淇河鲫与两野生鲫鱼群体遗传多样性的ISSR分析

应用ISSR技术对淇河鲫、金堤河鲫、沁河鲫3个群体进行遗传多样性研究。结果表明:(1)筛选8个引物共得到94个扩增位点,其中多态位点82个,多态位点百分率为87.23%。(2)Popgene分析结果,淇河鲫群体的遗传多样性水平相对较低,PPB=61.70%,H=0.2036,I=0.3085。(3)3个群体间的遗传分化系数GST=0.1303,基因流Nm=3.3359。表明3个群体的遗传分化尚未达到种群的分化水平。(4)金堤河鲫和沁河鲫群体间遗传距离较近,淇河鲫有一定的遗传分化,形成了较稳定的、独立的遗传结构。

ISSR Analysis of Genetic Diversity Among Crucian Carp Carassius auratus in Qihe River and the Other Two Crucian Carp in Wild

The genetic diversity was studied among 3 populations of crucian carp Carassius auratus,one in Qihe River,one in Jindihe River and the other in Qinhe River,by Inter-Simple Sequence Repeat(ISSR).A total of 94 discernible DNA fragments were observed by 8 primers analysed,in which 82 fragments were polymorphic,with polymorphic rate of 87.23%.POPGENE revealed that the genetic diversity of the population in Qihe River was comparatively low,with PPB of 61.70%,H of 0.2036,and I of 0.3085.The inter-populations Nei’s genetic differentiation coefficient was found to be 0.1303 and gene flow 3.3359,indicating that the genetic differentiation among three populations were not beyond the population level yet.Crucian carp in Jindihe River and in Qinhe River stocks had the relatively similar genetic distance,while the crucian carp in Qihe River stock had a degree of genetic differentiation and the stable and independent genetic structure.
