典型光活化毒素α-三噻吩对棉铃虫和亚洲玉米螟Na+-K+-ATPase的抑制作用

摘    要:以棉铃虫Helicoverpa armigera和亚洲玉米螟Ostrinia furnacalis为试虫,建立Na+-K+-ATPase最佳反应系统,研究典型光活化毒素α-三噻吩(简称α-T)对Na+-K+-ATPase的影响。棉铃虫Na+-K+-ATPase 活力最佳测试条件为酶源蛋白浓度6 μg/mL,反应温度35~40℃,反应时间6 min;亚洲玉米螟则为酶源蛋白浓度8 μg/mL,反应温度35℃,反应时间6 min。近紫外光照(300~400 nm)对棉铃虫和亚洲玉米螟离体Na+-K+-ATPase活力基本没有影响,但对活体活力有很强的抑制作用。光照和无光照条件下,α-T对两种昆虫离体和活体Na+-K+-ATPase活力均有不同程度的抑制作用。光照组α-T对亚洲玉米螟Na+-K+-ATPase的抑制率高于无光照组,且处理浓度或剂量越高,其抑制率越大;对棉铃虫Na+-K+-ATPase抑制作用不显著。

Abstract:The optimal reaction system of Na+-K+-ATPase from the larval heads of Helicoverpa armigera and Ostrinia furnacalis was set up, respectively. The optimal reaction condition for H. Armigera was 6 μg/mL for protein concentration, temperature 35~40℃ and time 6 min, for O. furnacalis was 8 μg/mL for protein concentration, temperature 35℃ and time 6 min. Under those optimized conditions, effects of α-terthienyl (α-T), a typical photoactivated insecticide, on Na+-K+-ATPase in insect pests were studied. The results showed that Na+-K+-ATPase activity in vivo was significantly inhibited by the near ultraviolet irradiation (300~400 nm), which almost had no effect on that in vitro. However, Na+-K+-ATPase activity in two insect pests after treated by α-T was inhibited both in vivo and in vitro whether irradiation or not. Inhibition rate of Na+-K+-ATPase activity from larvae of O. furnacalis in light group was higher than that in non-light group, and also showing good response-dosage relationship. This suggested that Na+-K+-ATPase might be involved in the mechanism of the action mode of α-T.
