松江鲈群体遗传多样性的ISSR分析

摘    要:利用ISSR分子标记技术对辽宁丹东和河北秦皇岛2个地区松江鲈(Trachidermus fasciatus)群体的遗传多样性进行了分析。结果显示:77个ISSR引物中有4个引物能扩增出清晰条带,4个引物对两个群体各24个样品扩增出27个位点。松江鲈丹东和秦皇岛两群体的多态位点比率均为44.44%,Shannon′s指数分别为0.2728、0.2836。Shannon′s指数和AMOVA分析均显示松江鲈的遗传变异主要来自于群体内个体间。松江鲈UPGMA系统树没有依据群体分别聚类,无明显遗传趋异。两个松江鲈野生群体的平均杂合度和多态位点与处于濒危状态的江豚类似,多态性位点比例低于濒危状态的平胸龟和长江刀鲚。结果表明:两个地区松江鲈群体的遗传多样性处于较低水平,遗传变异相对贫乏;松江鲈丹东和秦皇岛两个群体间无明显的遗传分化,亲缘关系较近。

Abstract:Inter-simple sequence repeat(ISSR)was used to detect the genetic variation of two geographical populations of Trachidermus fasciatus.The individuals of T.fasciatus were collected from Dandong(Liaoning)and Qinhuangdao(Hebei).Four primers,which were sifted from all 77 ISSR primers,produced 27 polymorphic markers from two populations.The proportions of polymorphic loci of Dandong and Qinhuangdao population were all 44.44%,and Shannon′s Lnformation index was 0.2728 and 0.2836.These results showed that the genetic diversity index of the two populations were at the same level.Shannon′s Information index and AMOVA analysis indicated that genetic variation mainly from individuals within populations and no significant genetic divergence between populations.The UPGMA tree based on ISSR data supported this result and no distinct variation was detected.The results of ISSR analysis indicated that two populations of T.fasciatus presented lower genetic diversity and there was a relatively lower level of genetic variation,there was no significant genetic differentiation between two populations.
