三种不同方法显示仔猪小肠肌间神经元的比较

本研究采用小肠铺片NADPH-d组化法、NADH-d组化法和抗NF免疫组化法分别对0日龄、5日龄、28日龄仔猪的肌间神经元进行了比较研究。结果表明:NADPH-d组化法可以选择性的标记合成NO的神经元亚群,大部分阳性肌间神经元符合DogielI型神经元的形态特征;NADH-d组化法可以非选择性的标记大部分神经元,且采用该法观察到的肌间神经元的密度明显高于NADPH-d组化法,但该法不能清晰的显示肌间神经丛的细微结构,不适合用于神经元类型的观察;抗NF免疫组化法可以清晰的显示肌间神经元的突起,28日龄仔猪体积较大的阳性神经元主要为DogielI型和DogielII型神经元。

The Comparative Study on the Myenteric Neurons in the Small Intestine of Piglets by Three Different Methods

The morphology of the myenteric neurons was studied comparatively,using the NADPH-diaphorase histochemical method,the NADH -diaphorase histochemical method and the anti neurofilament immunohistochemical method on the whole -mount preparations of the small intestine of the piglets aged0day,5days and28days.The results were as follows :the subpopulation of myenteric neurons,which were able to synthesize nitric oxid,could be marked selectively by the NADPH -diaphorase histochemical method.Most of the NADPH-diaphorase positivemyenteric neurons were Dogiel type I neurons.Most of the myenteric neurons were marked non -selectively by the NADH-diaphorase histochemical method.The spatial density of the NADH-d positive myenteric neurons was much higher than that of the NADPH-diaphorase positive myenteric neurons.But we could not see the smaller structure of the myenteric plexus clearly,so this method could not be used in the observation of the type of the myenteric neurons.The projectors of the myenteric neurons could be seen clearly by the anti neurofilament immunohistochemical method.Some big anti neurofilament immunoreactivity neurons were Dogiel type I neurons,and others were Dogiel II type neurons in the piglets aged28days.
