鸭黄病毒的分离鉴定

摘    要:从临床表现为产蛋下降、头颈歪斜、软脚为主要特征的发病蛋鸭卵巢、脑浆、肝、脾等组织中分离到1株病毒。分离毒能致死番鸭胚和鸡胚,人工感染产蛋期的蛋鸭能导致体温升高、产蛋急速下降,卵泡出血和萎缩;分离毒不能凝集鸡和鸽红细胞,能被鸡黄病毒高免血清特异中和;用禽黄病毒特异引物扩增为阳性。上述结果证实分离毒是黄病毒科黄病毒属的鸭黄病毒,是导致蛋鸭产蛋下降的病原。

Abstract:One novel flavivirus strain named DJD09 was isolated from a shelduck farm that experienced abrupt egg production decline and young ducks’ neurological symptoms.Poultry embryo deaths resulted from inoculating this virus into allantoic cavity with lesion of liver hemorrhage.This isolate had no haemagglutination activity to erythrocytes of pigeon and chicken.Healthy shelducks inoculated with this virus showed symptoms similar to those of field cases.In cross-neutralization tests,antiserum to chicken flavivirus CJD05 strain neutralized DJD09.Positive fragment was amplified by RT-PCR with the specific primers of avian flavivirus.These results indicated that DJD09 virus isolate is newly emerged duck flavivirus strain that cause an infectious disease in ducks.
