转BtCry3A基因杨树6个株系体内毒蛋白表达及对桑天牛的抗性鉴定

摘    要:为鉴定、筛选转BtCry3A基因抗虫杨树741杨对桑天牛有较强抗性的株系,选择741杨试验林中的6个转基因株系,检测外源基因整合的稳定性、可溶性蛋白和毒蛋白的表达量以及抗虫性等,并对6个转基因株系的抗虫性进行比较分析。结果表明:外源基因在6个转基因株系基因组中整合稳定,各株系的木质部在整个生长季节均检测到毒蛋白表达,且呈现一定的时间序列规律,均在8-9月份达到高峰期,10月份开始急剧下降,但表达量有差异,其中CC84株系的表达量在7-9月份都明显高于其它株系;6个株系对桑天牛成虫产卵和幼虫的生长均有一定的抑制作用,表现出一定抗性,其中CC84株系对桑天牛幼虫的致死率达到50%,并表现出较稳定的抗虫性。根据检测鉴定结果初步认为,741杨转基因株系CC84具有推广应用潜力。

Abstract:In order to identify and screen transgenic poplar strains with BtCry3A gene for strong resistance against Apriona germari,6 transgenic strains of poplar variety 741 were selected to detect the stability of integrated exogenous gene,the expression quantity of soluble protein and toxic protein,and the resistance against insect.Moreover,a comparative analysis was conducted on insect resistance of the 6 transgenic strains.The results showed that the exogenous gene was stably integrated in the 6 transgenic strains’ genomes.Toxic protein existed in xylem of each strain throughout the growing season.The toxic protein in each xylem showed certain temporal pattern,all of which reached peak value in August or September and started to drop quickly in October.However,the expression quantity was varied,among which strain CC84 had an obviously higher expression quantity over other strains from July to September.All 6 strains had certain inhibitory effect on oviposition of the adult and growth of the larva of A.germari,showing certain resistance.Among the 6 strains,CC84 had a lethal effect of 50% to A.germari larvae and the resistance was fairly stable.Based on these identification results,it is suggested that transgenic strain CC84 has high potential for practical application.
