鳄龟血液抗菌肽生物活性研究

为探讨鳄龟血液抗菌肽初提物的生物活性,采用纸片法和固体培养基连续稀释法对其进行抗菌效果检验,结果表明:分离纯化的血液抗菌肽对G-细菌和G+细菌均有不同程度的抑制或杀灭作用,但不同的微生物对其敏感性不同。其对致病性嗜水气单胞菌、葡萄球菌、枯草杆菌、蜡样芽孢杆菌和沙门氏菌的最小抑菌质量浓度分别为12.5,25,50,50,200μg/mL。鳄龟血液抗菌肽具有①耐热性:将其沸水浴5 min后仍然具有极强的抗菌活性,沸水浴10 min后其抗菌活性减弱;②耐酸性和不溶血性:pH值为6.0,5.0,4.0,2.5时,鳄龟血液抗菌肽对嗜水气单胞菌均有抑菌活性,当pH值下降至2.5时其抑菌圈最小。

Study on Biological Activity of Antimicrobial Peptides Extracted from Blood of Chelydra serpentina

In order to research the biological activity of antimicrobial peptides crudely extracted from the blood of Chelydra serpentina,the antibacterial teats were conducted by the methods of slip and serial dilution incubated on the solid medium.The results showed that the antimicrobial peptides could inhibit or kill Gram-positive and Gram negative bacteria.The minimal inhibition concentrations (MIC) of the antimicrobial peptides were measured by means of micro-titer-plate assay.The MIC of the peptides on A.hydrophila,Staphylococcus aureus,Bacillus subtilis,Bacillus cereus and Salmonell aderby was 12.5 μg/mL,25 μg/mL,50 μg/mL,50 μg/mL and 200 μg/mL,respectively.Besides,the antimicrobial peptides were provided with following characteristics:①Heat stability: their activity could maintain after being bathed at 100℃ for 5 min but be attenuated for 10 min.②Acid resistance and non-haemolyticus: it could inhibit A.hydrophila to grow in condition of pH6.0,5.0,4.0,2.5,but not inhibit the bacteria effectively in environment of pH2.5.
