摘    要:为更加高效利用抗稻瘟病基因Pid3/Pid3-A4,通过分析3K水稻测序数据中Pid3基因等位变异,设计了Pid3和Pid3-A4基因特异InDel分子标记Pdg-C与PA4-C,利用这2个标记对中国杂交水稻常用亲本材料或品种以及美国水稻微核心种质资源进行检测.结果 显示,Pdg-C和PA4-C能够分别快速、准确、经济地区分Pid3和Pid3-A4基因;通过克隆测序、表达分析及接菌鉴定,明确了水稻品种华占携带Pid3基因,栽培稻311100携带Pid3-A4基因,它们作为抗病基因供体更便于在水稻育种中利用.

Development and Application of Specific InDel Markers for Rice Blast Resistance Genes Pid3/Pid3-A4

Abstract:Gene-specific molecular markers play an important role in introducing and screening rice blast resistance genes.In order to utilize the blast resistance genes Pid3/Pid3-A4 more efficiently and screen out disease-resistant donor materials which carry Pid3/Pid3-A4 genes and are easier to utilize in rice blast resistance breeding,the allelic variation of Pid3 in cultivated rice revealed by the 3K rice re-sequencing project was used to design the Pid3 and Pid3-A4 specific InDel marker Pdg-C and PA4-C,respectively.Then these two markers were used to screen rice breeding materials in China and the USDA Rice Mini-core Collection.Finally,the disease resistance of the selected candidate materials and expression of Pid3/Pid3-A4 gene in these materials were identified.The results showed that Pdg-C and PA4-C could differentiate Pid3 and Pid3-A4 genes rapidly,accurately and economically,respectively,and could accurately detect the existence of these two genes in germplasm resources.By clone sequencing,expression analysis and inoculation identification,it was founded that the rice variety Huazhan contains Pid3 gene,and the cultivated rice 311100 from the USDA Rice Mini-core Collection contains Pid3-A4 gene.The newly identified disease-resistant donor materials carrying Pid3/Pid3-A4 genes are more convenient to be used in rice breeding.
