CPT1A基因在广西麻鸡的表达研究

摘    要:该研究旨在阐明CPT1A基因在广西麻鸡组织表达和时序表达谱,为研究鸡体内脂肪代谢提供一定的基础资料。选取胚胎期到性成熟时的广西麻鸡,在不同发育时间点采集胸大肌、腓肠肌、心脏和肝脏组织样品,利用实时荧光定量方法,分析CPT1A基因在不同组织不同发育阶段表达规律。组织表达结果显示,CPT1A在广西麻鸡不同发育阶段4种组织中表达差异性不同。在胚胎期,CPT1A基因在腓肠肌的表达量最高;1日龄和1周龄时,CPT1A在心脏表达量最高;5、7、9、13和16周龄时,CPT1A在胸大肌的表达量最高。时序表达结果显示,CPT1A在肝脏各个发育阶段表达量均较低。CPT1A在胸肌的表达量呈现逐渐上升的趋势,且变化趋势最显著,5、7、9、13、16周龄的表达量显著高于其他试验阶段。时序表达谱显示,CPT1A基因在同一发育阶段4种组织表达量不同,4种组织在不同发育阶段变化趋势不同。

Abstract:The aim of this study was to elucidate the tissue expression and temporal expression profiles of CPT1A gene,and provide some basic data for studying the mechanism of fat metabolism in chicken.Guangxi Partridge chickens from embryonic stage to market age were selected as experimental materials.Pectoral muscle,gastrocnemius,heart and liver tissue samples were collected at different developmental stage.The expression of CPT1A gene in different developmental stages of different tissues was analyzed by real-time fluorescence quantitative method.The results of tissue expression showed that CPT1A had different expression level in the four tissues at different developmental stages.The expression level of CPT1A gene in the gastrocnemius was highest during embryonic stage.At 1 d,and 1 week,the expression levels of CPT1A in the heart were highest.At 5,7,9,13 and 16 week,the expression levels of CPT1A in the pectoral muscle were highest.The temporal expression showed that the expression levels of CPT1A in the liver were lower at each experimental development stage.The expression of CPT1A in the pectoral muscle showed a gradual increase trend,and the trend of change was the most significant.The expression levels at 5,7,9,13 and 16 week in the pectoral muscle were significantly higher than in the other experimental stages.The results of spatial and tissues expression profiling showed that the expression levels of CPT1A gene in four tissues at the same developmental stage were different,and the trends of four tissues in different developmental stages were different.
