利用PCR方法从苎麻中检测出粉虱传双生病毒

从苎麻植株上采集了叶片呈现花叶和黄绿斑驳并扭曲的2个病样.用粉虱传双生病毒简并引物对2个样品进行了PCR检测,结果发现2个样品都能扩增到预期大小的DNA片段.PCR产物经克隆后进行序列测定,BLAST比对结果表明,此样品的DNA扩增片断序列与Hsinchu番茄曲叶病毒福建分离物(EF125190,EF125191)和台湾分离物(DQ866131)的同源性都为95%,推测侵染苎麻的双生病毒可能是Hsinchu番茄曲叶病毒.

PCR Detection of Whitefly-transmitted Geminiviruses from Ramie

Two virus-infected ramie samples showing mosaic and leaf roll symptoms were collected in Haikou, Hainan Province. A pair of degenerate primers were designed according to the conserved sequence of CP gene and intergenic of Whitefly-transmitted geminiviruses (WTGV), and the PCR products of expected size were amplified from the two samples. The specific fragments were cloned and sequenced. A comparison of the 500 bp sequences with the reported begomoviruses shows that the DNA fragment has 95 % nucleotide sequence identity with that of tomato leaf curl Hsinchu virus isolates from Fujian (EF125190,EF125191) and Taiwan (DQ866131), suggesting the ramie samples be infected by tomato leaf curl Hsinchu virus.
