盐胁迫条件下苦马豆幼苗根差异表达基因及功能注释

摘    要:苦马豆(Swainsona salsula Taubert)是新疆重要的耐盐抗旱豆科植物,通过染色体压片技术确定供试材料为二倍体,采用截形苜蓿(Medicago truncatula)基因组芯片(61278个探针),对苦马豆幼苗根部在未经盐处理和在180 mM NaCl溶液中处理24 h后的表达水平进行了分析,找出控制盐胁迫的关键基因,为耐盐基因的筛选提供依据。结果表明:有4057个基因在不同处理条件下存在表达差异,其中155个上调基因和105个下调基因;利用Gene Ontology注释对差异表达的基因从分子功能、细胞组件、生物学过程3个水平进行功能分类,搜索NCBI数据库,在截形苜蓿和拟南芥(Arabidopsis thaliana)中找到有高相似性功能已知的部分基因,表明差异表达的基因涉及逆境胁迫、信号转导、电子传递、蛋白代谢等多个方面,这些基因是植物根部盐胁迫相关基因。

Abstract:Swainsona salsula Taubert is a valuable and drought resistant salt tolerant legume in Xinjiang.Key genes controlling salt stress are investigated here for providing genetic screening basis of salt tolerance.S.salsula is confirmed as a diploid legume determined by chromosome pressing technology.Affymetrix Medicago genome gene chip containing 61,278 probes are used to identify differentially expressed genes of roots both without and with salt treatment(180 mM NaCl,24 hours).Results show that 4,057 genes were identified to be significantly differentially expressed between the two treatments.There are 155 up-regulated genes and 105 down-regulated genes.Gene ontology annotation is used to analyze differentially expressed genes at molecular function,cell components and biological process levels.By searching NCBI database,some genes show high similarity with known genes of Medicago truncatula and Arabidopsis thaliana.Some differentially expressed genes show high identity with stress-inducible genes,signal transduction genes,electron transport or protein metabolism genes related to salt-stress in plant roots.
