全蚕粉中1-脱氧野尻霉素的提取分离及分离物对肿瘤细胞的抑制作用

摘    要:利用柱前衍生化反相高效液相色谱法检测不同前处理全蚕粉中1-脱氧野尻霉素(1-deoxynojirimycin,DNJ)的提取分离效果,并研究其分离产物对人宫颈癌HeLa细胞的抗肿瘤作用。结果表明,超低温冷冻干燥、热风烘干和真空浓缩煎煮冷冻干燥所得全蚕粉中DNJ的质量分数分别为0.494 9%、0.383 9%和0.607 6%;冷冻干燥全蚕粉粉碎粒度为0.15、0.25和0.425 mm时,DNJ质量分数分别为0.494 9%、0.321 5%和0.089 5%;用稀酸浸渍及阳离子交换树脂提取与分离全蚕粉中的DNJ效果最佳,分离物中DNJ的质量分数达到1.08%。浓度为40 mg/mL的全蚕粉分离物对肿瘤细胞的生长抑制率达92.27%,可使肿瘤细胞增殖能力完全丧失。结果提示:采用真空浓缩煎煮结合超低温冷冻干燥、超细粉碎和稀酸浸渍及阳离子交换树脂分离技术,可有效提高全蚕粉中DNJ的提取、分离效率,含DNJ的全蚕粉分离物对HeLa肿瘤细胞的生长具有显著的抑制作用。
