基于线粒体COⅡ基因的乳白蚁序列分析和系统发育研究

摘    要:本研究克隆测序了我国口岸截获和国内采集的7种乳白蚁的线粒体细胞色素氧化酶Ⅱ(COⅡ)基因,并进行序列分析,构建了系统发育树,进行系统发育研究。研究结果表明:乳白蚁COⅡ基因长度为684bp,AT含量为61.2%,明显高于GC含量。乳白蚁属内各种之间的同源性为87.5%~96%,同种的不同地理种群之间的同源性均高于98.9%。塞庞乳白蚁和曲颚乳白蚁的同源性为98.9%~99.8%,系统进化树中聚集在一个进化枝,支持两者可能为同物异名种。

Abstract:This study cloned and sequenced the mitochondrial cytochrome oxidaseⅡ(COⅡ)genes of 7 species of Coptotermes intercepted at ports or collected from China,performed sequence analysis and constructed phylogenetic tree for researching phylogenetic relationship.The results showed that the length of COⅡgene of Coptotermes was 684 bp,and the AT content(61.2%)was significantly higher than the GC.The homology among various species in Coptotermes was 87.5%to 96%,and homology between different geographical populations of the same species was higher than 98.9%.The homology of C.curvignathus and C.sepangensis was reached to 98.9%-99.8%,and clustered in one clade in the phylogenetic tree,implied that these two species may be the same species.
