Cache Valley virus infection in Texas sheep flocks   总被引:1,自引:0,他引:1  
Cache Valley virus (CVV), an arbovirus indigenous to the United States, has been implicated as an important teratogenic agent in sheep. The prevalence and distribution of Texas sheep with CVV-specific antibody were investigated. In 1981, 19.1% of 366 sheep located in 22 counties of Texas had antibodies specific for CVV. Of 50 flocks examined in the major sheep-producing counties in Texas, 34 had sheep with antibodies that reacted with CVV, including all sheep tested in 6 flocks that were seropositive. Sera obtained from sheep at the Texas Agricultural Experiment Station at San Angelo between 1986 and 1989 were also examined for CVV-specific antibody because this flock was the subject of the episode of CVV-associated congenital malformations during the 1986 and 1987 lambing season. Approximately 8.6% of 104 sheep in 1986, 63.4% of 164 in 1987, 11.3% of 44 in 1988, and 71.9% of 89 in 1989 from the Texas Agricultural Experiment Station at San Angelo tested were seropositive. The data indicate that CVV infections in sheep were widespread in Texas in 1981 and that the virus is enzootic in sheep at the Texas Agricultural Experiment Station in San Angelo, where the episode of congenital malformations had initially been reported in 1987.  相似文献

Serologic evidence indicated that an episode of congenital abnormalities in sheep was caused by Cache Valley virus (CVV), a bunyavirus indigenous to the United States. To determine the teratogenic potential of CVV in sheep, fetuses were infected in utero between 27 and 54 days of gestation with an isolate (CK-102) obtained in 1987 from a sentinel sheep in San Angelo, Texas. The dams of these fetuses were euthanatized between 28 and 75 days after inoculation, and the fetuses were examined for malformations. Twenty-eight of 34 fetuses had congenital abnormalities, including arthrogryposis, hydranencephaly, mummification, reabsorption, and oligohydroamnion. Virus was isolated from the allantoic fluid of 11 of 17 fetuses euthanatized at less than 70 days of gestation. The virus-positive fetuses, which were all negative for CVV-neutralizing antibody, had lesions ranging from none to severe arthrogryposis and hydranencephaly. Virus was not recovered from the allantoic fluid of fetuses after 76 days' gestation when CVV-specific antibody could be detected in 5 of 8 fetuses examined. The 2 fetuses infected on days 50 and 54 of gestation appeared normal and 1 had antibody to CVV.  相似文献

An epizootic of abortions, weak lambs, stillbirths, and congenital arthrogryposis-hydranencephaly occurred in a sheep flock in West Texas. The outbreak began during the first week of January 1987 and continued through the third week of February 1987. Lambs born after February 1987 were not affected. A high incidence of antibodies to Cache Valley virus (Texas 7856 isolate) was demonstrated in the ewes' serum and in precolostral serum from affected lambs. No virus was isolated from tissues and body fluids of the affected lambs. The clinical, pathological, and immunological features of the epizootic were similar to those reported in Akabane virus infection in sheep. Although serological findings strongly suggest Cache Valley virus as the etiological agent of this outbreak, transmission studies are needed.  相似文献

Twenty-nine of 200 (14.5%) ewes on a farm in Cobleskill, NY aborted or had dead lambs during the lambing seasons of 1985 and 1986. Thirteen of 15 ewes that aborted had high Toxoplasma gondii antibody titers (1,024), via the modified agglutination test, and T gondii was isolated from the tissues of 1 fetus. In the 1987 lambing season, 5 ewes aborted, but not because of T gondii infection. Toxoplasma gondii antibodies were detected in 73.8% of sera obtained from 592 ewes in January 1987, indicating enzootic toxoplasmosis on this farm. Seropositivity increased with age; 40.2% of 1-year-old ewes had detectable antibody vs 89.2% of 2-year-old ewes.  相似文献

An outbreak of pseudorabies occurred in sheep housed with swine in the same building. Although the sheep and swine were not in physical contact, the lambs and ewes were exposed to air from the sows' section. Three dead lambs were submitted to the Iowa State University Veterinary Diagnostic Laboratory for necropsy. Grossly there were pulmonary congestion and multifocal pulmonary hemorrhages. Microscopic lesions were severe acute multifocal necrotizing bronchopneumonia with necrotizing vasculitis and intranuclear inclusion bodies within the neurons of the parabronchial ganglia. Bacterial cultures were negative for pathogenic agents; pseudorabies virus was isolated from ovine brain tissue. Viral antigen was demonstrated in the neurons of the parabronchial ganglia by immunoperoxidase staining. Electron microscopy revealed nucleocapsids in the parabronchial ganglionic neurons which contained basophilic intranuclear inclusion bodies. Viral DNA prepared from the ovine pseudorabies virus isolate was found by restriction endonuclease analysis to be related to the Indiana Funkhauser strain of pseudorabies virus.  相似文献

Background Several outbreaks of goitre, considered to be related to iodine deficiency, occurred in sheep flocks throughout Victoria in 2010. Objective We describe one outbreak in Merino–Border Leicester‐cross ewes and their lambs in north‐east Victoria that appeared to be associated with increased rainfall and pasture growth, particularly during the preceding summer and autumn. Results The outbreak was characterised by a four‐fold increase in neonatal lamb deaths and goitre, alopecia and poor skeletal development in the lambs. Most cases occurred in lambs born to 2‐year‐old crossbred ewes that had grazed long, lush perennial pastures throughout their entire pregnancy, whereas few cases occurred in mature crossbred or Merino ewes that had grazed shorter, annual pastures on hill country for 3 weeks in late pregnancy but were otherwise managed similarly. Conclusion Existing recommendations for south‐eastern Australia are that only spring‐lambing ewes in iodine‐deficient areas require iodine supplementation to prevent goitre in years with high autumn–winter rainfall. Aspects of this outbreak suggest that ewes lambing at other times of the year and grazing abundant pasture for prolonged periods may also require supplementation to prevent goitre, even if autumn–winter rainfall does not exceed previously established thresholds.  相似文献

Ovine fetuses, newborn lambs, and ovine colostrum were examined for ovine progressive pneumonia virus. The lambs and colostrum were also examined for specific antibody. Virus was isolated from 1 fetus, from 2 newborn lambs, and from most samples of colostrum. The fetus was about 100 days old and was carried by a seronegative ewe in contact with seropositive sheep. Both newborn lambs were carried by seropositive ewes. One lamb was dead at birth; the other lamb was normal and had not nursed. Antibody specific for the virus was present in the colostrum of 12 of 14 seropositive ewes and in the serum of 8 of 11 lambs that had nursed seropositive ewes, but not in the serum of lambs that had not nursed.  相似文献

: Sera from 1,448 adult ewes in 91 flocks, representing all 26 counties in the Republic of Ireland, were examined for pestivirus antibodies using a commercially available ELISA which detected IgG1 antibody to border disease virus. Eighty-one sheep (5.6%) in 42 flocks (46.0%) were antibody-positive. Within infected flocks, the mean seroprevalence level was 11.4% with a range of 6.3% to 30.0%. The highest antibody prevalence was detected in sheep from central lowland counties of Ireland. Comparative neutralisation testing of 42 ELISA-positive sera detected geometric mean antibody titres of 136 to the NADL strain of bovine viral diarrhoea virus (BVDV), 92 to the Moredun strain of border disease virus and 21 to the 137/4 strain of border disease virus. These results suggest that BVDV may be the major ruminant pestivirus infecting sheep in Ireland. Although there are high numbers of infected flocks, many sheep within such flocks remain antibody-negative and are at risk of giving birth to lambs with congenital border disease.  相似文献

A natural infection with border disease virus occurred in a flock on low ground in Argyll in the spring of 1984. The outbreak was unusual in that the typical clinical signs of border disease, ie, tremor and, or, fleece changes were not present; manifestations of disease were restricted to abortion and the birth of small weak lambs. The disease was shown to have been introduced to the flock by four healthy ewes persistently infected with border disease virus among a group of 39 purchased in October 1983. Further investigations in late August 1984 detected viraemia in six of seven ill-thriven lambs and four of 24 apparently healthy lambs. Attempted 'natural vaccination' of susceptible sheep by mixing them at grass for three months with groups of ewes and lambs known to contain virus excretors was largely unsuccessful as only four of 22 'sentinel' sheep seroconverted. In October 1984 the persistently infected purchased animals and all that year's lamb crop were removed from the farm. No disease occurred in 1985 when the lambing percentage was 129 per cent compared with 100 per cent in 1984. Two of the four persistently infected purchased ewes were mated at Moredun Research Institute in December 1984 and both produced healthy but persistently infected lambs.  相似文献

Thyroxine levels in serum of lambs and ewes were measured to determine their usefulness in assessing iodine nutrition of sheep. Lambs born on properties with no history of goitre had serum thyroxine concentrations more than twice that recorded in their mothers. These high values decreased to a level similar to that in the ewes after 8 weeks. In goitrous lambs younger than 2 weeks old, serum thyroxine concentrations were less than their ewe levels. Lambs born to ewes supplemented with iodine during pregnancy had higher thyroxine levels than lambs of control ewes.
A survey of 80 flocks of ewes in Victoria showed that 40% had a low mean thyroxine concentration (less than 50 nmol/l), but only one of these properties had reported an outbreak of goitre. It is suggested that serum thyroxine levels in newborn lambs may provide a more sensitive indicator of hypothyroidism associated with iodine deficiency than ewe levels.  相似文献

From 1985 to 1989 lambs persistently infected with border disease virus (BDV) were produced for comparative immunological studies by infecting 57 susceptible pregnant ewes between 50 and 60 days' gestation with Moredun or Oban strains of BDV. Ewes were infected either by injection with virus grown in cell culture or by housing with lambs excreting BDV. There was no significant difference in the outcomes of these different methods of infection. There was a significant difference in the number of viable lambs born to ewes receiving the two viruses. Of 41 ewes infected with Moredun virus 21 produced 32 live lambs of which 17 were reared to 1 month old (53% viability). Of 16 ewes receiving Oban virus 10 gave birth to 17 live lambs of which 15 were reared to 1 month old (88% viability). All the lambs born to ewes infected with Moredun BDV had varying signs of tremor and increased hairiness ("hairy-shakers") while those born to ewes infected with the Oban virus had no obvious clinical signs. Survival of the lambs was poor. Up until February 1991, 14 Moredun and 10 Oban sheep between the ages of 4 months and 5.5 yr had died from a variety of causes. The two commonest causes were a chronic wasting syndrome and a mucosal disease-like syndrome which was associated with the recovery of cytopathic BDV. Mating of unrelated persistently infected sheep was largely unproductive although 2 lambs were reared.  相似文献

An outbreak of nervous disease and abnormally hairy fleece was observed among newborn lambs in a closed flock of sheep in southern Ontario. The outbreak was investigated and found to closely parallel syndromes described in the British Isles and Australia and variously referred to as border disease, hypomyelinogenesis congenita and hairy shaker syndrome. Clinically affected lambs represented only a portion of the animals affected, considerable losses being caused by reproductive failure in the ewes and suboptimal performance in subclinically affected lambs. Sheep from the affected flock showed a strong serological response to the causative agent of bovine virus diarrhea/mucosal disease. The episode appeared to be chronologically related to the introduction of beef heifers from a sales barn. The paper discusses the results of clinico-epidemiological investigation of the outbreak in the light of current knowledge of border disease-like syndromes.  相似文献

An experiment was designed to investigate whether a condition in Australian sheep with clinical and pathological similarites to Border Disease was caused by the infection of the pregnant ewe with a Mucosal Disease virus (MDV). Forty ewes, at 58 to 63 days after mating, were inoculated with material from lambs in which all, some or none of the tissues examined contained MDV. The clinical condition was observed only in lambs born to ewes inoculated with MDV-positive material and then only to ewes in the group which had serological evidence of MDV infection. It is concluded that the Border Disease-like condition in Australian sheep is caused by the infection of the pregnant ewe with a Mucosal Disease virus.  相似文献

In a Swedish sheep flock comprising 202 ewes and 13 rams, a pair of twin lambs born in the spring of 1990 demonstrated signs of border disease (BD) and were persistently infected (PI) with border disease virus (BDV). Investigation showed that BDV had been introduced in the preceding tupping period 5-6 months earlier by a bought-in ewe which, on the basis of immunoperoxidase- and polymerase chain reaction techniques, was shown to be PI with BDV. Only 7 of the ewes, all of which had been in close contact with the PI ewe, seroconverted during the subsequent gestation. Apart from the PI twin lambs the losses caused by BDV were restricted to 2 barren ewes. The twin lambs, the PI ewe and lambs from the other 4 ewes that seroconverted were removed from the flock. The flock was thereafter free from an ongoing infection with BDV as shown by the absence of seroconversion.In addition, 5 heifers in late pregnancy most probably seroconverted to bovine virus diarrhoea virus (B VDV) when kept in close contact with the same PI ewe during the winter of 1989-90. When these heifers were reintroduced to the BVDV-free dairy herd from which they originated, their serum antibody titres ranged between 1:250 and 1:1250. Neither these heifers - nor their calves–caused any spread of the infection in the herd, as indicated by the absence of seroconversion in 70 cows. The present investigation shows that in the control of both BDV in sheep and BVDV in cattle, it is important to ensure that the risk of transmission of pestivirus between the 2 species is minimized.  相似文献

An outbreak of muscle disease affected approximately 20 of 600 ewes in spring 1987 in south-east Scotland. The clinical signs were a flaccid paralysis of the hind limbs and in severe cases collapse. Serum creatine kinase and aspartate aminotransferase activities were increased. Clinically affected sheep had a mean reciprocal serum antibody titre in a sarcocystis immunofluorescence antibody test of 557 whereas 22 sheep from the same flock, sampled one year earlier, showed a mean reciprocal titre of only 51. Histologically a heavy infestation of sarcocysts, myodegeneration and a non-suppurative myositis centred on degenerating sarcocysts were observed in a wide range of skeletal muscles and myocardium from four affected sheep. Monensin sodium had been inadvertently included in the protein pellet used in the feed for one week before the onset of the disease.  相似文献

An outbreak of bluetongue and the first isolation of the virus in the Sudan are reported. The disease occurred in sheep stressed by walking for five days when biting arthropods were prevalent. Estimates of the morbidity and mortality rates ranged from about 30 per cent and 2 per cent respectively in adult sheep to around 80 per cent and 100 per cent respectively in lambs. The virus was isolated by the inoculation of suckling mice and embryonated eggs with whole blood from febrile sheep. In a gel precipitation test it reacted with specific antiserum to type 10 BT8 strain. No other agent was isolated. Given the relatively mild nature of bluetongue in indigenous sheep, it is believed that the long walking stress coupled with exposure to sunlight might have aggravated the severity of the disease in this particular outbreak.  相似文献

A condition in Australian sheep resembling border disease was transmitted by the inoculation of pregnant ewes with material from affected lambs. This material contained mucosal disease virus (MDV). Twenty-two lambs comprising 6 from uninoculated control ewes, together with 11 with hairy coats and 5 with normal coats from inoculated ewes, were observed from 7 to 182 days after birth. Nine of the lambs from inoculated ewes died during the experiment from a variety of causes. Glial cell abnormalities were observed in control and affected lambs, but only 4 of the 11 hairy lambs were judged to have abnormal glial cells. There were no consistent histopathological findings indicative of MDV infection. MDV was recovered from tissues of all 11 hairy lambs, but not from any of the lambs with normal coats. The hairy lambs appeared to be immunologically tolerant to the virus. Susceptible sheep in contact with the hairy lambs were infected with MDV. It is suggested that a condition in Australian lambs characterised by hairiness of the birth coat and poor viability is due to foetal infection with a mucosal disease virus.  相似文献

A survey by mail in 1979 of 995 sheep farmers in Victoria enquiring as to the occurrence of hairy shaker disease (HSD) in lambs on their farms resulted in a 48% response. A total of 17 farms reported the presence of HSD. One hundred and eleven ewes produced lambs with HSD representing a small proportion only of the ewes (0.8%) on these farms and an even smaller percentage of the total ewes (0.03%) in the survey. Abortions were not prevalent on the majority of farms reporting HSD. There was no evidence that HSD was more common in any particular area in Victoria. As was anticipated mucosal disease virus/bovine virus diarrhoea (MDV) antibody was often present in serums from affected sheep. It was concluded that at present, the hairy shaker form of border disease did not appear to be an economically important problem in Victoria.  相似文献

Epizootic outbreaks of congenital malformations in sheep are rare and have, to the best of our knowledge, never been reported before in Europe. This paper describes relevant preliminary findings from the first epizootic outbreak of ovine congenital malformations in the Netherlands. Between 25 November and 20 December 2011, congenital malformations in newborn lambs on sheep farms throughout the country were reported to the Animal Health Service in Deventer. Subsequently, small ruminant veterinary specialists visited these farms and collected relevant information from farmers by means of questionnaires. The deformities varied from mild to severe, and ewes were reported to have given birth to both normal and deformed lambs; both male and female lambs were affected. Most of the affected lambs were delivered at term. Besides malformed and normal lambs, dummy lambs, unable to suckle, were born also on these farms. None of the ewes had shown clinical signs during gestation or at parturition. Dystocia was common, because of the lambs' deformities. Lambs were submitted for post-mortem examination, and samples of brain tissue were collected for virus detection. The main macroscopic findings included arthrogryposis, torticollis, scoliosis and kyphosis, brachygnathia inferior, and mild-to-marked hypoplasia of the cerebrum, cerebellum and spinal cord. Preliminary data from the first ten affected farms suggest that nutritional deficiencies, intoxication, and genetic factors are not likely to have caused the malformations. Preliminary diagnostic analyses of precolostral serum samples excluded border disease virus, bovine viral diarrhoea virus, and bluetongue virus. In December 2011, samples of brain tissue from 54 lambs were sent to the Central Veterinary Institute of Wageningen University Research, Lelystad. Real-time PCR detected the presence of a virus, provisionally named the Schmallenberg virus, in brain tissue from 22 of the 54 lambs, which originated from seven of eight farms that had submitted lambs for post-mortem examination. This Schmallenberg virus was first reported in Germany and seems to be related to the Shamonda, Aino, and Akabane viruses, all of which belong to the Simbu serogroup of the genus Orthobunyavirus of the family Bunyaviridae. These preliminary findings suggest that the Schmallenberg virus is the most likely cause of this epizootic of ovine congenital malformations, which is the first such outbreak reported in Europe.  相似文献
