摘    要:利用ISSR分子标记对来源于不同地区的冬瓜种质资源亲缘关系进行分析.从67个ISSR引物中筛选出多态性强、重复性好的7条引物,对57份冬瓜资源基因组DNA进行扩增.结果表明:共扩增出50条谱带,其中多态性带34个,多态性位点百分率为68.0%.冬瓜种质间的遗传相似系数在0.26~1.00之间,大部分在0.70以上,表明供试的57份材料间遗传基础狭窄.利用UPGMA聚类分析,可将供试57份冬瓜种质划分为6个类群,类群的划分与地理来源有较高的相关性.本研究分类具有较高的应用价值,已利用本研究分类结果配制出杂种优势强的新组合.

Abstract:ISSR markers were employed to analyze the phylogenetic relationships of wax gourd germplasms from different regions. Seven polymorphic and reproducible primers were selected from 67 ISSR primers for PCR amplification in 57 samples. Totally, 50 bands were amplified, including 34 polymorphic bands. The polymorphic percentage is 68.0%. The genetic similarity among wax gourd germplasms ranged from 0.26~1.00, and most of which is higher than 0.70, indicating their narrow genetic diversity. Cluster analysis with UPGMA method showed that the 57 germplasm could be divided into six groups, which has a higher correlation with the geographical origin of the germplasms. The classification in the study has a great application value. A new variety with strong heterosis was bred based on the above study.
