摘    要:以四川白鹅为材料,用PCR-SSCP方法检测了PRL基因内含子2扩增片段,结果在群体中共检测到AA、AB两种基因型。对不同基因型个体测序分析发现,四川白鹅PRL基因内含子2上存在一个新的SNP(A126G)位点。此外,将测序获得的四川白鹅PRL基因内含子2序列与GenBank上发表的皖西白鹅、浙东白鹅、马岗鹅、莱茵鹅PRL基因序列比对发现,鹅PRL基因内含子2区段存在一个(GA)n简单重复序列,且浙东白鹅、马岗鹅、莱茵鹅相比四川白鹅和皖西白鹅少一个GA重复,即存在一个简单重复序列多态性位点。

Abstract:The single nucleotide polymorphism (SNP) sites of prolactin gene intron 2 fragment were detected in Sichuan white goose by PCR-SSCP method. The result showed that there were two SSCP genotype (AA and AB) individuals in the experiment geese. By sequencing of the PCR products of different genotype individuals, one new mutation (A126G) was found in the intron 2 fragment of prolactin gene. In addition, there was a (GA)n simple repeat sequence polymorphism site was found in the intron 2 fragment by aligning the sequence of Sichuan white goose with reported prolactin gene sequences of Wanxi white goose, Zhedong white goose, Magang goose and Rhine goose. There was one more GA repeat time in the intron 2 fragment of Sichuan white goose and Wanxi white goose than the other goose breeds. These polymorphism sites will be useful for screening goose laying performance relative genetic marker.
