硫辛酸对大口黑鲈氧化应激相关酶活性的影响

摘    要:为研究硫辛酸对大口黑鲈抗氧化性能的影响,通过腹腔注射不同浓度硫辛酸评估其对大口黑鲈抗氧化酶活性的影响。该试验挑选60尾健康大口黑鲈随机分成3组,根据鱼体质量分别注射含有不同剂量的硫辛酸,设定对照组(0 mg/kg)、T20组(20 mg/kg)和T40组(40 mg/kg),注射6 h、12 h、24 h后分离其血清和肝脏,测定血清中过氧化氢酶(CAT)活性、肝脏中过氧化氢酶(CAT)和总超氧化物歧化酶(T-SOD)活性。结果表明:与对照组相比,T20组和T40组中血清CAT酶活性在6 h和12 h均显著提高(P<0.05);T20组肝脏CAT酶活性与对照组没有差异(P>0.05),T40组肝脏CAT酶活性在处理12 h后显著高于对照组和T20组;与对照组相比,T20组肝脏T-SOD酶活性在3个时间点均显著提高(P<0.05),而T40组肝脏T-SOD活性在6 h和12 h显著提高(P<0.05),且T20组T-SOD酶活性高于T40组。研究结果表明,注射硫辛酸可以促进大口黑鲈血清与肝脏中CAT和T-SOD酶活性,且该促进作用与硫辛酸使用剂量及处理时间有关。

Abstract:In order to explore the effect of lipoic acid on the antioxidant properties,the effect of different lipoic acid doses on the antioxidant enzyme activity of largemouth bass was evaluated by intraperitoneal injection.In this experiment,60 healthy largemouth bass were randomly divided into 3 groups,and different doses of lipoic acid were injected according to the fish body weight.The groups were set as control(0 mg/kg),T20(20 mg/kg)and T40(40 mg/kg).The serum and liver were separated at 6,12,and 24 hours after injection,and the activity of catalase(CAT)in serum,catalase(CAT)and total superoxide dismutase(T-SOD)in the liver were measured.The results showed that the serum CAT enzyme activity in the T20 and T40 groups was significantly increased at 6h and 12 h compared with the control group(P<0.05).In the liver,CAT activity in the T20 group was not influenced by lipoic acid compared with control group(P>0.05),CAT activity of the T40 group was significantly higher than that of the control group and T20 group after 12 hours of treatment.Compared with the control group,the liver T-SOD activity of the T20 group was significantly increased at three time points(P<0.05),while the T40 group T-SOD activity was significantly increased at 6 h and 12 h(P<0.05),and the T-SOD activity of the T20 group was higher than that of the T40 group.The results indicated that the injection of lipoic acid could promote the activity of CAT and T-SOD in the serum and liver of largemouth bass.This promotion was related to the dosage of lipoic acid and the treatment time.
