家蚕5龄幼虫不同时期中部丝腺细胞蛋白质双向电泳分析

摘    要:对5龄家蚕不同发育时期的中部丝腺细胞蛋白质变化进行分析研究,有利于发现与丝胶蛋白分泌有关的功能蛋白质。采用蛋白质双向电泳和图像分析技术,发现同一区段的家蚕中部丝腺细胞在5龄第2天、第4天和熟蚕期的蛋白质组成具有显著差异:中部丝腺前部区段5龄第2天表达的特异性蛋白质斑点有8个,第4天有16个,熟蚕期有24个;中部区段5龄第2天表达的特异性蛋白质斑点有9个,第4天有33个,熟蚕期有30个;后部区段5龄第2天表达的特异性蛋白质斑点有10个,第4天有7个,熟蚕期有25个。

Abstract:Investigation on the protein varieties in the middle silkgland cells of silkworm,Bombyx(mori),during 5~(th) instar stage could find out the functional proteins concerning with the secretion of the sericin protein.Using two dimensional polyacrylamide gel electrophoresis(2D-PAGE) and the image analysis technology,the results indicated that a significant difference in the protein composition of the same part of middle silkgland cells among 2~(nd) day,4~(th) day and the mature stage during 5~(th) instar.Eight special protein spots were expressed in anterior part of middle silkgland cells in 2~(nd) day,16 in 4~(th) day and 24 in the mature stage. In middle part of middle silkgland cells,9,33,and 34 especial protein spots were detected in 2~(nd) day,in 4~(th) day and in the mature stage,respectively.In posterior part of middle silkgland cells,10,7,and 25 especial protein spots were found out in 2~(nd) day,in 4~(th) day and in the mature stage,respectively.
