摘    要:利用EST-SSR分子标记,基于最适取样策略构建了15份老芒麦种质的DNA指纹图谱,并对其遗传多样性进行了评价。结果显示,16对引物扩增出的条带多态信息含量变化范围为0.0457(Ps569)~0.4667(Elw1468s087),表明15份老芒麦材料具有较丰富的遗传多样性。其中,甘南老芒麦种群遗传多样性最丰富(H=0.2165,I=0.3285),其次是F 2种群(H=0.1662,I=0.2439)、育成品种(H=0.1583,I=0.2311)和俄罗斯种质(H=0.1078,I=0.1573)。利用引物Elw5627s404和Ltc0157构建的DNA指纹图谱,可将所有15份材料进行区分。从地理类群分析,4个地区老芒麦种质的遗传相似度变化范围为0.7536~0.8728,遗传距离变化范围为0.1361~0.2829。遗传结构聚类分析将15份材料分为三大类:类群Ⅰ为甘南的6份材料,类群Ⅱ为混合种群,类群Ⅲ为3份F 2种质。

Abstract:In the study,the DNA fingerprints of 15 Elymus sibiricus accessions were constructed based on the optimal sampling strategy using EST-SSR molecular markers,and their genetic diversity was evaluated.The results showed that the polymorphic information content(PIC)of bands amplified by 16 pairs of primers ranged from 0.0457(Ps569)to 0.4667(Elw1468s087),which indicated that 15 E.sibiricus accessions had rich genetic diversity.Among them,Gannan population had the most abundant genetic diversity(H=0.2165,I=0.3285),followed by the F 2 population(H=0.1662,I=0.2439),bred varieties(H=0.1583,I=0.2311)and Russian population(H=0.1078,I=0.1573).The DNA fingerprints constructed with primers Elw5627s404 and Ltc0157 could distinguish all 15 accessions.From the geographical group analysis,the genetic similarity(GI)of E.sibiricus accessions in four regions ranged from 0.7536 to 0.8728,and the genetic distance(GD)varied from 0.1361 to 0.2829.Cluster analysis of genetic structure showed that 15 accessions were divided into three groups:GroupⅠwas 6 accessions from Gannan,GroupⅡwas a mixed population,and GroupⅢwas 3 accessions from F 2.
