一株狂犬病病毒街毒株的分离鉴定与组织培养

摘    要:对来自河北省无极县某地区疑似狂犬病病犬脑组织进行FAT、电镜观察、RT-PCR扩增,以及MIT法等确诊为狂犬病,对分离到的这株狂犬病街毒命名为WJ07-1,分析N蛋白和G蛋白序列,主要抗原位点发生轻度变异,同源性分析发现N基因与ZhejiangWz(H)株同源性最高为99.0%,G基因与JSL27株同源性最高为99.4%。对分离到的狂犬病病毒进行组织培养发现这株狂犬病病毒街毒株对细胞的适应性和感染性很弱,需要进行多次传代培养,才可以适应N2a、BHK-21细胞。

Abstract:FAT, electron microscope, RT-PCR,and MIT were used to diagnose a street (rabies) virus strain from Wuji County Hebei province,which was named WJ07-1. Few variation of the major antigen sites were found after analysing the N gene and G gene sequences, homology of nucleotide of N gene with ZhejiangWz (H) was 99.0%; homology of nucleotide of G gene with JSL27 was 99.4%. Adaptability and infectivity on cells were found weak af- ter tissue culture,More subcultures are needed for it to adapt N2a cells and BHK-21 cells.
