基于线粒体控制区D-loop序列的内蒙古大鳍鼓鳔鳅群体遗传结构分析

摘    要:为了解内蒙古地区大鳍鼓鳔鳅(Herzenstein)群体的遗传背景和分化情况,对该地区52个大鳍鼓鳔鳅样本的线粒体控制区D-loop序列进行了遗传多样性和遗传分化分析。结果显示,在947 bp的D-loop区序列中,碱基A、T、C、G的平均含量分别为34.5%、28.0%、21.3%、16.2%,表现出较强的AT偏好性;有27个样本在645位点均缺失C碱基,突变总数为21;D-loop区序列存在多态性位点/突变位点20个,单倍型数(h)、单倍型多样性(H d)、核苷酸多样性(π)分别为17、0.912和0.0045;系统进化树分析显示,样本D-loop区序列分布在2个大的分支上,表明该群体结构形成了一定的遗传分化;样本的D-loop序列间的遗传距离均在0.010以下,说明样本的D-loop区序列亲缘关系较近。

Abstract:In order to investigate the genetic background and differentiation of Herzenstein population in Inner Mongolia,52 Herzenstein samples were analyzed by sequencing their mitochondrial control region(D-loop region).The results showed that the contents of A,T,C and G base in the 947 bp fragment were 34.5%,28.0%,21.3%,16.2%,respectively,indicating a preference for A and T base.Among the 52 samples,27 samples lost C base at 645 sites,with a total of 21 mutations.Totally 20 variable sites were identified in samples.Overall haplotype(h)index,haplotype diversity(H d)and nucleotide diversity(π)were 17,0.912 and 0.0045,respectively.The genetic distances between the D-loop regions of 52 samples were all below 0.010,indicating that the genetic relationship between the samples was relatively close.
