ABA对PEG胁迫下狗牙根可溶性蛋白质的影响

摘    要:为了探明ABA对水分胁迫下可溶性蛋白质的影响,以狗牙根的幼苗为材料,分为PEG处理和ABA预处理24 h PEG处理2组,研究了16 h内可溶性蛋白质变化.SDS -PAGE结果表明,PEG处理后可溶性蛋白质含量先下降后上升,Rf值为0.05,0.29,0.38,0.80,0.87(分子质量198.4,93.3,70.6,19.0,15.5 kD)的蛋白质谱带随胁迫时间延长逐渐消失,同时也新增了几条Rf值为0.10,0.14,0.35,0.63, 0.97谱带(分子质量165.8,149.3,76.5,32.7,11.4 kD).ABA处理24 h后,可溶性蛋白质种类和含量均高于CK,与PEG胁迫16 h后的蛋白质谱带近似,增加了如Rf值为0.05,0.14,0.64的蛋白质谱带(分子质量196.0,148.4,31.6 kD).ABA处理24 h后,可溶性蛋白质含量则缓慢降低;推测ABA参与了这些与抗旱有关的蛋白质的基因表达.

Abstract:The effects of ABA on soluble proteins in seedlings of Cynodon dactylon (PEG treated groups and PEG treated groups following ABA pretreatment) under 40% PEG stress were investigated by using SDS-PAGE (sodium dodecyl sulphate polyacrylamide gel electrophoresis) in this paper. The results showed that the soluble protein content in the seedlings dropped and then ascended in the PEG treated groups. Some protein bands with Rf value 0.05,0.29, 0.38, 0.80, 0.87 (molecular weight 198.4,93.3,70.6,19.0,15.5 kD) disappeared away with increasing water stress and some new protein bands with Rf value 0.10, 0.14, 0.35, 0.63, 0.97 ( molecular weight 165.8,149.3,76.5,32.7,11.4 kD) emerged in C. dactylon after 40% PEG treatment.The soluble protein content in C. dactylon pretreated by ABA for 24 h were higher than CK, and increased protein bands with Rf value 0.05,0.14,0.64 (molecular weight 196.0,148.4,31.6 kD) which were similar with that of 16 h PEG treatment. And then the content of protein decreased with increasing the PEG treatment time after 24 h ABA pretreatment. The experiments suggested that ABA participates in signaling mechanisms of the expression of some drought-induced proteins.
