摘 要: | 建立了测定鱼血浆、肌肉、肝胰脏、肾脏中培氟沙星含量的超高液相色谱一紫外法。将组织样品剪碎后用磷酸二氢钾溶液提取,固相萃取柱净化,0.1%甲酸水溶液:LN(V:V=90:10)流动相洗脱。该方法血浆的检出限为0.01μg·mL-1,肌肉、肝胰脏、肾脏的检出限均为0.02μg·g-1,线性范围为0.01-10μg·mL-1,平均回收率为70.2-95.1%。本方法的前处理过程操作简便,避免使用有机溶剂,减少对环境的污染和人体健康的伤害。

Abstract: | A method of ultra performance liquid chromatography was developed for detection of pefloxacin residues in plasma, muscle, hepatopancreas and kidney in fish. The tissue samples were extracted with different aqueous potassium dihydrogenphosphate solution, and purified by solid phase extraction cartridge, the acetonitrile and water (containing 0.1% formic acid) as the mobile phase. It was found that the limit of quantification was 0.01μg·mL-1 with linear ranges from 0.01μg·mL-1 to 10μg·mL-1. The average recoveries were ranged from 70.2 to 95.1%. This method was proved to be easy processing, avoiding use of organic solvents leading to reduce in pollution to environment and in health damage of human. |
