摘    要:本研究应用原位杂交法、免疫组化、PCR法对肌肉注射猪瘟基因疫苗pcDST、pcDSW后小鼠胫前肌中质粒存在的方式、部位及时间进行了动态观察,结果表明猪 病毒基因疫苗注射3h后质粒就到达细胞核,至20d在细胞核内仍呈阳性反应;pcDST质粒在肌肉内存在120d,pcDSW质粒在肌肉存在150d;pcDST质粒肌肉注射后第3d,pcDSW质粒肌肉注射后第5d,在胫前肌肌肉中可检测到表达产物,持续表达20d以上。表明构建的猪瘟基因疫苗经肌肉注射是较好的免疫途径。

Abstract:In situ hybridization,immunohistochemistry and PCR were used to detect the CSF DNA vaccine expressing plasmids,pcDST and pcDSW,which were injected into the muscle of mice.The pattern and the time of their appearance in the muscle were also observed.The results demonstrated that pcDST and pcDSW could be seen in the nucleus of muscle cells by in situ hybridization 3 h after injection,and was positive untill 20 days.pcDST existed in the muscle for about 120 days and pcDSW for 150 days by PCR analysis.The products expressed could be detected 3 days after injection with pcDST and 5 days with pcDSW,the expressing persisted untill 20 days.Intramuscle injection is a good route for CSF DNA vaccine.
