马氏珠母贝大亚湾和三亚野生种群内自繁及种群间杂交一代的RAPD分析

摘    要:运用RAPD技术分析了广东大亚湾野生种群内繁殖一代DDl群体和海南三亚野生种群内繁殖一代SSl、两野生种群间杂交一代DS1群体各18个个体的遗传多样性。筛选的28个含10碱基的随机引物中,其中11个随机引物可产生稳定和可重复的多态扩增结果,共检测出86个位点,其中DDl群体有74个基因位点,其中56个表现多态,多态比例为75.68%;SSl群体79个基因位点,其中57个表现多态,多态比例是72.15%;DSl群体有72个基因位点,其中64个表现多态,多态比例为88.89%。用修正的Shannon表型多态性指数量化3个群体的遗传多态性,DDl、SSl和DS13个群体的遗传多态性分别为0.2280,0.2ll5和0.2456。不同地理种群间杂交能够增加子代的遗传多态性。

Abstract:Pinctada martensii (Dunker) is one of the most important cultured pearl oysters in the southeast of China. In order to improve efficiency of genetic breeding by marker-assisted selec-tion, 28 primers were used to test different populations by random amplified polymorphic DNA (RAPD). The individuals of wild populations were collected separately from Daya Bay, Guangdong province and Sanya ,Hainan province. On the same condition, let them mate (inner-population) and cross (between-population) respectively, getting three populations markered DD1,SS1 and DS1. A-mong the 28 primers ,11 produced highly reproducible profiles and generated 86 bands. There were 74 gene sites in DD1 population,including 56 polymorphic gene sites ,79 gene sites in SS1 population, including 57 polymorphic gene sites, 72 gene sites in DS1 population, including 56 polymorphic gene sites. The polymorphic proportions were 75.68 % (DD1), 72.15 % (SS1) and 88.89 % (DS1) respec-tively. The genetic diversity indexes of the three populations (DD1, SS1 and DS1) are 0.247, 0.237and 0.261 respectively. The crossing from two different geographical wild populations in-creased the genetic diversity of the first filial generation.
