摘    要:基于河流弧菌的toxR基因设计了6对引物,分别为F1、F2、F3、F4、F5、F6.以标准河流弧菌HL01、HL02、HL03、HL04做梯度PCR检测.试验结果表明,仅F2和F4能获得分子量分别为332 bp和358 bp的目的条带.利用其他27株非河流弧菌进行PCR特异性检测,结果表明,引物F2和F4的特异性较强,但F2和F4这两对引物的灵敏度也有一定的差异,F2的灵敏为9.3×103 cfu/ml,F4的灵敏度为9.3×102cfu/ml.

Abstract:Six pairs of primers,numbered from F1 to F6,were designed for rapid detection of Vibrio fluvialis according to its toxR gene.Gradient PCR was introduced to detect the feasibility of these primes with standard strains of V.fluvialis(HL01,HL02,HL03 and HL04).The results showed that only F2 and F4 amplified the expected products,332 bp and 358 bp,respectively.Furthermore,27 strains of non-V.fluvialis were used to determine the specificity of F2 and F4.The results showed that both F2 and F4 had high specificity...
