| 摘 要： | 应用胶体金免疫层析技术（GICA）进行磺胺甲嗯唑（SMZ）的残留检测研究。制备抗磺胺甲嗯唑多克隆抗体，并采用柠檬酸三钠法制备20nm胶体金颗粒。胶体金标记多抗后喷涂到玻璃纤维膜上，特异性抗原（SMZ-OVA）以线状包被在硝酸纤维素膜上，制成快速检测试纸条。滴加检测溶液后，若呈现一条红色条带则为阴性，无红色条带则为阳性。该方法对SMZ标准溶液的灵敏度达到50ng／mL，试纸条显色结果清晰，反应灵敏，整个检测反应在5～10min内完成，稳定性及重复性良好。

| Abstract: | A gold immunochromatographic assay(GICA) was applied to determination of sulfamethoxazole(SMZ) residues.Prepare polyclonal anti-sulfamethoxazole antibodies, tri-sodium citrate with aqueous gold chloride were mixed to make gold sol.Colloidal golds were coupled with polyclonal antibody and then to be solidified to glass(fibers),SMZ-ovalbumin was immobilised on nitrocellose membrane.Using SMZ standand solution to test,a negative result was the appearance of a red line on the test stip,while a positive result was the absence of the red line.The device had a cut-off value of 50 ng/mL in SMZ standard solution.The whole test of the GICA could be finished within 10 minutes.The stabilization and repetition performed well. |
