摘    要:在建立竞争ELISA方法的基础上,首次研制出检测磺胺二甲嘧啶的单克隆抗体快速检测试剂盒,并对其检测限、精密度、检测范围以及鸡肌肉组织中的添加回收实验做了详细研究。本试剂盒的检测限为1.0ng/m1,检测范围为1.0-81.0ng/m1,批内变异系数<8.9%,批间变异系数<9.5%,在10、60和200ng/m1水平鸡肌肉组织中添加,回收率为64.5%-85.5%,变异系数为6.0%-18.6%。与同类相关德国产试剂盒相比较,阳性符合率为100%。

Abstract:A sensitive and specific ELISA for SM2was established by anti-sulfamethazine monoclonal antibody. In the assay,sulfamethazine residue in sample was allowed to compete with the sulfamethazine-OVA conjugate immobilized on microtiter wells for binding to a limited amount of the anti-sulfamethazine monoclonal antibody, Horseradish peroxidase coupled to the goat IgG of anti-mouse IgG was selected as a tracer. Standard curves were constructed over 1 - 81 ng/ml of sulfamethazine,and the limit of detection of the ELISA was 1 ng/ml. Recoveries of sulfamethazine in spiked samples between 10, 60 and 200 ng/ml ranged from 64. 5% to 85. 5%. Precision of the assay as determined by interassay and intraassay variabilities was below 10%. Among 6 sulfonamide analogues tested for specificity .Sulfadimethoxine and sulfamer-azine showed significant cross-reaction in the assay. The assay is rapid,sensitive,and convenient to perform, and the test is readily adaptable to high-volume testing of sample in slaughterhouses.
