GnRH促鸡卵泡颗粒细胞孕酮合成功能作用机制的研究

摘    要:本实验选用掌握产蛋规律的莱航鸡,在排卵前16-18h剖腹取卵泡,分离和纯化颗粒细胞。采用无血清贴壁培养法,初步探讨了鸡促性腺激素释放激素促颗粒细胞孕酮合成的作用机制。获得以下结果:(1)PKC阻断剂H7(25-50μmol/L)能阻断GnRH-Ⅱ促颗粒细胞P4合成作用;(2)磷酸二酯酶抑制剂MIX(10^-5-10^-3mol/L)能促进P4分泌,但对GnRH-Ⅱ促P4合成效应无明显影响;(3)钙

Abstract:ThepresentexperimentswereconductedtostudythemechanismofGnRHstimulatingprogesterone(P4)productionbyculturedchickengranulosacells(GC)Thedifferentsizeatfollicles(F1F4)wereobtainedfromlayinghens(Leghornstrain)whichwerekilledat1618hbeforeovulationTheGCswereisolatedfromthegranulosalayerswith01%collagenasefor5to7minutesandculturedinaserumfreemediumTheresultsshowthat(1)H7,(2550mol/L)couldblockGnRHinducedP4production;(2)MIX(10-510-3mol/L)alonestimulatedtheprogesteroneproduction,buthadnosignificanteffectonP4productioninducedbyGnRH;(3)P4productioninducedbyGnRHweredecreasedsignificantlyinthepresenceofEGTA(13mmol/L),butitseffectwasconvertedbyaddingCa2 15mol/L;(4)TheactionofVerapamil(thecalciumchannelblocker)suppressedP4productionstimulatedbyGnRHwasnotsignificantTheresultsmentionedaboveindicatethatthePKCsystemmaymediatetherelationofP4productionbyGnRHinchickenGC;andtheactionofintracellularcalciumisalsoimportant
