油菜F2无性系群体稳定性的SSR检测

摘    要:为了研究F2无性系群体作为可重复作图群体的效率,将油菜F2种子进行组织培养,以试管苗茎尖、子叶和下胚轴为材料,分别进行快繁培养,获得F2无性系群体.以各F2试管苗子叶DNA(原供体)作为模板,用100对覆盖油菜全基因组的SSR引物进行扩增,从中选取20对扩增多态性高且带纹较强的引物,对03H109 F2无性系群体进行SSR扩增反应.比较再生植株和原供体间的SSR标记带,结果表明茎尖再生后代的变异率为0.0059%,接近自然变异率,而子叶和下胚轴再生后代的变异率分别为0.5271%和0.2095%,因此认为以茎尖再生所构建的F2无性系群体可以作为可重复作图群体.

Abstract:In order to test the efficiency of clonal F_2 population as a repetitive colony in genetic mapping, plantlets were regenerated by tissue culture from F_2 rapeseed plants. A clonal F_2 population was propagated by stem apex, cotyledon and hypocotyls of the plantlets. SSR amplified with 100 pairs of primers which cover the whole rapeseed genome using each cotyledon DNA as template. Clone of 03H109 and the original F_2 population as the control were studied using SSR by 20 pairs of primers which were selected for high polymorphism from 100 pairs. Only one DNA polymorphism was found among 16932 pair of plants, indicating 0.0059% variation in the stem apex, which is lower than natural variation. The rate of variation in cotyledon and hypocotyl regenerants were 0.5271% and (0.2095%.) Thus, the clonal F_2 population propagated from stem apex could be regarded as a repetitive population for genetic mapping.
