| 摘 要： | 应用荧光定量PCR技术,根据兔病毒性出血症病毒的保守基因VP60设计了1对引物和1段Taqman MGB探针,建立了用于检测兔病毒性出血症病毒的实时荧光定量RT-PCR方法。试验中能够检出的RHDV VP60基因质粒拷贝数达103数量级,能够检测到RHDV病毒核酸最低量可以达到5 pg,未检出其他病原的RNA。试验结果表明,建立的TaqMan MGB探针实时荧光定量RT-PCR方法的特异性、敏感性、重复性均达到试验设计要求,能快速检测临床样品中的兔病毒性出血症病毒,适合于兔各脏器及肌肉组织中兔病毒性出血症病毒的快速诊断和检测。

| Abstract: | Based on the fluorescence quantitative PCR technology,referenced rabbit hemorrhage disease virus's(RHDV)conservative gene VP60,designed primers and TaqMan MGB probe.The method has been used in detecting the rabbit hemorrhage disease virus by real-time RT-PCR.The result indicated no other viral RNA was delected,and it can detect the VP60 gene plasmid copy number reaching 103 magnitudes,also can detect the RHDV to 5 pg.The specificity and the sensitivity of the method achieved the experimental design request.The method can fast detect the clinical sample of RHDV within 5 hours,also can be fit for the detection of viscera and meat of rabbits. |
