| 摘 要： | 非洲猪瘟给全球养猪业造成了严重危害,需要建立更加敏感的PCR检测方法。根据非洲猪瘟病毒(ASFV)的VP72基因序列,设计PCR引物,建立了ASFV PCR检测方法。以合成的VP72基因片段为阳性对照,进行敏感性试验和特异性试验。同时与世界动物卫生组织(OIE)推荐的PCR引物进行对比试验。结果显示,建立的方法可用于ASFV检测,且对其他病毒检测时无交叉反应,特异性良好;敏感性可达到10-8,比OIE推荐的方法高10倍,为ASFV检测提供了一种新的PCR方法。

| Abstract: | A more sensitive PCR method was required to detect African swine fever virus(ASFV)because of that serious harm to swine industry.The PCR method was established based on VP72 gene fragment of ASFV.Subsequently,the sensitivity and specificity tests were performed with synthetic VP72 gene segment as positive control.At the same time,a comparison of sensitivity was carried out between PCR primers recommended by OIE and designed in this research.Finally,the PCR method of ASFV was established sucessfully with good specificity(no cross-reaction with other viruses)and high sensitivity(10-8).Specially,the sensitivity of our primer was higher about 10 times than that of PCR primer recommended by OIE.A new PCR method was provided for ASFV detection. |
