重组型番鸭细小病毒JH10株的分子特征解析

摘    要:为了解析番鸭细小病毒(MDPV)分离株JH10的分子特征,本研究对该毒株全基因组进行了扩增、测序、重组分析及遗传进化树的构建。结果显示:JH10株基因组由5071个核苷酸组成,P9启动子区和VP3基因内部约1.1 kb区域经历了重组,GPV鹅胚化弱毒株SYG61v参与了P9启动子区的重组,而GPV强毒株LH、B和SYG61v不同程度地参与了VP3基因内部的重组;以VP3基因重组片段构建遗传进化树,JH10株与8个经典型GPV株处于同一个进化分支中,而以毗邻的上下游非重组区片段分别构建的遗传进化树中,JH10株则与4个经典型MDPV株处于同一分支。本研究结果表明,JH10株为经典型MDPV和GPV之间的重组产物,该研究结果为进一步阐明重组型MDPV的致病机理奠定了基础。

Abstract:In order to unravel the genomic characteristics of a Muscovy duck parvovirus(MDPV)strain JH10,the whole genome was amplifi ed and sequenced for recombination and phylogenetic analysis.The result showed that the genome of JH10 comprised of 5071 nucleotides.Recombination analysis was executed with the software Simplot 3.5.1 and RDP4,which demonstrated that JH10 underwent two recombination events,among which the goose embryo-adapted vaccine strain SYG61v was solely involved in the recombination event that occurred in the P9 promoter region,and the virulent GPV strain LH,B and SYG61v was involved in the recombination event that occurred in the middle 1.1 kb region of the VP3 gene in various degrees.The phylogenetic tree was constructed based on the 1.1 kb recombination region in the VP3 gene,which showed that JH10 clustered with all classical GPV strains while strain JH10 clustered with all classical MDPV strains in the phylogenetic trees constructed based on the two DNA fragments adjacent to the 1.1 kb recombination region.These results indicated that strain JH10 derived from recombination between the classical MDPV and GPV strains.The present study laid a solid foundation for elucidating the pathogenesis of recombinant MDPV in the future.
