), three flavone glycosides (isoscutellarein 7-O-[6'-O-acetyl-β-allopyranosyl-(1→2)]-β-glucopyranoside (11), 4'-O-methyisoscutellarein 7-O-[6'-O-acetyl-β-allopyranosyl-(1→2)]-β-glucopyranoside (12), 3'-hydroxy-4'-O-methyisoscutellarein 7-O-[6'-O-acetyl-β-allopyranosyl-(1→2)]-β-glucopyranoside (13) and a benzylalcohol derivative (di-O-methylcrenatin) were obtained and identified. The structures were elucidated on the basis of NMR and HRMS data. All compounds were tested for their antioxidant activity by in vitro TEAC assay and some of them exhibited moderate activity (0.97-1.44 mM) when compared with the reference compound (quercetin 1.86 mM). Glycosides 6-13, the most active compounds in the TEAC assay, were also tested by flow cytometry to evaluate their ability to affect the levels of reactive oxygen species (ROS) in human prostate cancer cells (PC3).     

Phenolics from Bidens bipinnata and their amylase inhibitory properties

A new chlorinated flavonoid, 3, 6, 8-trichloro-5, 7, 3', 4'-tetrahydroxyflavone (1), a new biscoumaric acid derivative, 4-O-(2″, 3″-O-diacetyl-6″-O-p-coumaroyl-β-d-glucopyranosyl)-p-coumaric acid (2), and 8, 3', 4'-trihydroxyflavone-7-O-β-d-glucopyranoside (3) together with twenty-four known compounds (4-27) were isolated from the whole plant of Bidens bipinnata. All chemical structures were established on the basis of UV-, MS- and NMR ((1)H, (13)C, (1)H-(1)H COSY, HMQC and HMBC) spectroscopic data. Some of the isolated compounds were tested for the inhibition of α-amylase. The result showed that isookanin (6) was a potent inhibitor of α-amylase (IC(50)=0.447mg/ml).     

Flavonol glycosides from Mentha lavandulacea.

In addition to kaempferol 3-O-rhamnoside-7-O-glucoside, the methanol extract of the aerial parts of Mentha lavandulacea yielded three new flavonoids identified as kaempferol 3-O-glucosyl-(1-->2)-rhamnoside-7-O-glucoside (1), kaempferol 3-O-(6"'-p-coumaroylglucosyl)-(1-->2)-rhamnoside-7-O-glucoside (2) and kaempferol 3-O-(4"'-p-coumaroylglucosyl)-(1-->2)-rhamnoside-7-O-glucoside (3).     

New neolignans from Talauma ovata

The hexane extract of the bark of Talauma ovata yielded three new natural neolignans, identified as acetyl oleiferin-C (4), acetyl oleiferin-F (7) and acetyl oleiferin-G (8), together with the known compounds dihydroguaiaretic acid (1), austrobailignan-5 (2), oleiferin-C (3), austrobailignan-6 (5) and oleiferin-F (6).

New coumarins from Cedrelopsis grevei

Two new coumarins, 7-methoxy-5-prenylcoumarin (isocedrelopsin) (1) and 3',4'-dihydrobraylin (2), were isolated from the trunk bark of Cedrelopsis grevei, along with five known coumarins, microfolicoumarin (6,7-dimethoxy-5-prenylcoumarin, (3), obliquin (4), 8-methoxyobliquin (5), aesculetin (6), cedrelopsin (7) and scoparone (8).     

In vivo antidepressant activity of sesquiterpenes from the roots of Valeriana fauriei Briq

Antidepressant activity-guided fractionation of the MeOH extract of Valeriana fauriei Briq. roots resulted in the isolation of two new germacrane-type sesquiterpenes (1-2) in addition to seven known ones (3-9). Their structures were elucidated as 1β,10α-dihydroxyl-8α-acetoxyl-10β,11,11-trimethyl-4-formyl-bicyclogermacren-E-4(5)-ene (1), 1β-hydroxyl-8α-acetoxyl-11,11-dimethyl-4-formyl-bicyclogermacren-E-4(5),10(14)-diene (2), bicyclo[8,1,0]5β-hydroxyl-7β-1acetoxyl-5α,11,11'-trimethyl -E-1(10)-ene-4α,15-olide (3), 8α-acetoxyl-3α,4α,10-trihydroxyl-guaia-1(2)-ene-12,6α-olide (4), 2-Ethylhexyl-4-hydroxybenzoate (5), 11αH-gemacra-1(10)E,4Z-diene-3-one-12,6α-olide (6), β-Sitoterol (7), isovaleric acid (8), isoborneol acetate (9), using a combination of UV, IR, mass spectroscopy, 1D and 2D NMR spectroscopy. The antidepressant activity of compounds 1-4 was investigated by the FST on mice. Among them, compounds 3 and 4 showed the significant antidepressant activity (*, P<0.01).     

Isoflavone content and estrogenic activity of different batches of red clover (Trifolium pratense L.) extracts: An in vitro study in MCF-7 cells

The estrogenicity of different batches of red clover (Trifolium pratense L., Fabaceae; RCL) extracts and its relationship with the isoflavone content were assessed by measuring MCF-7 cell proliferation by flow cytometry and propidium iodide staining. RCL extracts were compared to estradiol (E2) and to the main RCL isoflavones biochanin A, daidzein, genistein and formononetin. Isoflavone content in the extracts was assayed by HPLC.E2 and isoflavones increased MCF-7 proliferation in a concentration-dependent fashion, with the following potency order: E2 >>> genistein > biochanin A = daidzein > formononetin. Extracts increased MCF-7 proliferation with different potencies, which in four out of five extracts correlated with the ratios 5,7-dihydroxyisoflavones/7-hydroxyisoflavones. The efficacy of all extracts increased with decreasing genistein contents. A solution containing the main isoflavones at the average concentration of RCL extracts increased MCF-7 proliferation with higher potency and steeper concentration–response curve. The effects of E2, of RCL extracts and of the isoflavone solution were inhibited by the estrogen receptor antagonist 4-hydroxytamoxifen.Flow cytometric analysis of MCF-7 proliferation is a suitable bioassay for the estrogenicity of RCL extracts, thus expanding the characterization of individual batches beyond assessment of chemical composition and contributing to improved standardization of quality and activity.     

Constituents of Cassia laevigata.

Calendin (1), cinnamic acid (2), 3-hydroxy-1-(4-hydroxy-3-methoxyphenyl)-propan-1-one (3), 2,3-dihydroxy-1-(4-hydroxy-3-methoxyphenyl)-propan-1-one (4), 3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-propan-1-one (5), syringic acid (6) and vanillic acid (7) have been isolated from a dichloromethane extract of the leaves and branches of Cassia laevigata.     

Flavonoids with antiplasmodial and cytotoxic activities of Macaranga triloba

A new flavanone derivative, malaysianone A (1), four prenylated flavanones, 6-prenyl-3'-methoxyeriodictyol (2), nymphaeol B (3), nymphaeol C (4) and 6-farnesyl-3',4',5,7-tetrahydroxyflavanone (5), and two coumarins, 5,7-dihydroxycoumarin (6) and scopoletin (7), were isolated from the dichloromethane extract of the inflorescences of Macaranga triloba. The structures of these compounds were elucidated based on spectroscopic methods including nuclear magnetic resonance (NMR-1D and 2D), UV, IR and mass spectrometry. The cytotoxic activity of the compounds was tested against several cell lines, with 5 inhibiting very strongly the growth of HeLa and HL-60 cells (IC(50): 1.3 μg/ml and 3.3 μg/ml, respectively). Compound 5 also showed strong antiplasmodial activity (IC(50): 0.06 μM).     

An anti-inflammatory and immunomodulatory polysaccharide from Orbignya phalerata

A polysaccharide, a glucan with mean M_r of 1.0×10⁶ (MP1), was isolated from the mesocarp of fruits of Orbignya phalerata. Chemical and spectroscopic studies indicated that MP1 has a highly branched glucan type structure composed of α-(1→4) linked -glucopyranose residues with (3→4), (4→6), and with (3→6) branching points. MP1 enhanced phagocytosis in vivo and exhibited anti-inflammatory activity.     

漆树木粉醇提物中多酚化合物的分离及结构表征

采用硅胶柱层析、中低压色谱和制备高效液相色谱协同活性跟踪方法,对漆树木粉醇提取物乙酸乙酯部位的具有抑制酪氨酸酶活性的化合物进行分离,并用红外、质谱和核磁等色谱分析手段对化合物的结构进行表征。从漆树木粉提取物乙酸乙酯部位分离得到7种多酚化合物,分别为没食子酸(1)、黄颜木素(2)、漆黄素(3)、硫黄菊素(4)、3,4',7-三羟基二氢黄酮醇(5)、槲皮素(6)和紫铆花素(7)。各化合物抑制酪氨酸酶活性(半数抑制质量浓度,IC_(50))由强到弱顺序:硫黄菊素(3.74 mg/L)、紫铆花素(4.32 mg/L)、槲皮素(18.73 mg/L)、漆黄素(69.4 mg/L)、没食子酸(89.5 mg/L)、黄颜木素(100 mg/L)和3,4',7-三羟基二氢黄酮醇(164.07 mg/L)。     

Triterpenoids from Calophyllum inophyllum and their growth inhibitory effects on human leukemia HL-60 cells

A new friedelane-type triterpene (1), along with seven known triterpenoids, was isolated from the stems and leaves of Calophyllum inophyllum Linn. Their structures were established as 3β, 23-epoxy-friedelan-28-oic acid (1), friedelin (2), epifriedelanol (3), canophyllal (4), canophyllol (5), canophyllic acid (6), 3-oxo-friedelan-28-oic acid (7), and oleanolic acid (8) by spectroscopic methods (NMR, EI-MS). The growth inhibitory effects of these triterpenoids on human leukemia HL-60 cells were determined.     

Antifungal limonoids from the fruits of Khaya senegalensis

Investigation of the fruits of Khaya senegalensis resulted in the isolation of three new mexicanolide limonoids containing a rare conjugated diene lactone system named seneganolide A (1), 2-hydroxyseneganolide A (2) and 2-acetoxyseneganolide A (3). Two known limonoids, 3-deacetyl-7-deacetoxy-7-oxokhivorin (4) and methyl 6-hydroxyangolensate (5), were also found. The structures of the new compounds were elucidated on the basis of spectral methods. The antifungal activity of compounds 1, 3 and 5 was tested against the fungus Botrytis cinerea.
