中药复合剂AFF-Ⅰ口服液对小鼠急性肝损伤血液生化指标的影响

摘    要:为了研究中药复合剂AFF-Ⅰ口服液对四氯化碳(CCl4)导致小鼠急性肝损伤的保护效果,将60只昆明种小鼠随机分为正常对照、造模、阳性药物对照(联苯双酯)、中药复合剂AFF-Ⅰ口服液高、中、低剂量6组。正常对照组、造模组分别灌服等量的生理盐水,阳性药物对照组按150mg/kg灌胃联苯双酯溶液,中药复合剂AFF-Ⅰ口服液高、中、低剂量组分别按45、30、15mL/kg灌胃,2次/d,连续给药7d。末次给药后1h,正常对照组腹腔注射花生油20mL/kg,其余组腹腔注射10mL/L CCl4花生油溶液20mL/kg,16h后测定血清中天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)含量和肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)含量。结果表明,中药复合剂AFF-Ⅰ口服液低、中、高剂量组能显著降低CCl4导致小鼠急性肝损伤血清中AST、ALT含量(P<0.01),显著降低CCl4所致急性肝损伤小鼠肝组织MDA含量,提高肝组织SOD活力(P<0.01)。研究表明,中药复合剂AFF-Ⅰ口服液对小鼠的急性肝损伤具有保护作用。

Abstract:To study the hepato-protective effect of AFF-1 oral solution on carbon tetrachloride(CCl4) induced acute hepatic injury in mice,sixty KM mice were randomly divided into normal control group,model group,positive drug control group-bifendate pill group,AFF-1 oral solution low dose group,medium dose group and high dose group.In normal control group and model group were administrated equal amounts of saline,respectively.The mice bifendate pill group were intragastrically administrated with 150 mg/kg.In anti-hepatitis-jaundice oral solution low dose group(15 mL/kg),medium dose group(30 mL/kg) and high dose group(45 mL/kg) were intragastrically administrated continuously for 7 days,twice a day.After the last dose 1 h,in normal control group were injected intraperitoneally 20 mL/kg peanut oil,in the remaining group were intraperitoneal injection of 0.1% CCl4 peanut oil solution 20 mL/kg.Sixteen hours later,blood was collected to detect the activities of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in sera,and liver homogenate was prepared to determine the activities of superoxide dismutase(SOD) and the content of malondialdehyde(MDA).The mice in high,middle and low dosages of the AFF-1 oral solution groups,the activities of ALT and AST in sera were significantly decreased(P<0.01).The content of MDA in liver was significantly decreased and the activity of SOD was significantly increased.Conclusion: AFF-1 oral solution has a good protective effect on the acute hepatic injury in mice induced by CCl4.
