摘    要:利用冻融破碎、乙醇沉淀及凝胶层析等手段从培养至平衡期后的扁藻和螺旋藻细胞内提取分离出不同级分的多糖,对其理化性质和结构特征进行了研究和对比分析。结果表明,同一乙醇体积分数范围沉淀的扁藻多糖的分子量比螺旋藻多糖的分子量略大,并且多糖中结合的蛋白质比螺旋藻多糖结合的蛋白质略高;但扁藻多糖的糖基含量、溶解度和相对黏度却低于螺旋藻多糖。红外光谱结果表明螺旋藻多糖和扁藻多糖中均含有硫酸基,后者还含有氨基。这些结果说明不同微藻多糖及同一微藻的不同级分多糖,其组成不相同,而且扁藻多糖比螺旋藻多糖更复杂。所得结果为2种微藻多糖的实际应用提供了理论和试验基础。

Abstract:Different polysaccharides were extracted and purified from algae Platymonas subcordiformis and Spirulina platensis after they grew in the stationary phase by means of frozen-thawing,ethanol-precipitation and SephadexG-150 gel chromatography,and their properties and structural characters were analyzed. The results showed that the molecular weight of the polysaccharide from P.subcordiformis was bigger than that of polysaccharide from S.platensis,and there was more protein combined with polysaccharide in P.subcordiformis than in S.platensis,but the content of glycon-group,solubility and relative viscosity of polysaccharide from P.subcordiformis were lower than those of polysaccharide from S.platensis.The infrared spectrum exhibited that the two polysaccharides contained sulphuric group,and the polysaccharide from P.subcordiformis also contained amidogen.The results showed that there were differences in character and structure of polysaccharides from the two microalgae and provided theoretical and experimental foundation for their practical application.
