| 摘 要： | 用线性化质粒与酵母细胞混合后进行电转化,培养细胞,菌体计数,通过计算转化率探索ECM 830电转仪的转化条件,确定出毕赤酵母电转化最佳条件,低电压模式;电压300 V;脉冲长度15 ms;脉冲次数为1次;为ECM 830电转仪在酵母转化中的应用提供依据. |

| Abstract: | The linear plasmid was mixed with Pichia pastoris,and the mixture was transformed by ECM 830 electroporator.After culturing and enumerating Pichia pastoris,the electransformation condition was searched according to the transformation coefficient.The optimal protocol for electroporation of Pichia pastoris was as followings:votage 300V,pulse length 15ms,pulse once.The results provided the reference for using ECM 830 electroporator in the electransformation of Pichia pastoris. |
