氟苯尼考粉中非法添加烟酰胺和氨茶碱的HPLC-PDA检测方法的建立

摘    要:为检测氟苯尼考粉中非法添加的烟酰胺和氨茶碱,以十八烷基键合硅胶为填充剂,0.1%甲酸溶液(三乙胺调节pH值至3.0)-甲醇为流动相,建立了HPLC-PDA检测方法,并采用峰纯度检查和光谱相似度检查辅助对照品比对方法,对非法添加药物进行确证。在此液相色谱条件下,烟酰胺、氨茶碱与其他物质峰分离良好。按外标法以峰面积计算,烟酰胺和氨茶碱的平均回收率分别为99.8%和99.6%.RSD分别为0.1%和0.3%。结果表明,该检测方法准确、可靠,可用于同时测定氟苯尼考粉中非法添加的烟酰胺和氨茶碱。

Determination of Nicotinamide and Aminophylline in Florfenicol Powder by HPLC-PDA

Abstract:A method for the determination of nicotinamide and aminophylline in the florfenicol powder was developed by the high performance liquid chromatography with photo-diode array detector (HPLC-PDA). It was tested with C18 column, using gradient elution with 0.1% formic acid solution (adjusting pH to 3.0 by triethylamine) and methanol as the mobile phase. The extract wavelength was 261nm. Peak purity test and spectrum similar test were helped to identify the nicotinamide and aminophylline. The mean recovery of nicotinamide and aminophylline were 99.8% and 99.6%, and RSD were 0.1% and 0.3%, respectively. In conclusion, the method is simple, accurate and reliable for the determination of nicotinamide and aminophylline in florfenicol powder.
