Globally, bread wheat production is threatened by fungal diseases, including the devastating disease Septoria tritici blotch (STB). Given the global importance of STB, and the difficulty in identifying novel sources of resistance to this disease, we screened a variety of wheat genotypes, including wild, ancestral, and mutagenized lines, for their STB response. This delineated a panel of wild wheat relatives and Watkins collection lines with exceptional resistance to a range of Zymoseptoria tritici isolates, some of which are highly virulent on modern, elite wheat varieties. Additionally, we characterized the STB susceptibility of 500 lines of the wheat cultivar Cadenza TILLING population and developed backcross derivatives of two TILLING lines that show dominant partial resistance to STB. These backcross lines are partially resistant to multiple isolates of Z. tritici, and, with the wild and ancestral lines identified, provide a useful reservoir of STB-resistant germplasm for use in wheat breeding programmes.  相似文献

Septoria tritici blotch (STB) caused by the fungal pathogen Zymoseptoria tritici continues to be the most economically destructive disease of winter wheat throughout Ireland. Due to the widespread development of fungicide resistance in the Irish Z. tritici population, integrated strategies to control STB are increasingly necessary. A key component of such strategies will be the deployment of winter wheat cultivars with improved levels of STB resistance. Unfortunately, due to the nature of Z. tritici, such resistances are at risk of being overcome by the pathogen. In late summer 2020, foci of STB were observed across a range of winter wheat cultivars under evaluation for recommendation in Ireland. Common amongst these was the cultivar Cougar in each of their pedigree. To determine if the foci observed in 2020 resulted from strains virulent to Cougar, isolate collections were established and virulence screens conducted on Cougar and a range of the cultivars currently under evaluation. These confirmed the presence of Cougar-virulent strains in the Irish Z. tritici population, and that this virulence affects not just Cougar, but also cultivars derived from it. Although the foci observed in 2020 were in both fungicide-untreated and -treated plots, there was no evidence that these strains are more sensitive or resistant to fungicides compared to the wider Irish Z. tritici population, with moderate resistance to the SDHIs and azoles dominating. Combined, the present study confirms the need to ensure a diversity of control measures for STB, including ensuring a range of STB resistances are used.  相似文献

Zymoseptoria tritici, the causal agent of septoria tritici blotch (STB), remains a significant threat to European wheat production with the continuous emergence of fungicide resistance in Z. tritici strains eroding the economic sustainability of wheat production systems. The life cycle of Z. tritici is characterized by a presymptomatic phase (latent period, LP) after which the pathogen switches to an aggressive necrotrophic stage, when lesions bearing pycnidia quickly manifest on the leaf. As minimal knowledge of the possible role of the LP in supporting STB resistance/susceptibility exists, the goal of this study was to investigate the spatial and temporal association between the LP and disease progression across three locations (Ireland – Waterford, Carlow; UK – Norwich) that represent commercially high, medium and low STB pressure environments. Completed over two seasons (2013–2015) with commercially grown cultivars, the potential of the LP in stalling STB epidemics was significant as identified with cv. Stigg, whose high level of partial resistance was characterized by a lengthened LP (c. 36 days) under the high disease pressure environment of Waterford. However, once the LP concluded it was followed by a rate of disease progression in cv. Stigg that was comparable to that observed in the more susceptible commercial varieties. Complementary analysis, via logistic modelling of intensive disease assessments made at Carlow and Waterford in 2015, further highlighted the value of a lengthened LP in supporting strong partial resistance against STB disease of wheat.  相似文献

Infection efficiency is a key epidemiological parameter that determines the proportion of pathogen spores able to infect and cause lesions once they have landed on a susceptible plant tissue. In this study, an improved method to measure infection efficiency of Zymoseptoria tritici using a replicated greenhouse experiment is presented. Zymoseptoria tritici is a fungal pathogen that infects wheat leaves and causes septoria tritici blotch (STB), a major disease of wheat worldwide. A novel experimental setup was devised, where living wheat leaves were attached to metal plates, allowing for time-resolved imaging of disease progress in planta. Because lesions were continuously appearing, expanding and merging during the period of up to 3 weeks, daily measurements were necessary for accurate counting of lesions. Reference membranes were also used to characterize the density and spatial distribution of spores inoculated onto leaf surfaces. In this way, the relationship between the number of lesions and the number of viable spores deposited on the leaves was captured and an infection efficiency of about 4% was estimated from the slope of this relationship. This study provides a proof of principle for accurate and reliable measurement of infection efficiency of Z. tritici. The method opens opportunities for determining the genetic basis of the component of quantitative resistance that suppresses infection efficiency. This knowledge would improve breeding for quantitative resistance against STB, a control measure considered more durable than deployment of major resistance genes.  相似文献

Demethylation inhibitor (DMI) and succinate dehydrogenase inhibitor (SDHI) fungicides are currently relied upon for the control of septoria tritici blotch (STB) in European wheat fields. However, multiple mutations have occurred over time in the genes encoding the targeted proteins that have led to a practical loss of fungicide efficacies. Among the different amino acid substitutions in Zymoseptoria tritici associated with resistance to these fungicides, S524T in CYP51 (DMI target) and H152R in SdhC (SDHI target) are regarded as conferring the highest resistance factors to DMI and SDHI, respectively. To facilitate further studies on the monitoring and selection of these substitutions in Z. tritici populations, a multiplex allele-specific quantitative PCR (qPCR) assay allowing for estimation of both allele frequencies in bulk DNA matrices was developed. The assay was then used on complex DNA samples originating from a spore trap network set up in Belgium, Denmark, Sweden, and Ireland in 2017 and 2018, as well as on leaf samples with symptoms. The S524T allele was present in all field samples and its proportion was significantly higher in Ireland than in Belgium, whereas the proportion of H152R was only sporadically present in both countries. The frequency of S524T varied greatly in the airborne inoculum of all four countries; however, the H152R allele was never detected in the airborne inoculum. The method developed in this study can be readily adopted by other laboratories and used for multiple applications including resistance monitoring in field populations of Z. tritici.  相似文献

Zymoseptoria tritici, the causal agent of septoria tritici blotch, a serious foliar disease of wheat, is a necrotrophic pathogen that undergoes a long latent period. Emergence of insensitivity to fungicides, and pesticide reduction policies, mean there is a pressing need to understand septoria and control it through greater varietal resistance. Stb6 and Stb15, the most common qualitative resistance genes in modern wheat cultivars, determine specific resistance to avirulent fungal genotypes following a gene‐for‐gene relationship. This study investigated compatible and incompatible interactions of wheat with Z. tritici using eight combinations of cultivars and isolates, with the aim of identifying molecular responses that could be used as markers for disease resistance during the early, symptomless phase of colonization. The accumulation of TaMPK3 was estimated using western blotting, and the expression of genes implicated in gene‐for‐gene interactions of plants with a wide range of other pathogens was measured by qRT‐PCR during the presymptomatic stages of infection. Production of TaMPK3 and expression of most of the genes responded to inoculation with Z. tritici but varied considerably between experimental replicates. However, there was no significant difference between compatible and incompatible interactions in any of the responses tested. These results demonstrate that the molecular biology of the gene‐for‐gene interaction between wheat and Zymoseptoria is unlike that in many other plant diseases, indicate that environmental conditions may strongly influence early responses of wheat to infection by Z. tritici, and emphasize the importance of including both compatible and incompatible interactions when investigating the biology of this complex pathosystem.  相似文献

This study provides new insights into the role of photoprotection in preformed and induced defence of two wheat genotypes with contrasting phenotypes to infection by Zymoseptoria tritici. We investigated the mechanisms of the photoprotective response during early infection, including nonphotochemical quenching (NPQ), β-carotene-derived xanthophylls, reactive oxygen species, and the phytohormones abscisic acid (ABA), jasmonic acid (JA), and salicylic acid (SA). Furthermore, we quantified the effects of pathogenesis on photosynthesis, stomatal control, and expression of plant defence molecular markers. The photoprotective mechanism of successful defence involved the qI component of NPQ leading to rapid down-regulation of photosystem II quantum yield and chlorophyll a:b, increased biosynthesis of the xanthophyll neoxanthin and ABA, and the expression of chloroplast-specific enzymes to engage in scavenging of O2●−. Elevated ABA in the resistant genotype correlated with preformed leaf defence traits including low stomatal density, increased expression of wax biosynthesis, and lignification. Z. tritici exhibited reduced germination and branching on the resistant host genotype and hijacked stomatal control in both genotypes by enhancing stomatal sensitivity to light. Increased biosynthesis of JA and anthocyanins, in contrast to SA, were quantified in the incompatible interaction. Our results indicate that ABA and JA in antagonistic action to SA were associated with defence in the resistant genotype, Cougar, against Z. tritici.  相似文献

Stem rust caused by Puccinia graminis f. sp. tritici is one of the most devastating diseases of wheat. Breakdown of host resistance under field conditions triggered by the evolution of new pathogenic races and pathotypes is a perennial threat for wheat cultivation. Rice, often grown in a rice–wheat cropping system, is immune to rust infection. Our microscopic studies revealed that P. graminis f. sp. tritici, although displaying nearly identical uredospore germination, stomatal entry, and epi- and endophytic mycelial growth in rice and wheat, failed to sporulate to cause rust disease in rice. We identified 18 key defence signalling genes in rice and unravelled their elicitation dynamics in time-course studies during infection. ICS1, NPR1-3, PRs, EDS1, PAD4, FMO1 (salicylic acid [SA] signalling), and ethylene-related genes (ACO4 and ACS6) were strongly elicited in rice. However, genes from the jasmonic acid (JA) signalling pathway (LOX2, AOS2, MYC2, PDF2.2, JAZ8, JAZ10) showed a delayed response during colonization in rice compared to an early or no induction in wheat. However, the JA/ethylene marker gene PDF2.2 was strongly induced in wheat as early as 12 hr postinoculation. Furthermore, rice and wheat displayed specific profiles of accumulation of various phenolic acids during P. graminis f. sp. tritici 40A infection. We propose a model where a differential modulation of the SA/JA-dependent defence network may modulate nonhost resistance. A deeper understanding of the molecular mechanism governing differential elicitation of defence signalling may provide a novel resistance mechanism for the sustainable management of rust diseases.  相似文献

Septoria tritici blotch (STB) disease of wheat is caused by the fungal pathogen Zymoseptoria tritici. It is the most important foliar disease of wheat in western Europe and affects wheat cultivation worldwide. The combination of intensive fungicide usage, a polycyclic asexual life cycle and an active sexual cycle has led to the emergence of fungal strains resistant/tolerant to all the major classes of fungicides used in its control. The hallmark of this disease is a long, symptomless latent phase that precedes the onset of visible symptoms. Understanding the processes that occur during the symptomless phase of infection is paramount in developing alternative strategies for disease control; however, large gaps in our knowledge of the disease remain. The known unknowns of the latent stage of infection can be summarized in three questions. Does the fungus initiate or manipulate host defences to trigger programmed cell death in order to facilitate nutrient acquisition or is the host acting exclusively? Does the fungus feed during both the latent phase and the necrotrophic phase like a true hemibiotroph? Does the long latent phase serve a beneficial function for the fungus or is it simply an artefact of evolution? This review aims to distil observations made during studies that have directly or indirectly contributed to answering these questions and points towards their most likely answers.  相似文献

Tan spot caused by Pyrenophora tritici‐repentis is a disease present in all wheat‐producing countries and silicon (Si) treatment of wheat plants has been shown to increase plant resistance to tan spot. In this study, the effect of phenylpropanoid metabolism on resistance to tan spot was evaluated and some phenolic compounds that accumulated in response to Ptritici‐repentis attack were identified. Furthermore, the effect of Si on phenylalanine ammonia‐lyase (PAL) activity and phenolic compound accumulation were determined in situ. Antifungal activity of differentially accumulated phenolic compounds was also evaluated in in vitro tests. Results showed that the increase in concentration of phenolic compounds was greatest at the onset of infection, and that some compounds showed fungitoxic effects including fungal tip swelling, granulation of germ tube and hyphae, and hyphal hyperbranching. Silicon‐induced reduction in both lesion size and tan spot disease progression were associated with activation of phenylpropanoid metabolism. PAL activity and accumulation of antifungal phenolic compounds were greater in pathogen‐inoculated plants supplied with Si. In these plants, fluorescence indicative of accumulation of phenolic compounds occurred early in epidermal cells and its intensity increased during the evaluation period, showing higher numbers of fluorescent cells around infected cells. Thus, the combined responses of cell fluorescence at sites of infection, increased PAL activity and accumulation of phenols indicate that Si strengthened wheat defence responses to infection by Ptritici‐repentis, reducing the severity of tan spot.  相似文献

Septoria tritici blotch (STB), caused by Mycosphaerella graminicola has recently become one of the devastating diseases in Iran causing significant yield losses on most commercial bread and durum wheat cultivars. Iran is located in the Fertile Crescent, a region where wheat was historically domesticated; and, thus, landraces derived from this region are of considerable global interest for identification of new sources of resistance to various stresses. Here, we report on the resistance responses of 45 tetraploid wheat landraces collected from different provinces of Iran to eight M. graminicola isolates. In total 138 isolate-specific resistances were found among all interactions (n = 360). The highest number of specific resistances (30 out of 32 interactions) was found in wheat landraces collected from West Azarbaijan. In contrast, all landraces from Kordestan were highly susceptible to M. graminicola isolates and only one isolate-specific resistance was identified among 106 isolate-wheat interactions. Kermanshah landraces showed the highest resistance variation against different isolates. About 57 isolate-specific resistances were identified among 104 interactions. Ilam landraces were highly resistant to STB as 28 specific resistances were observed among 32 interactions. Markazi (n = 2) and Sistan-Baluchestan (n = 1) were susceptible to all isolates tested. Landraces from Lorestan were generally susceptible to isolates tested as 26 susceptible responses were observed out of 32 interactions. Our results indicate that landraces collected from the Fertile Crescent region may possess diverse effective resistance genes or valuable broad spectrum resistance genes, and that their identification is of interest and can be exploited in breeding programs.  相似文献

Zymoseptoria tritici is the causal agent of septoria tritici blotch (STB), a foliar wheat disease important worldwide. Succinate dehydrogenase inhibitors (SDHIs) have been used in cereals for effective control of STB for several years, but resistance towards SDHIs has been reported in several phytopathogenic fungi. Resistance mechanisms are target‐site mutations in the genes coding for subunits B, C and D of the succinate dehydrogenase (SDH) enzyme. Previous monitoring data in Europe indicated the presence of single isolates of Z. tritici with reduced SDHI sensitivity. These isolates carried mutations leading to amino acid exchanges: C‐T79N, C‐W80S in 2012; C‐N86S in 2013; B‐N225T and C‐T79N in 2014; and C‐V166M, B‐T268I, C‐N86S, C‐T79N and C‐H152R in 2015. The current study provides results from microtitre and greenhouse experiments to give an insight into the impact of different mutations in field isolates on various SDHIs. In microtitre tests, the highest EC50 values for all tested SDHIs were obtained with mutants carrying C‐H152R. Curative greenhouse tests with various SDHIs confirmed the findings of microtitre tests that isolates with C‐H152R are, in general, controlled with lower efficacy than isolates carrying B‐T268I, C‐T79N and C‐N86S. SDHI‐resistant isolates of Z. tritici found in the field were shown to have cross‐resistance towards all SDHIs tested. So far, SDHI‐resistant isolates of Z. tritici have been found in low frequencies in Europe. Therefore, FRAC recommendations for resistance management in cereals, including a limited number of applications, alternation and combination with other MOAs, should be followed to prolong SDHI field efficacy.  相似文献

Twenty-four wheat cultivars and breeding lines were screened for isolate-specific resistance to septoria tritici blotch (STB) caused by 12 isolates of Mycosphaerella graminicola. New isolate-specific resistances that could be used in wheat breeding were identified. Major sources of resistance to STB used in world breeding programmes for decades, such as Kavkaz-K4500, Veranopolis, Catbird and TE9111, have several isolate-specific resistances. This suggests that 'pyramiding' several resistance genes in one cultivar may be an effective and durable strategy for breeding for resistance to STB in wheat. Several cultivars, including Arina, Milan and Senat, had high levels of partial resistance to most isolates tested as well as isolate-specific resistances. Resistance to isolate IPO323 was common, present in all but one of the major sources of resistance tested. This suggests that resistance to IPO323 may be an indicator of varietal resistance to STB in the field.  相似文献

The wheat stem rust fungus, Puccinia graminis f. sp. tritici (Pgt), race TTKSK and related races pose a serious threat to world wheat production. Knowing the effectiveness of wheat stem rust resistance (Sr) genes against Pgt race TTKSK is fundamental in mitigating this threat through resistance breeding. Sr15 was previously identified as being ineffective against Pgt race TTKSK. Here, multirace disease phenotyping data, linkage analyses, allelism testing and haplotype analyses are presented to support the conclusion that Sr15 is effective against Pgt race TTKSK. Resistance to race TTKSK was mapped to Sr15 in a biparental population. Thirty-two accessions with Sr15 displayed seedling resistance phenotypes against race TTKSK. However, these accessions were susceptible as seedlings at high temperatures (22–25 °C), consistent with previous reports that the interaction between avirulent Pgt isolates and Sr15 is temperature-sensitive. Markers STS638, wri4 and KASP_IWB30995 were found to predict the presence of Sr15, suggesting the utility of these assays for marker-assisted selection in breeding programmes. The effectiveness of Sr15 to specific Pgt races and temperatures makes it a less-desirable TTKSK-effective gene. Wheat lines assayed as resistant to race TTKSK at the seedling stage may possess Sr15 and breeders should be aware of the limitations of Sr15 for conferring stem rust resistance.  相似文献

Known and unknown genes conferring seedling and adult plant resistance (APR) to leaf rust, stem rust and stripe rust were detected either singly or in combination in a set of 136 African wheat genotypes using multi-pathotype tests with characterized Australian Puccinia triticina (Pt), P. graminis f. sp. tritici (Pgt) and P. striiformis f. sp. tritici (Pst) pathotypes. Lines Beladi 132, IYN 68/9.44, Kenya Kifaru and Kenya Mbweha were postulated to carry resistance against multiple pathotypes of Pt, Pgt and Pst, whereas IAR/W/163-3, Grano Di Moggio Tipo 44 and Trigo 48 had resistance against all pathotypes tested in the current study. Field evaluation with the three rust pathogens detected low to high APR in more than 50% of lines, and while most tested positive with markers linked to known APR genes (csLV34, csLV46G22, TM10KASPAR, csGS, Cfb5006 and csSr2), many carried unidentified and useful resistance to all three rusts. Genetic analysis of F3 mapping populations based on seven genotypes showed either monogenic or digenic inheritance of APR to leaf rust, stem rust and stripe rust. The lines postulated to carry effective uncharacterized seedling genes and APR genes are of great potential value in diversifying resistance to help achieve durable control of all three rust diseases of wheat.  相似文献

我国地方品种是小麦白粉病抗性的重要来源之一,为了对地方品种抗源的利用奠定基础,采用常规杂交方法,以感病品种Chancellor分别与我国小麦抗病地方品种蚂蚱麦、小白冬麦、游白兰、红卷芒进行正交和反交,获得F1、F2代。根据白粉菌菌株的毒谱选用E09菌株对Chancellor与小白冬麦、游白兰、红卷芒的杂交后代进行苗期抗性鉴定和统计分析,选用E30菌株对Chancellor与蚂蚱麦的杂交后代进行苗期抗性鉴定和统计分析。结果表明4个品种在正、反交情况下均表现出由一对隐性基因控制的抗性,说明这4个地方品种属于核遗传,其抗性是由一对隐性基因控制的。  相似文献

为明确中国不同春麦区小麦地方种质对当前小麦生产上流行的条锈病菌Puccinia striiformis f.sp.tritic的抗性水平及其所含抗性基因,利用条锈病菌生理小种条中32(CYR32)和条中34(CYR34)及混合生理小种(致病类群)对来自5个春麦区的196份小麦地方种质进行苗期、成株期抗性鉴定,并通过6个已知条锈病抗性基因Yr9、Yr18、Yr26、Yr48、Yr65Yr67对其所含重要抗性基因进行分子标记检测。结果显示,在苗期,有11份小麦地方种质对CYR32表现出抗性,有12份对CYR34表现出抗性,分别占供试种质总数的5.61%和6.12%;有6份对CYR32和CYR34均表现出抗性;在成株期,有59份小麦地方种质在5个田间诱导环境下表现出稳定的抗性。有119份小麦地方种质检测到含抗性基因,其中有3份携带Yr9,有50份携带Yr18,有43份携带Yr48,有54份携带Yr65,所有供试种质均未检测到Yr26Yr67,抗性基因的组合分析发现,共有31份小麦地方种质携带4种抗性基因组合类型Yr9+Yr18、Yr18+Yr48、Yr18+Yr65Yr48+Yr65。表明来自中国5个春麦区的小麦地方种质条锈病抗性表型呈多样性,且携带目前在小麦抗病育种和生产上有效的条锈病抗性基因(组合),建议加大对小麦地方种质的保护和应用力度。  相似文献
