摘    要:【目的】明确SNP标记位点与罗氏沼虾生长性状间的关联性,为罗氏沼虾生长性状候选基因的寻找、生长性状调控机理及后期分子标记辅助育种研究打下基础。【方法】采用直接测序技术对25个SNP标记位点在罗氏沼虾生长性状极端群体中进行多态性检测。采用卡方检验(χ2)筛选出2个极端(体长极大和极小)罗氏沼虾群体中与生长状况存在潜在相关性的SNP标记位点,进一步对罗氏沼虾浙江群体60个个体进行SNP基因型与生长性状(体长、头胸甲宽、第一腹节宽、第一腹节高和体重)的关联分析。运用一般线性模型对SNP标记位点与罗氏沼虾5个生长性状的关联性进行检测。【结果】卡方检验结果显示,SNP6、SNP7、SNP16、SNP20和SNP24等5个标记位点在罗氏沼虾极端群体中与生长性状存在潜在相关。5个潜在相关SNP标记位点与生长性状的相关性分析结果表明,SNP6、SNP7、SNP16和SNP24等4个位点与生长性状呈显著(P<0.05,下同)或极显著(P<0.01,下同)相关,其中位点SNP6与体长和体重关联极显著,与头胸甲宽关联显著;SNP7与体长、头胸甲宽、第一腹节宽和体重关联极显著,与第一腹节高关联显著;SNP16与头胸甲宽、第一腹节宽、第一腹节高和体重关联极显著,与体长关联显著;SNP24仅与第一腹节高关联显著。对浙江群体中60尾个体体长50%小个体和体长50%大个体2个群体中4个SNP位点的基因型频率进行统计,结果显示,SNP6和SNP24 2个标记与生长的关联性显著,SNP7和SNP16与生长的关联性极显著。【结论】HSP90和HGS基因可能是与罗氏沼虾生长相关的重要功能基因,研究结果为下一步基因定位及分子标记辅助育种提供了更多的标记基础。

Abstract:【Objective】 To clarify the relationship between SNP marker loci and growth traits of Macrobrachium rosenbergii, so as to lay a foundation for the search of candidate genes for growth traits, the regulation mechanism of growth traits and the study of molecular marker-assisted breeding in the later stage.【Method】 The direct sequencing technique was used to detect the polymorphism of 25 SNP markers in the extreme population of growth traits of M. rosenbergii. Chi-square test(χ2) was used to screen two SNP marker loci with potential correlation with growth status in two extreme populations(maximum and minimum in body length) of M. rosenbergii. Further, the association between SNP genotypes and growth traits(body length, carapace width, first abdominal segment width, first abdominal segment height and body weight) was analyzed in 60 individuals of M. rosenbergii Zhejiang population. The general linear model was used to detect the correlation between SNP marker loci and five growth traits of M. rosenbergii.【Result】 Chi-square test showed that SNP6, SNP7, SNP16, SNP20 and SNP24 were potentially associated with growth traits in M. rosenbergii population. The results of correlation analysis between five potential correlation SNP marker loci and growth traits showed that SNP6, SNP7, SNP16 and SNP24 were significantly(P<0.05, the same below) or extremely significantly(P<0.01, the same below) correlated with growth traits, in which SNP6 was extremely significantly correlated with body length and weight, and significantly correlated with carapace width, while SNP7 was extremely significantly correlated with body length, carapace width, first ventral segment width and body weight, and significantly correlated with first ventral segment height. SNP16 was extremely significantly correlated with carapace width, first abdominal segment width, first abdominal segment height and body weight, and significantly correlated with body length, while SNP24 was only significantly correlated with the first abdominal segment height. The genotypic frequencies of 4 SNP loci in 60 individuals with 50% small body length individuals and 50% large body length individuals in Zhejiang population were statistically analyzed. The results showed that 2 markers in SNP6 and SNP24 were significantly associated with growth, and SNP7 and SNP16 were extremely significantly associated with growth.【Conclusion】 HSP90 and HGS genes may be important functional genes related to the growth of M. rosenbergii.
