猪MAP2K6基因多态性及其与经济性状的关联分析

摘    要:本实验旨在探究猪丝裂原活化蛋白激酶激酶6(Mitogen-Activated Protein Kinase Kinase 6,MAP2K6)多态性与猪重要经济性状的关系,为猪育种提供新的标记资源。实验选用法系大白猪为研究对象,采用质谱技术对猪MAP2K6基因的SNPs位点进行基因分型,同时利用线性混合模型分析单个标记位点与猪重要经济性状的相关性,共检测出4个SNPs,通过与生长育肥和乳头数性状的关联分析表明,rs345304630(SNP1)、rs325278117(SNP2)和rs332017877(SNP3)与达100 kg体重日龄和右乳头数显著相关,rs325752048(SNP4)与眼肌面积显著相关;连锁不平衡结果表明,MAP2K6基因SNP1、SNP2和SNP3处于强连锁不平衡状态。结果提示,在育种进程中,通过对优质基因型的选择来缩短母猪达100 kg体重日龄,增加母猪的泌乳和带仔能力,对提高哺乳仔猪的存活率和断奶重具有重要意义,也可为猪生长育肥和乳头数性状的分子标记的挖掘和利用提供一定的理论依据。

Abstract:The study aimed to study the association between MAP2K6 gene and growth trait in pigs,and to provide new markers for pig breeding.In this study,we used France Large White pigs as the research object to genotype the SNPs of MAP2K6 by mass spectrometry.At the same time,a linear mixed model was used to analyze the correlation between individual marker loci and important traits of pigs.Four single nucleotide polymorphisms(SNPs)were detected by analyzing the correlation between SNPs and growth,fertility and papillary number.The results showed that the polymorphism sites rs345304630(SNP1),rs325278117(SNP2)and rs332017877(SNP3)were significantly associated with the traits of the age at 100 kg and the number of right side teats.The polymorphism site rs325752048(SNP4)significant correlation with the eye muscle area.The results of linkage disequilibrium showed that SNP1,SNP2 and SNP3 of MAP2K6 gene were in a strong linkage disequilibrium state.The results showed that selecting high-quality genotypes to shorten the sow's age up to 100 kg body weight,increase the ability of sows to lactate and carry piglets,is of great significance for improving the survival rate and weaning weight of suckling piglets,but also for pig growth and nipple growth.The mining and utilization of molecular markers for number traits provide a certain theoretical basis.
