鱼腥草愈伤组织的诱导技术

摘    要:以鱼腥草茎秆和叶片为外植体,进行外植体消毒和愈伤组织诱导条件的研究。结果表明:外植体在0.1%升汞中浸泡10 min可达到良好的消毒效果,污染率低于2%;愈伤诱导所用的茎秆和叶片两种器官中,适宜的外植体是茎秆,其诱导率可达31.25%;愈伤诱导时,24 h暗培养比12 h黑暗与12 h光照交替培养诱导率高;在含2,4-D或6-BA的3种培养基中,仅在MS+2.0 mg.L-16-BA+6.0 g.L-1琼脂+30 g.L-1蔗糖的诱导培养基中诱导出了愈伤组织,诱导率最高为28.57%。

Abstract:Taking the stem and leaf of Houttuynia cordata Thunb.as explants,the explants sterilization and the callus induction conditions were researched.Results were as follows: explants were putting in 0.1% HgCl2 for 10 minutes,the rate of pollution is inferior to 2%;comparing callus induction of stem and leaf suitable explant is stem,the rate of callus induction is 31.25%,Callus induction training environment 24 h dark is better than 12 h dark and 12 h light,In three kinds of 2,4-D or 6-BA mediam,only in MS+2.0 mg·L-1 6-BA+6.0 g·L-1 agar+30 g·L-1 sucrose of induction medium the callus induction rate is highest,reaching 28.57%.
