This article studies the effect of a boar's birth season and breed on semen parameters and its further reproductive performance. Research material consisted of 72 boars from three breeds (24 Polish Large White PLW, 24 Polish Landrace PL, 24 Duroc × Pietrain D×P). During the whole period of the study, selected semen parameters were analysed: semen volume, spermatozoa concentration, total number of spermatozoa, total number of motile spermatozoa, number of insemination doses and also reproductive indicators: farrowing rate, total born litter size, total number of piglets born live and still, and average piglet weight. Boars born in the winter and summer months demonstrated the highest spermatozoa concentrations (383.25 and 392.37 × 106/ml), total number of spermatozoa (91.75 and 93.21 × 109), total number of motile spermatozoa (76.10 and 77.99 × 109) and number of insemination doses (24.53 and 24.89; p ≤0.01). Statistically lower values for these parameters were observed for boars born in the spring and especially in autumn (p ≤0.01). The significant impact of birth season on farrowing rate (p  0.05) and average piglet weight (p  0.05) was confirmed for PLW boars. For the PL breed, only the total number of piglets born live was proven to be significantly affected (p  0.05). No impact of birth season was shown on semen quality or reproductive performance for D×P boars. In our study, we showed that the birth season of a boar had a more impact on the level of semen parameters, and less on the reproductive performance indicators. The results indicated that both the quality of semen and reproductive performance varied in terms of the study factors, as well as between individual breeds of boars involved in the experiment.  相似文献

The study evaluated the reproductive performance of primiparous sows submitted to post‐cervical insemination (PCAI) compared with cervical artificial insemination (CAI). Difficulty with catheter introduction, the occurrence of bleeding or semen backflow during insemination, and volume and sperm cell backflow up to 60 min after insemination were also evaluated. Sows were homogenously distributed, according to body weight loss in lactation, lactation length, weaned piglets, weaning‐to‐oestrus interval and total born in previous farrowing, in two treatments: PCAI (n = 165) with 1.5 × 109 sperm cells in 45 ml (2.4 ± 0.04 doses per sow) and CAI (n = 165) with 3 × 109 sperm cells in 90 ml (2.5 ± 0.04 doses per sow). During PCAI, sows were inseminated in the absence of boars. Transabdominal real‐time ultrasonography was performed at oestrus onset, immediately before the first insemination and at 24 h after last insemination. There was no difference (P > 0.05) between treatments in farrowing rate (91.5% × 89.1%) and litter size (12.5 × 11.9 piglets born, respectively for PCAI and CAI sows). Successful passage of the intrauterine catheter in all the inseminations was possible in 86.8% (165/190) of sows initially allocated to PCAI treatment. Difficulty of introducing the catheter in at least one insemination did not affect the reproductive performance of PCAI sows (P > 0.05). Bleeding during insemination did not affect (P > 0.05) the farrowing rate in both treatments, but litter size was reduced in CAI and PCAI sows (P ≤ 0.06). Percentage of spermatozoa present in backflow within 1 h after insemination was greater in CAI than PCAI sows (P < 0.01). More than 85% of primiparous sows can be successfully post‐cervical inseminated with doses containing 1.5 × 109 sperm cells in the absence of the boar during insemination without impairing the reproductive performance.  相似文献

This study investigates the association of semen traits with boar fertility. The fertility outcome (farrowing rate – FR and total piglets born – TB) of 14 boars was obtained from a field trial conducted during 10 week of breeding period on a commercial farm using multiparous sows (n = 948) through single‐sire mating with 2 × 109 motile sperm cells per artificial insemination (AI) dose. Sperm motion parameters, evaluated with computer‐assisted semen analysis system in raw and stored semen at 17°C for 240 h, in addition to morphological sperm defects, measured on the collection day, were included in the analysis to determine which semen traits were important to discriminate the fertility potential of ejaculates from these boars. The data underwent multivariate cluster, canonical and discriminant analyses. Four clusters of boars were formed based on fertility outcome. One boar, with the lowest FR and TB values (89.7% and 11.98), and two boars, with the highest FR and TB values (97.8% and 14.16), were placed in different clusters. The other boars were separated in two distinct clusters (four and seven boars), including boars with intermediate TB (12.64 and 13.22) but divergent values for FR (95.9% vs 91.8%). Semen traits with higher discriminatory power included total motility, progressive motility, amplitude of lateral head displacement and cytoplasmatic droplets. Through multivariate discriminant analysis, more than 80% of the 140 ejaculates were correctly classified into their own group, showing that this analysis may be an efficient statistical tool to improve the discrimination of potential fertility of boars. Nevertheless, the validation of the relationship between fertility and semen traits using this statistical approach needs to be performed on a larger number of farms and with a greater number of boars.  相似文献

The present study aimed to evaluate differences in area, volume and testicular weight among groups related to breed and age of boar and establish associations between testicular parameters and seminal characteristics. Seminal quality has been studied using computer-assisted semen analysis (CASA) systems that reduce the subjectivity of kinematic parameters. Based on the data obtained through this system, sperm quality indexes and their derived parameters have been assessed to identify a better estimator for determining movement and velocity parameters. We also established indexes of velocity and sperm movement for boar semen. To this end, we evaluated 191 ejaculate samples from 63 boars. Differences related to the effects of season and age on quality boar semen were found (p ≤ .05) in the animal groups. Indexes were established for velocity and sperm movement based on CASA-Mot, and we proposed a sperm movement index (SMI) and sperm velocity index (SVI) with the CASA-Mot variables. Correlations were found (p < .05) among sperm indexes and CASA-Mot variables. Boar characteristics were found to be associated with some CASA-Mot variables, and the estimated SMI and SVI indexes predicted a greater variation in kinematic characteristics compared to the parameters evaluated separately using the CASA-Mot system.  相似文献

Motility is the most widely used indicator of sperm quality. Computer‐Assisted Semen Analysis (CASA) allows the objective evaluation of sperm motility parameters. CASA technology is a common tool to predict semen doses in farm animal reproduction. The kinds of video cameras used until now for image acquisition have presented limited frame rates (FR), which have a negative influence on the quality of the obtained data. The aim of the present work was to define the optimal frame rate for a correct evaluation of boar sperm motility and its subpopulation structure. Eighteen ejaculates from nine mature boars of the Pietrain breed were used. Using the ISAS®v1 CASA‐Mot system, with a video camera working up to 200 Hz, six FRs (25, 50, 75, 100, 150 and 200 fps) were compared. ISAS®D4C20 counting chambers, warmed to 37°C, were used. FR affected all the kinematic parameters, with curvilinear velocity (VCL) and BCF the most sensitive ones. All the parameters showed differences among animals. Non‐linear correlation showed the asymptotic level for VCL at 212 fps, being the highest FR for all the parameters. For future studies based just on progressive motility, almost 100 fps FR for 0.5 s must be used, while when kinematics must be considered, almost 212 fps for one‐second should be analysed. Three principal components were obtained (velocity, progressivity and oscillation), being similar at 50 and 200 fps. Cells were grouped in four subpopulations but with different kinematic and cellular distribution at both FRs.  相似文献

The objective of this study was to evaluate genotype by environment interactions for growth rate and carcass leanness in organic and conventional pig production environments. Organic breeding values for fattening traits were estimated for 37 Hampshire AI-boars based on slaughter records registered for 1805 crossbred offspring raised in an organic environment. The offspring were born and raised in herds certified for organic production. The statistical model included the fixed effects of sex, litter size at 2 weeks and herd. It also included the random effects of herd-year-season, birth litter and animal. Conventional breeding values for the same boars were captured from the breeding organization’s genetic evaluation. In the organic environment h2 was estimated to 0.30 and 0.37 for growth rate and carcass leanness, respectively (rg = − 0.11). Spearman rank correlations between organic and conventional breeding values, based on 29 boars with ≥ 20 progenies, were 0.48 for growth rate and 0.42 for carcass leanness. Both correlations were significantly different from 0 and 1. In conclusion, the results of the present study indicate weak genotype by environment interactions for both growth rate and carcass leanness in organic and conventional pig production environments, and there is some re-ranking of boars’ breeding values between environments.  相似文献

The fertilization capacity of sex-sorted sperms is seriously decreased, which inhibits its wide application. However, little information is still available about the effect of vitamin C (VC) and lycopene (Lyc) on the fertilization capacity of sex-sorted bull sperm. In this study, the washing medium and fertilization medium of sex-sorted sperm from three bull individuals were supplemented with different concentrations of VC (0, 1 × 10–3, 1 × 10–4, 1 × 10–5, 1 × 10–6 M) or Lyc (0, 1 × 10–4, 1 × 10–5, 1 × 10–6, 1 × 10–7). After washing twice and incubation for 1.5 hr, the malondialdehyde (MDA) level, phosphatidylserine (PS) translocation, membrane potential (Δψm) and IVF (in vitro fertilization) ability of sex-sorted sperm were investigated. For the sex-sorted sperm of bulls A, B and C, 1 × 10–3 M VC or 1 × 10–4 M Lyc treatment significantly decreased their MDA levels and PS translocation and increased their Δψm levels and cleavage rates after IVF. When blastocysts were concerned, 1 × 10–4 M Lyc significantly improved the blastocyst rates and their IFN-tau expression of bulls A and C. In conclusion, supplementation of 1 × 10–3 M VC or 1 × 10–4 M Lyc in washing and fertilization medium contributed greatly to improving the fertilization capacity of sex-sorted bull sperm during IVF procedure.  相似文献

Supplementation of exogenous growth hormone (GH) during prepuberty advances onset of spermatogenesis in boars, but the mechanism of action is unknown. The present study is an investigation of the presence and characteristics of testicular growth hormone receptors (GHR). A total of 36 boars were castrated, three boars every 10 days, between the ages of 10 and 120 days. Testicular membrane preparations of 10, 20, 30, 50, 70, 100 and 120‐day‐old boars were used to determine 125I‐bGH binding and Scatchard analysis. Liver from a 60‐kg barrow was used for comparison. Specific 125I‐bGH binding to testicular membrane preparations occurred in all age groups with the exception of 20‐day‐old boars at levels of 30–40% of liver binding. At 30 days of age the unlabelled bGH at 1.1 ng/tube achieved half maximal inhibition (ID50). Results of Scatchard analysis indicated a single class of binding sites. Binding affinity was 2.89 × 109m with a binding capacity of 12 fmole/mg membrane protein. The results from this study suggest that GH may act directly on the cells of the prepubertal boar testis.  相似文献

We investigated the effects of dietary supplementation with Bacillus subtilis PB6 (B. subtilis PB6) during late gestation and lactation on sow reproductive performance, antioxidant indices, and gut microbiota. A total of 32 healthy Landrace × Yorkshire sows on d 90 of gestation were randomly assigned to 2 groups, with 16 replicates per group, receiving basal diet (CON) or the basal diet + 0.2% B. subtilis PB6, containing 4.0 × 108 CFU/kg of feed (BS). The litter sizes (total born) and numbers of piglets born alive were larger in the BS group (P < 0.01), whereas the weights of piglets born alive and the piglet birth intervals were lower in the BS group (P < 0.05). Although the litter weights and piglet bodyweights (after cross-fostering) were lower after BS treatment (P < 0.05), the litter sizes, litter weights, lactation survival rate, and litter weight gains at weaning were higher in BS group (P < 0.05). The concentrations of malondialdehyde (MDA) in the sow sera at parturition were lower in the BS group (P < 0.01). The serum total antioxidant capacity (T-AOC) at parturition and the serum catalase (CAT) concentrations on d 21 of lactation were higher in the BS group (P < 0.05). Dietary supplementation with B. subtilis PB6 (P < 0.05) reduced the serum endotoxin concentrations in the sows and the serum cortisol concentrations of the piglets at d 14 of lactation. The α-diversity indices of microbial were higher in the CON group (P < 0.05). At the phylum level, B. subtilis PB6 supplementation increased the relative abundances of Gemmatimonadete and Acidobacteria (both P < 0.01) and reduced those of Proteobacteria, and Actinobacteria (both P < 0.05). At the genus level, B. subtilis PB6 supplementation increased the relative abundance of Ruminococcaceae_UCG-013 cc (P < 0.05) and reduced that of Streptococcus (P < 0.05). This study demonstrated that adding 4.0 × 108 CFU/kg B. subtilis PB6 to sows’ feed during late gestation and lactation could shorten piglet birth intervals, enhance the growth performance of suckling piglets, and improve the gut health of sows during late gestation.  相似文献

The aims were to evaluate sperm DNA fragmentation (SDF) in boars through the dispersion of their chromatin in raw semen samples, quantifying the extent of SDF, and to assess dynamic aspects of sperm DNA damage after incubation to obtain the rate of sperm DNA fragmentation (rSDF) under thermal conditions similar to the uterus (37°C) over a period of up to 24 hr and to correlate the reproductive outcome of the sows with the SDF of the boars at ejaculation. The study was performed on a pig‐breeding farm in southern Uruguay. Sixty‐one ejaculates from five of the most frequently used hybrid boars were evaluated. Semen was collected weekly from each of the boars, using the gloved‐hand technique and discarding the jelly‐like fraction of the ejaculate. Fresh semen was kept in a water bath at 37°C and protected from light, and was thereafter processed with Sperm‐Sus‐Halomax® to evaluate SDF. The smears for time 0 (T0) were made on farm, and thereafter smears were made at the laboratory at 4 hr of obtaining the semen (T4), then every 2 hr (T6, T8, T10, T12) and a final fixation at 24 hr (T24). Differences in SDF were observed among exposure times for all boars (p < .05), but not between T10 and T12 (p = .7751) nor T4 and T24 (p = .9113). In none of the T24 samples, sperm heads could be seen with chromatin dispersion halos. Furthermore, there were differences among boars when comparing sperm rSDF (p < .05). Farrowing rate was not affected by SDF at T0 (r = .38, p = .75), nor was litter size (r = .16, p = .70). With the present experimental conditions, we have not been able to show a relationship between sperm DNA fragmentation at ejaculation and reproductive performance. However, this could be a result of the low number of ejaculates and boars used.  相似文献

A pedigree including 1538 individuals of the endangered pig breed ‘Bunte Bentheimer’ and 3008 records of the fertility traits ‘number of piglets born alive’ (NBA) and ‘number of piglets weaned’ (NW) were used to i) characterize the population structure, ii) to estimate genetic (co)variance components and estimated breeding values (EBVs) and iii) to use EBVs for the application of the concept of optimal genetic contributions. The average coefficient of inbreeding increased from F = 0.103 to = 0.121 within the two recent cohorts. Average rate of inbreeding amounted to 1.66%, which resulted in an effective population size of Ne = 30 animals in the recent cohort. Average generation interval was 3.07 years considering the whole pedigree, and in total, only 612 sows and boars generated offspring. Estimated heritabilities for both traits NBA and NW were 0.12, and the estimated genetic correlation between both traits was 0.96. The variance component due to the service sire was higher than in commercial pig breeds, presumably due to the widespread use of natural service boars. The EBVs for NBA from 333 selection candidates (63 boars and 270 sows) were used to determine optimal genetic contributions. Based on selected animals and their optimal genetic contributions, specific mating designs were evaluated to minimize inbreeding in the next generation. Best results were achieved when using a simulated annealing algorithm and allowing artificial insemination.  相似文献

为探讨不同配种方式对二元母猪产仔数的影响,以杜洛克猪、长白猪、大白猪、皮特兰猪四个品种公猪精液采用人工授精的方法对11654头长大二元母猪配种,对所产11654窝总产仔数、窝产活仔数、窝损失等进行统计和分析,分析不同配种模式对二元母猪繁殖性能的影响。结果表明,配种次数、不同品种公猪组合配种对二元母猪总产仔数、产活仔数以及窝损失均无显著影响(P〉0.05),但杜洛克公猪所配二元母猪的总产仔数和窝产仔数均高于大白和长白猪(P〈0.05);采用长×杜×大组合配种的二元母猪总产仔数和窝产活仔数均高于其它三种组合。试验结果表明,在生产中,可在长白、大白、杜洛克猪中任选两个品种,通过二次配种的方式提高母猪的产仔数。  相似文献

The objective of this study was to develop a prediction equation for methane‐related traits in beef cattle and evaluate this equation using datasets with different cattle breeds and roughage rates. Enteric methane emission (CH4, l/day) was measured using open‐circuit respiration chambers. Dry matter intake (DMI, kg/day), body weight (BW, kg), daily gain (DG, kg), total digestible nutrients (TDN, %DMI), and roughage rate (Rrate, %) were used as independent variables, and methane‐related traits—CH4, CH4 per DMI (CH4/DMI, l/kg), and methane conversion factor (MCF, %)—were used as dependent variables. The best‐fit equations to predict methane‐related traits using a total of 76 records were CH4 = –676.7 + 0.04194 × BW + 29.88 × DMI + 7.883 × TDN + 4.367 × Rrate, CH4/DMI = –52.24 – 1.193 × 10–3 × BW – 5.905 × DG + 1.077 × TDN + 0.5008 × Rrate, and MCF = –11.43 – 5.308 × 10–4 × BW – 1.223 × DG + 0.2336 × TDN + 0.1157 × Rrate. The predictive ability of the developed equations differed between roughage rates but not between breeds. For CH4, the predictive ability of the developed equations was better compared with previously reported equations in the low roughage rate dataset, but not in the high roughage rate dataset. Our results suggest that the developed equations of methane‐related traits can be applied in beef cattle fed with low roughage diets.  相似文献

Nine Belgian Landrace (BL) boars were used to produce crossbred BL × Dutch Yorkshire (BL × DY) boars to be compared with DY boars as terminal sires. BL × DY boars had 0.5 mm more side fat at 94 kg than DY boars (P < 0.001); growth rate did not differ between both groups.Claw development of BL × DY boars was considered significantly better (P < 0.001) than that of DY boars at 6–8 months of age, causing overall leg quality appraisal to be significantly better (P < 0.025).Young BL × DY boars showed more and earlier sexual activity, whereas only 4.0% of these boars had to be culled due to reproductive failure, relative to 10.0% of DY boars (P < 0.005).Analysis of semen of the 9 BL AI boars, and of 19 BL × DY and 19 DY AI boars indicated weakly significant breed differences (P < 0.1) for the number of spermatozoa per ejaculate and no significant breed differences (P > 0.1) for the number of live spermatozoa per ejaculate.For both traits, BL × DY boars showed the smallest numbers of cells and BL boars the largest.Inseminations with semen from BL × DY boars (doses being standardized for the number of cells) were succesful more often (19.0%) than with DY semen (88.3%) (P < 0.001); in addition, the litters contained more piglets at birth (11.43 vs. 10.97 piglets) (P < 0.001).End products sired by BL × DY boars contained 0.305 kg less trimmed ham + loin in an 80-kg carcass than progeny of DY boars (P < 0.1); other fattening and slaughter traits did not differ significantly.  相似文献

One hundred thirty Yorkshire-Landrace (YL), Chester White-Landrace (CL) and Chester White-Yorkshire (CY) F1 crossbred sows were mated randomly to Duroc (D), Hampshire (H), Farmers Hybrid 414 (FH414) or Farmers Hybrid 929 (FH929) boars. These matings produced 321 litters and 3,379 pigs, which were used to determine the effect of sire breeding and dam breeding on preweaning and postweaning performance of the pigs and litters. Litter size born alive, litter size at 56 d, litter weight at 56 d and litter weight per day of age to 100 kg were lower in litters sired by H boars than in those sired by D boars. However, sizes and weights of litters sired by D and H boars were not different from those sired by FH414 and FH929 boars. Litter sizes at birth were larger for litters out of CL and CY sows than those out of YL sows, but significant differences were not found for litter sizes at 21 d, 56 d or at marketing. Pigs sired by H boars were heavier at birth than pigs sired by D boars. Pigs sired by H boars did not differ significantly in weights at 21 and 56 d or in postweaning daily gains or days to 100 kg from pigs sired by D boars. Pigs out of CL dams were heavier at birth, 21 d and 56 d of age than pigs out of CY dams, but did not differ significantly in postweaning gains or in days to 100 kg. Pigs sired by D and H boars had less ultrasonic fat than pigs sired by FH414 and FH929 boars. Breed and sire differences in sperm cryosurvival have been noted, with negative implications for sperm cryobanking and assisted reproduction programmes. This study hypothesized that these differences could be modified by using lower molecular weight cryoprotectants. Therefore, the effect of replacing glycerol (GLY) with ethylene glycol (EG) on differential cryosurvival of semen from two Sanga cattle breeds (Mashona vs. Tuli) was determined. Three to five ejaculates were collected from each of ten bulls (3-8 years) by electro-ejaculation, diluted in three Tris-egg yolk extenders (Triladyl®, 7% GLY-based and 7% EG-based) and evaluated for sperm motility, viability and morphology at three time periods (fresh – 0 hr, pre-freeze – 4 hr and post-thaw). Tuli bulls produced larger (11.8 ± 0.31 ml vs. 8.5 ± 0.38 ml) and more concentrated ejaculates of lower fresh semen quality. Breeds differed across time for motility and morphology, but not viability. Mashona bull semen had significantly higher motility and normal morphology values at each sampling time. Bulls classified as poor freezers had lower concentration (0.70 ± 0.09 × 109 sperm/ml vs. 1.37 ± 0.10 × 109 sperm/ml), sperm motility index (SMI, 35.0 ± 3.4 % vs. 67.8 ± 2.1 %) and viability (69.7 ± 1.1 % vs. 75.7 ± 1.0 %) compared to good freezers. Maintenance of semen quality by GLY and EG did not differ between breeds, poor and good freezers, or age groups. The interaction breed by extender across time did not reach statistical significance for all variables. The study revealed that bull and breed variation in sperm quality and cryosurvival is not modified by replacing GLY with EG, suggesting that cryostress tolerance of sperm may be under control of mechanisms other than differential response to GLY cytotoxicity.  相似文献

The aim of this study was to evaluate fertility and sex ratios after artificial insemination in dogs under field conditions. Semen was cryopreserved as unsorted (control) or was separated into X‐ and Y‐chromosome‐bearing sperm using a cell sorter. Sixty female dogs were inseminated with frozen–thawed spermatozoa of 100 × 106 unsorted (a dose in practice) and 4 × 106 sorted (X and Y group, respectively). A total of 20 dogs became pregnant and 126 puppies were born from the three groups. The percentage of parturition was similar for the X (5/20; 25.0%) and Y (4/20; 20.0%) group (P > 0.05), but lower than controls (11/20; 55.0%) (P < 0.05). Ultimately 28 out of the 32 puppies produced from X group were female (87.5%) and 19/22 (86.4%) puppies of Y group were male. In contrast, sex ratio (51.4% to 48.6%) in the control was significantly different from the X, Y group (P < 0.05). However, male and female puppies in the control had similar birth weights and weaning weights to those from the X and Y groups. This preliminary information indicated that normal puppies of predicted sex can be produced with low numbers of sorted cryopreserved dog spermatozoa at a farm level, making sperm‐sexing technology potentially applicable for elite breeding units.  相似文献

Surface litter protects rangeland soils against wind and water erosion and provides food and nesting materials for wildlife and insects. However, the ability of grassland systems to provide these services depends on the little studied topic of seasonal surface litter decomposition. Seasonal and annual surface litter decomposition rates were determined between 2014 and 2015 in central and western South Dakota at three mixed-grass prairie locations. Residue bags containing surface litter were placed in the field in late fall (1 November) of 2014 and removed after the winter (1 April), spring (1 July), and summer + fall seasons (1 November) of 2015. The litter was analyzed for total C, total N, acid detergent fiber (ADF), and acid detergent lignin (ADL). Average winter temperatures ranged from −5oC to −15oC, while summer temperatures ranged from 10oC to 35oC. Litter decomposition was lowest during the winter (0.57−0.86 g [kg × day]−1) and greatest during the summer + fall (2.12−2.69 g [kg × day]−1). Over the entire season, 40.8−62% of the surface litter decomposed. Winter litter decomposition was positively correlated with air temperature (r = 0.62, P < 0.01) and snow depth (r = 0.61, P < 0.01), and negatively correlated with C/N ratio (r = −0.65, P < 0.01), ADF (r = −0.35, P < 0.05), and ADL (r = −0.25, P < 0.05) concentrations. These findings indicate that winter decomposition cannot be ignored and that winter surface litter decomposition increases with snow depth.  相似文献

Ketorolac (KET) is a nonsteroidal anti‐inflammatory drug approved for the use in humans that possesses a potent analgesic activity, comparable to morphine, and could represent a useful tool to control acute pain also in animals. The clinical efficacy and pharmacokinetic profile of intravenous (IV) ketorolac tromethamine (0.5 mg/kg) were studied in 15 dogs undergoing gonadectomy. Intra‐operative cardiorespiratory variables were monitored, and post‐operative pain was assessed using a subjective pain score (0–24) in all dogs, whereas the pharmacokinetic profile of the drug was determined in 10 animals. During surgery, mean minimal alveolar concentration of isoflurane was 1.69 ± 0.11%, and normocapnia and spontaneous ventilation were maintained in all animals. During pain assessment, no significant differences between males and females were found, and in no case rescue analgesia was necessary. No adverse effects were reported. Serum samples were purified by solid‐phase extraction and analysed by HPLC with UV‐Vis detection. A large variability was observed in serum concentrations. The kinetics of ketorolac was described by a noncompartmental analysis. The elimination half‐life (t½λz) and ClB were 10.95 ± 7.06 h and 92.66 ± 84.49 mL/h/kg, respectively, and Vdss and Vz were 1030.09 ± 620.50 mL/kg and 1512.25 ± 799.13 mL/kg, respectively. AUC(0→last) and MRT(0→last) were 6.08 ± 3.28 h × μg/mL and 5.59 ± 2.12 h, respectively. The results indicate that ketorolac possess good post‐operative analgesic effects until about 6 h after administration in dogs undergoing moderately painful surgery.  相似文献
