猪霍乱沙门菌C500运载猪瘟病毒新型基因疫苗在家兔体内的免疫应答

摘    要:为研究猪霍乱沙门菌C500(S.C500)运载猪瘟病毒(CSFV)新型基因疫苗口服免疫家兔的体内免疫应答特点,采用电转化法将CSFV新型基因疫苗转化到S.C500,构建重组工程菌,口服免疫接种家兔,检测CSFV、S.C500特异性抗体;并用猪瘟兔化弱毒疫苗及猪伤寒沙门菌野毒株依次进行攻毒试验。结果显示,成功构建CSFV新型基因疫苗重组菌S.C500/pCB-ME2-IL-15,S.C500/pCC-ME2-IL-15。口服免疫家兔可以诱导产生抗CSFV和S.C500的特异性ELISA抗体,且S.C500/pCC-ME2-IL-15略显优势。经三免后免疫家兔能够部分抵抗猪瘟兔化弱毒疫苗与猪伤寒沙门菌野毒株的攻击。试验提示以S.C500为CSFV新型基因疫苗运载体的猪用重组活菌苗具有可行性。

Abstract:In order to research the characteristics of the rabbits immune response in vivo by oral immunization with Salmonella choleraesuis C500(S.C500) carrying classical swine fever virus(CSFV) novel-gene-vaccine.The CSFV vaccine expression vectors were transformed into S.C500 by electroporation to construct re-engineering strains.The experimental rabbits were vaccined with the recombinant bacteria by oral immunization and detected for CSFV and S.C500 specific antibody.The immunized rabbits were attacked with HCLV vaccine and pig wild strain Salmonella typhi in turn.The results showed that the recombinant bacteria,which named S.C500/pCB-ME2-IL-15 and S.C500/pCC-ME2-IL-15,were successfully constructed for CSFV novel-gene-vaccine.The immunized rabbits produced specific ELISA antibodies of CSFV and S.C500,and the S.C500/pCC-ME2-IL-15 was slightly comparative advantages.The experimental rabbits with three times immunized could partially resist the challenge of HCLV vaccine and pig wild strain Salmonella typhi.The research demonstrated the potential of developing porcine recombinant live vaccine,which use S.C500 as CSFV novel-gene-vaccine transporter.
